Use of methylation filtration and Cot fractionation for analysis of genome composition and comparative genomics in bread wheat

We investigated the compositional and structural differences in sequences derived from different fractions of wheat genomic DNA obtained using methylation filtration and Cot fractionation. Comparative analysis of these sequences revealed large compositional and structural variations in terms of GC content, different structural elements including repeat sequences （e.g., transposable elements and simple sequence repeats）, protein coding genes, and non-coding RNA genes. A correlation between methylation status [determined on the basis of selective inclusion/ exclusion in methylation-filtered （MF） library] of different repeat elements and expression level was observed. The expression levels were determined by comparing MF sequences with expressed sequence tags （ESTs） available in the public domain. Only a limited overlap among ME high Cot （HC）, and ESTs was observed, suggesting that these sequences may largely either represent the low-copy non-transcribed sequences or include genes with low expression levels. Thus, these results indicated a need to study MF and HC sequences along with ESTs to fully appreciate complexity of wheat gene space.

The prediction of water quality in terms of variables like dissolved oxygen (DO), biochemical oxygen demand (BOD), pH value, total dissolved solids (TDS) and salinity etc. is useful for evaluating the use of water for various related purposes. The widely used Streeter and Phelps models for computing biochemical oxygen demand and its impact on dissolved oxygen do not account for the settleable component of BOD and related implications. The model also does not account for the impact of storage zone on the stream’s DO. In the present work an attempt is made to develop a model which simultaneously accounts for the settleable component of BOD and the effect of storage zones onriver’s DO. An application of the model to real field data suggests that the cumulative impact of settleable BOD and presence of storage zone in the river is to shift the critical deficit closer to the point source and magnify its amount.

Chest Drain Insertion-Made Easy with Gum Bougie

Background: Surgical Chest drain insertion is a lifesaving procedure. It requires skills and experience towards its prompt insertion. Quite often there is delay or failure caused in its insertion due to excessive fat and loose skin in the axillary area while using the conventional methods. Numerous guidelines exist advising its safe placement, but mishaps still occur, as reported multiple times, while placing the chest drain. In addition to this, a delay or failure of its successful insertion can lead to disastrous consequences. Case: We would like to share, with fellow readers, a novel technique which we used in placing a surgical chest drain successfully. We used a gum bougie to guide the chest drain into the pleural cavity instead of using metal trocar, stellate or long forceps. Conclusion: By using this method, we experienced that not only it reduced the amount of time taken for the procedure, but it also had positive impact on the ease of insertion without fear of causing damage to internal organs, which ultimately boosted the operator’s confidence.

Pre-Operative Diagnostic Dilemma-Ovarian Ectopic Pregnancy

Primary ovarian pregnancy is one of the rarest varieties of ectopic pregnancies. Patients frequently present with abdominal pain and menstrual irregularities. Preoperative diagnosis is challenging but transvaginal sonography has often been helpful. A diagnostic delay may lead to rupture, secondary implantation or operative difficulties. Therefore, awareness of this rare condition is important in reducing the associated risks. Here, we report a case report of ovarian pregnancy presented. A 29-year-old woman, nulliparous, was admitted to the hospital forsuspected ectopic pregnancy. A transvaginal sonography failed to hint at ovarian ectopic. She was successfully managed by surgery and histological studies confirmed an ovarian pregnancy.

Provenance variations in proximate principles,mineral matter,total phenols and phytochemicals of Melia dubia drupes:an unexplored alternate livestock feed stock

This research was carried out on nine provenances from the north Western Ghats to quantify proximate principles,mineral matter,total phenols and other phytochemicals in Melia dubia drupe *** biometric,biomass attributes,proximate principles,mineral matter and total phenols diff ered signifi cantly among the *** average drupe length and width was 26.5 and 22.3 mm,*** drupe and pulp fresh weight,pulp dry weight and stone weight were 783.1,552.1,165.1,and 179.8 g/100 drupes,***,dry matter and moisture per cent were 70.5%,29.9%and 70.0%,*** crude fi bre,protein,ether extract,total ash and nitrogen-free extract were 8.3%,7.6%,5.1%,6.3%,and 72.6%,*** Ca,Mg,P and K levels were 0.5%,0.2%,0.2%,and 2.0%,*** phenols were 0.4%.Cluster analysis showed that the study area may be divided into three distinct provenances,each having diff erent drupe traits and proximate composition,including mineral ***-chromatography mass-spectrometry analysis revealed 27 phytochemicals,many of which possess benefi cial biological *** implications of this provenance variation study should be useful for future improvement in terms of better drupe pulp nutrition for livestock *** study suggests that *** pulp could be categorized as a good feed source with low fi bre,energy,fat and mineral *** proximate parameters,mineral matter content and total phenols are within prescribed limits.

Introduction Unconventional alternatives such as aerobic and anaerobic effluent from starch industry contain essential nutrients for Btk active ingredient *** from starch industry is rich in carbon and nitrogen and can replace expensive feedstock used during the fermentation *** The main objective of this study was to achieve a biopesticide formulation from starch industry wastewater(SIW)with high entomotoxicity(UI/ml)of larvae comparable to Foray 76B,which is a commercial *** Bacillus thuringiensis var kurstaki HD1(Btk)strain was cultivated and sub-cultured to aerobic,anaerobic digested effluent and ***-treatment was carried on these different substrates to enhance the residual carbon required for Btk growth and delta endotoxin *** 48 hours of fermentation,cells count and delta-endotoxin were determined.A biopesticide formulation containing fermented broth and adjuvants was fed to larvae to determine larvae *** Btk cell growth and sporulation profile in SIW media displayed a high total cell count and viable spores compared to btk growth in anaerobic or aerobic media after 48h *** maximum endotoxin concentration in the SIW medium was 435μg/mL,whereas,in anaerobic and aerobic effluent,the maximum concentrations were at 161μg/mL and 136μg/mL,*** acidic treatment was performed at pH 2 for these substrates,entomotoxicity obtained from aerobic and anaerobic biopesticide formulations displayed significantly higher entomoxicity than the untreated *** entomotoxicity of SIW treated at pH 2 was equivalent to the standard Foray 76B which is 20,000 IU/μ*** Anaerobic and aerobic effluent did not contain enough total organic carbon to augment Btk growth and *** pre-treated at pH 2 provided significant organic matter for Btk growth and resulted in larval mortality equivalent to the com ercial biopesticide Foray 76B.

Digital data have become a torrent engulfing every area of business, science and engineering disciplines, gushing into every economy, every organization and every user of digital technology. In the age of big data, deriving values and insights from big data using rich analytics becomes important for achieving competitiveness, success and leadership in every field. The Internet of Things (IoT) is causing the number and types of products to emit data at an unprecedented rate. Heterogeneity, scale, timeliness, complexity, and privacy problems with large data impede progress at all phases of the pipeline that can create value from data issues. With the push of such massive data, we are entering a new era of computing driven by novel and ground breaking research innovation on elastic parallelism, partitioning and scalability. Designing a scalable system for analysing, processing and mining huge real world datasets has become one of the challenging problems facing both systems researchers and data management researchers. In this paper, we will give an overview of computing infrastructure for IoT data processing, focusing on architectural and major challenges of massive data. We will briefly discuss about emerging computing infrastructure and technologies that are promising for improving massive data management.

Fuzzy Fault Tree Analysis for Fault Diagnosis of Cannula Fault in Power Transformer

Being one of the most expensive components of an electrical power plant, the failures of a power transformer can result in serious power system issues. So fault diagnosis for power transformer is highly important to ensure an uninterrupted power supply. Due to information transmission mistakes as well as arisen errors while processing data in surveying and monitoring state information of transformer, uncertain and incomplete information may be produced. Based on these points, this paper presents an intelligent fault diagnosis method of power transformer using fuzzy fault tree analysis (FTA) and beta distribution for failure possibility estimation. By using the technique we proposed herein, the continuous attribute values are transformed into the fuzzy numbers to give a realistic estimate of failure possibility of a basic event in FTA. Further, it explains a new approach based on Euclidean distance between fuzzy numbers, to rank the basic events in accordance with their Fuzzy Importance Index.

Hydrogen sulfide intervention in cystathionine-β-synthase mutant mouse helps restore ocular homeostasis

AIM: To investigate the applications of hydrogen sulfide(H2 S) in eye-specific ailments in ***: Heterozygous cystathionine-β-synthase(CBS+/-) and wild-type C57 BL/6 J(WT) mice fed with or without high methionine diet(HMD) were administered either phosphate buffered saline(PBS) or the slow-release H2 S donor: GYY4137. Several analyses were performed to study GYY4137 effects by examining retinal lysates for key protein expressions along with plasma glutamate and glutathione estimations. Intraocular pressure(IOP) was monitored during GYY4137 treatment; barium sulfate and bovine serum albumin conjugated fluorescein isothiocyanate(BSA-FITC) angiographies were performed for examining vasculature and its permeability post-treatment. Visionguided behavior was also tested employing novel object recognition test(NORT) and light-dark box test(LDBT) ***: CBS deficiency(CBS+/-) coupled with HMD led disruption of methionine/homocysteine(Hcy) metabolism leading to hyperhomocysteinemia(HHcy) in CBS+/-mice as reflected by increased Hcy, and s-adenosylhomocysteine hydrolase(SAHH) levels. Unlike CBS, cystathionine-γ lyase(CSE), methylenetetrahydrofolate reductase(MTHFR) levels which were reduced but compensated by GYY4137intervention. Heightened oxidative and endoplasmic reticulum(ER) stress responses were mitigated by GYY4137 effects along with enhanced glutathione(GSH) levels. Increased glutamate levels in CBS+/-strain were prominent than WT mice and these mice also exhibited higher IOP that was lowered by GYY4137 treatment. CBS deficiency also resulted in vision-guided behavioral impairment as revealed by NORT and LDBT findings. Interestingly, GYY4137 was able to improve CBS+/-mice behavior together with lowering their glutamate levels. Blood-retinal barrier(BRB) appeared compromised in CBS+/-with vessels' leakage that was mitigated in GYY4137 treated group.
