自上个世纪90年代末以来,国内学者和政策决策者对集群的集群政策表现出日益浓厚的兴趣。由于现行的集群理论源自西方工业化国家,作为发展中的中国能否和如何吸收运用这些西方理论和研究经验成为讨论的焦点之一。本文的目的是通过探讨西方集群研究和集群政策的理论基础,以期能带给中国的相关研究一些有益的启示。本文首先对波特和经合组织两学派的集群概念的起源、发展和着重点进行了回顾。然后分析了集群起源和发展的条件、生命周期、发展机制、空间维度、类型等西方集群研究的要素,阐述了西方工业国家的集群政策概念及相关评判。相对于发达国家丰富的关于技术创新区域的研究而言,针对发展中国家创新系统和集群的理论性及经验性研究则要少得多。所以发展中国家的集群研究及集群政策需要更合适自身实际情况的理论。最后笔者以集群理论和中国实际情况为例,通过问题的分析、理论的分析和理论的调整三步骤说明新理论发展的可能途径。

Recurrent severe gastrointestinal bleeding and malabsorption due to extensive habitual megacolon

Dilatation of the colon and the rectum, which is not attributable to aganglionosis, is a rare finding and can be the result of intractable chronic constipation. We report a rare case of a 29-year-old male patient with impressive megacolon, in whom Hirschsprung's or Chagas disease was ruled out. In the present case, dilatation of the colon was most likely due to a behavioral disorder with habitual failure of defecation. Chronic stool retention led to a bizarre bulging of the large bowel with displacement of the other abdominal organs and severe occult blood loss. Because of two episodes of life-threatening gastrointestinal bleeding despite conventional treatment of constipation, a surgical approach for bowel restoration was necessary. Temporary loop ileostomy had to be performed for depressurization of the large bowel and the subsequent possibility for effective antegrade colonic lavage to remove impacted stools. Shortly after the operation, the patient was healthy and could easily manage the handling of the ileostomy. However, the course of the megacolon in this young adult cannot be predicted and the follow-up will have to reveal if regression of this extreme colonic distension with reestablishment of regular rectal perception will occur.

Chimeric protein probes for C5a receptors through fusion of the anaphylatoxin C5a core region with a small-molecule antagonist

Blockade of the interaction of anaphylatoxin C5a with its receptor C5aR1 has been actively studied as a potential treatment for many inflammatory diseases;but current C5a antagonists exhibit inadequate potency and poor species cross-reactivity, and novel biochemical tools are needed to investigate whether the core region of C5a contains important interaction epitopes that can explain these limitations. Herein, we report the development of chimeric protein C5a probes containing both the complete core region of rat or human C5a, and the small-molecule antagonist PMX53-1. These probes were chemically synthesized through hydrazide-based native chemical ligation of a linear peptide hydrazide with the requisite cyclopeptidic antagonist, both of which were made by solid-phase synthesis. Quasi-racemic X-ray crystallography established that attachment of PMX53-1 did not affect the structure of the core region of C5a. Subsequent C5aR1 activity assays demonstrated the probes can provide valuable insights into the development of C5a antagonists;for example, they exhibited significantly better binding affinity and much improved species cross-reactivity than PMX53-1, supporting the notion that the effect of some epitopes outside the C-terminus of C5a should be taken into consideration when designing better C5a antagonists. Surprisingly, the core region of C5a was found to partially agonize C5aR1, suggesting the presence of more than one agonistic interaction in the binding of C5a to C5aR1. This study exemplifies the value of chemical protein synthesis in developing novel receptor probes for drug discovery research.

The family of voltage-gated （Shaker-like） potassium channels in plants includes both inward-rectifying （Kin） channels that allow plant cells to accumulate K＋ and outward-rectifying （Kout） channels that mediate K＋ efflux. Despite their dose structural similarities, Kin and Kout channels differ in their gating sensitivity towards voltage and the extracellular K＋ concentration. We have carried out a systematic program of domain swapping between the Kout channel SKOR and the Kin channel KAT1 to examine the impacts on gating of the pore regions, the S4, S5, and the S6 helices. We found that, in particular, the N-terminal part of the S5 played a critical role in KAT1 and SKOR gating. Our findings were supported by molecular dynamics of KAT1 and SKOR homology models. In silico analysis revealed that during channel opening and closing, displacement of certain residues, especially in the S5 and S6 segments, is more pronounced in KAT1 than in SKOR. From our analysis of the S4-S6 region, we conclude that gating （and K＋-sensing in SKOR） depend on a number of structural elements that are dispersed over this -145-residue sequence and that these place additional constraints on configurational rearrangement of the channels during gating.

Media and Information Literacy in Inclusive Education: A Team Teaching Concept at the Technische Universitat Dortmund

There is a desideratum in the research about the interdependencies of inclusive education and media education. Digital media can facilitate individualized and cooperative learning. Additionally, digital media can be used to promote inclusion. However, which factors influence the acquisition of media and information literacy in inclusive education? Comparative international studies, e.g., the International Computer and Information Literacy Study (ICILS), show how social challenges, such as participation and handling heterogeneity at school, can be addressed through media. In numerous studies, it becomes apparent that German teachers do not sufficiently see the potential of digital media, especially for the improvement of learning opportunities for individuals with diverse learning requirements. German teachers are not well able to apply digital media appropriately in their lessons. Teachers need to be qualified and to design modern education with accessible media. In Germany, the ratio between available computers and pupils (1:4) is far better than the rate between teaching with and without computers. According to the teacher’s reports, only 9% of teachers are using digital media daily. Approximately 40% use digital media seldom or never. It seems that the cause is a skeptical attitude towards media among teachers, especially in humanities. That may result from missing didactical competences regarding digital media. The idea of Technical Pedagogical Content Knowledge (TPACK) and extending that explanatory model for teachers’ competencies on the inclusive practices builds the theoretical background for the design of joint seminars by the Research Center Youth-Media-Education as part of the Faculty for Cultural Studies and the Research Cluster Technology for Inclusion and Participation at the University of Dortmund. 近年来,中资企业的研发国际化倾向日益明显,海外专利申请数量激增。中资跨国公司的海外R&D中心区位与空间组织结构,以及对企业创新能力提升的影响成为国内外经济地理学研究的热点问题之一。以深圳华为技术有限公司为案例,基于世界知识产权组织专利数据与企业访谈资料,分析了华为研发国际化的途径和成效。研究发现:华为通过在全球卓越中心建立海外R&D中心以及与国际一流企业、大学与科研机构的跨境合作获得领先的研发资源,并通过总部协调实现知识由海外卓越中心向中国的流动。这种主动技术获取途径既保障华为获得全球领先的技术资源,又保障了知识商业化的成果归华为所有,是华为成长为全球领先企业的主要途径。
