本研究采用燃烧后四极质谱(QMS)检测装置测定地面水中有机悬浮物的元素组成。用离心法收集几种类型水中的固体物质。结果表明,本方法可用于水特性鉴定等方面。包括予处理和元素分析约需30分钟。无论水污染研究还是废物和天然有机物代谢机理的探讨,都越来越对地面水中不溶物内的有机质部分的元素组成感兴趣。有机质的组成极为复杂,因为它可能含有多种工业组分及脂肪、蛋白质和碳水化合物的代谢物。所以,进行全面的结梅分析以确定化合物的种类及其含量是不实际的而且几乎是不可能的。然而,元素分析可以给出化合物种类的有用信息,如脂肪或蛋白质的存在。最近Baumeister(1978)进行了地面水中悬浮物的CHN分析(即碳氢氮分析,译注)。他发现,各种不同试样的颗粒中C/N比率颇为一致。这可说明均来源于生物。

Cultivated potato is a clonally propagated autotetraploid species with a highly heterogeneous *** assemblies of six cultivars including two chromosome-scale phased genome assemblies revealed extensive allelic diversity,including altered coding and transcript sequences,preferential allele expression,and structural variation that collectively result in a highly complex transcriptome and predicted proteome,which are distributed across the homologous *** species contribute to the extensive allelic diversity in tetraploid cultivars,demonstrating ancestral introgressions predating modern breeding *** a clonally propagated autotetraploid that undergoes limited meiosis,dysfunctional and deleterious alleles are not purged in tetraploid *** a quarter of the loci bore mutations are predicted to have a high negative impact on protein function,complicating breeder’s efforts to reduce genetic *** StCDF1 locus controls maturity,and analysis of six tetraploid genomes revealed that 12 allelic variants of StCDF1 are correlated with maturity in a dosage-dependent *** of the complexity of the tetraploid potato genome with its rampant structural variation and embedded deleterious and dysfunctional alleles will be key not only to implementing precision breeding of tetraploid cultivars but also to the construction of homozygous,diploid potato germplasm containing favorable alleles to capitalize on heterosis in F1 hybrids.

Dense genetic maps create a base for QTL analysis of important traits and future implementation of marker-assisted *** tetraploid rose,the existing linkage maps include<300 markers to cover 28 linkage groups(4 homologous sets of 7 chromosomes).Here we used the 68k WagRhSNP Axiom single-nucleotide polymorphism(SNP)array for rose,in combination with SNP dosage calling at the tetraploid level,to genotype offspring from the garden rose cultivar‘Red New Dawn’.The offspring proved to be not from a single bi-parental *** rose breeding,crosses with unintended parents occur *** developed a strategy to separate progeny into putative populations,even while one of the parents was unknown,using principle component analysis on pairwise genetic distances based on sets of selected SNP markers that were homozygous,and therefore uninformative for one *** of the inferred populations was consistent with self-fertilization of‘Red New Dawn’.Subsequently,linkage maps were generated for a bi-parental and a self-pollinated population with‘Red New Dawn’as the common maternal *** densest map,for the selfed parent,had 1929 SNP markers on 25 linkage groups,covering 1765.5 cM at an average marker distance of 0.9 *** with the strawberry(Fragaria vesca)genome was *** ICM1 corresponded to *** pseudochromosome 7(Fv7),ICM4 to Fv4,ICM5 to Fv3,ICM6 to Fv2 and ICM7 to *** ICM2 corresponded to parts of *** pseudochromosomes 1 and 6,whereas ICM3 is syntenic to the remainder of Fv6.

Monocarboxylate transporter 8 (MCT8) is a thyroid hormone transporter, the gen e of which is located on the X chromosome. We tested whether mutations in MCT8 c ause severe psychomotor retardation and high serum triiodothyronine (T3) concent rations in five unrelated young boys. The coding sequence of MCT8 was analysed b y PCR and direct sequencing of its six exons. In two patients,gene deletions of 2·4 kb and 24 kb were recorded and in three patients missense mutations Ala150 Val, Arg171 stop, and Leu397Pro were identified. We suggest that this novel syndrome of X linked psychomotor retardation is due to a defect in T3 entry i nto neurons through MCT8, resulting in impaired T3 action and metabolism.

The adenosine triphosphate (ATP) binding cassette (ABC) transporters ABCG5 and ABCG8 have been shown to mediate hepatic and intestinal excretion of cholesterol. In various (genetically modified) murine models, a strong relationship was found between hepatic expression of ABCG5/ABCG8 and biliary cholesterol content. Our study aimed to relate levels of hepatic expression of ABCG5 and ABCG8 to biliary excretion of cholesterol in man. From 24 patients who had received a liver transplant, bile samples were collected daily after transplantation over a 2-week period to determine biliary composition. Expression of ABCG5, ABCG8, MDR3, and BSEP was assessed by real-time polymerase chain reaction (PCR) in liver biopsy specimens collected before and after transplantation. Levels of hepatic ABCG5, ABCG8, and MDR3 messenger RNA (mRNA) were strongly correlated. After transplantation, the biliary secretion rate of cholesterol continuously increased, coinciding with gradual increases in bile salt and phospholipid secretion. In contrast, hepatic levels of ABCG5 and ABCG8 mRNA remained unchanged. Surprisingly, no correlation was found between the hepatic expression of ABCG5 and ABCG8 and rates of biliary cholesterol secretion, normalized for biliary phospholipid secretion. As expected, the concentration of biliary phospholipids correlated well with MDR3 expression. In conclusion, the strong relationship between ABCG5 and ABCG8 gene expression is consistent with the coordinate regulation of both genes, and in line with heterodimerization of both proteins into a functional transporter. Hepatic ABCG5/ABCG8 expression, at least during the early phase after transplantation, is not directly related to biliary cholesterol secretion in humans. This finding suggests the existence of alternative pathways for the hepatobiliary transport of cholesterol that are not controlled by ABCG5/ABCG8.

Is overhydration in peritoneal dialysis patients associated with cardiac mortality that might be reversible?

AIM To study the relationship between overhydration(OH) in peritoneal dialysis(PD) patients and cardiac *** OH, as measured by body composition monitor(BCM), is associated with increased mortality in dialysis patients. BCM has been used to guide treatment on the assumption that correcting OH will improve cardiac morbidity and mortality although data demonstrating causality that is reversible is limited. We wished to determine if OH in PD patients predicted cardiac mortality, and if there was a correlation between OH and cardiac troponin-T(cT n T) levels. Finally, we wished to determine if improving OH values would lead to a decrement in cT n T. All prevalent PD patients over the study period of 57 mo who had contemporaneous BCM and cT nT measurements were followed irrespective of transplantation or PD technique failure. We also studied a cohort of patients with who had severe OH(> +2L).The Fresenius Body Composition Monitor was used to obtain hydration parameters. cT nT levels were done as part of routine clinical care. Data was analysed using SPSS version *** There were 48 deaths in the 336 patients. The patients that died from cardiac or non-cardiac causes were similar with respect to their age, incidence of diabetes mellitus, gender, ethnicity and cause of renal failure. However, the patients with cardiac causes of death had significantly shorter dialysis vintage(10.3 mo vs 37.0 mo, P < 0.0001) and were significantly more overhydrated by BCM measurement(2.95 L vs 1.35 L, P < 0.05). The mean(standard error of the means) hydration status of the 336 patients was +1.15(0.12) L and the median [interquartile range(IQR)] cT nT level was 43.5(20-90) ng/L. The cT nT results were not normally distributed and were therefore transformed logarithmically. There was a statistically significant correlation between Log(cT nT) with the OH value(Spearman r value 0.425, P < 0.0001).
