Comparative Evaluation of Traditional Susceptibility Testing for MRSA with the PCR Approach

Methicillin-resistant Staphylococcus aureus (MRSA) is a multi-drug resistant pathogen, which is responsible for increasing cases of serious diseases, including life-threatening diseases and nosocomial and community-acquired infections. Laboratory identification of MRSA is crucial and essential both for initiation of appropriate antimicrobial therapies and for effective infection control strategies that are designed to limit the spread of MRSA. In spite of the availability of commercial kits for MRSA detection in the market, the Clinical and Laboratory Standards Institute (CLSI) recommends the use of phenotypic methods, such as the disk diffusion method with oxacillin or with cefoxitin, as well as a serial dilution method with oxacillin. Nevertheless, some studies have shown that results obtained with such phenotypic methods are controversial. The aim of the study described in this paper was to comparatively evaluate the traditional susceptibility testing for MRSA with PCR as the gold standard assay. Analysis of collection (n = 68) isolates of Staphylococcus aureus revealed that the serial dilution method with oxacillin possessed the highest sensitivity (at 100%). In contrast, the disk diffusion methods with oxacillin and cefoxitin showed lower sensitivity (95.83%, 95% CI (78.81%-99.30%)). Furthermore, the borderline value of zone inhibition diameters for cefoxitin might be considered as a risk, and they may give false-susceptible result.

Diversity, novelty, antimicrobial activity, and new antibiotics of cultivable endophytic actinobacteria isolated from psammophytes collected from Taklamakan Desert

Three hundred and twenty endophytic actinobacterial strains were isolated from psammophytes collected from Taklamakan Desert and identified. Among them, three strains already had been identified as new species of two genera and sixteen isolates showed relatively low 16 S rRNA similarities < 98.6% to validly described species. Seventy-five of the isolates were selected as representative strains to screen antibacterial activity and mechanism. Forty-seven strains showed antagonistic activity against at least one of the indicator bacteria. Two Streptomyces strains produced bioactive compounds inducing DNA damage, and two Streptomyces strains produced bioactive compounds with inhibitory activity on protein biosynthesis. Notably, the strain Streptomyces sp. 8P21H-1 that demonstrated both strong antibacterial activity and inhibitory activity on protein biosynthesis was prioritized for exploring new *** the strategy of integrating genetics-based discovery program and MS/MS-based molecular networking, two new streptogramin-type antibiotics, i.e., acetyl-griseoviridin and desulphurizing griseoviridin, along with known griseoviridin, were isolated from the culture broth of strain 8P21H-1. Their chemical structures were determined by HR-MS, and 1D and 2D NMR. Desulphurizing griseoviridin and griseoviridin exhibited antibacterial activities by inhibiting translation.

Genes associated with testicular germ cell tumors and testicular dysgenesis in patients with testicular microlithiasis

Testicular microlithiasis （TM） is one of the symptoms of testicular dysgenesis syndrome （TDS）. TM is particularly interesting as an informative marker of testicular germ cell tumors （TGCTs）. KIT ligand gene （KITLG）, BCL2 antagonist/killer 1 （BAK1）, and sprouty RTK signaling antagonist 4 （SPRY4） genes are associated with a high risk of TGCTs, whereas bone morphogenetic protein 7 gene （BMP7）, transforming growth factor beta receptor 3 gene （TGFBR3）, and homeobox D cluster genes （HOXD） are related to TDS. Using polymerase chain reaction-restriction fragment length polymorphism （PCR-RFLP） analysis, we investigated allele and genotype frequencies for KITLG （rs995030, rs1508595）, SPRY4 （rs4624820, rs6897876）, BAK1 （rs210138）, BMP7 （rs388286）, TGFBR3 （rs12082710）, and HOXD （rs17198432） in 142 TGCT patients, 137 TM patients, and 153 fertile men （control group）. We found significant differences in the KITLG GG_rs995030 genotype in TM （P= 0.01） and TGCT patients （P = 0.0005） compared with the control. We also revealed strong associations between KITLGrs1508595 and TM （G allele, P = 0.003; GG genotype, P= 0.01） and between KITLG_rs1508595 and TGCTs （G allele, P-- 0.0001; GG genotype, P = 0.0007）. Moreover, there was a significant difference in BMP7_rs388286 between the TGCT group and the control （T allele, P = 0.00004; TT genotype, P = 0.00006） and between the TM group and the control （T allele, P= 0.04）. HOXDalso demonstrated a strong association with TGCTs （rs17198432 A allele, P = 0.0001; AA genotype, P = 0.001）. Furthermore, significant differences were found between the TGCT group and the control in the BAK1_rs210138 G allele （P= 0.03） and the GG genotype （P= 0.01）. KITLG and BMP7genes, associated with the development of TGCTs, may also be related to TM. In summary, the KITLG GG_rs995030, GG_rs1508595, BMP7 TT_rs388286, HOXD AA_rs17198432, and BAK1 GG_rs210138 genotypes were associated with a high risk of TGCT development.

目的研究P/Q型钙通道在小鼠海马神经元的突触传递中的重要作用。方法 1培养小鼠海马神经元用于研究,采用全细胞膜片钳记录技术,记录培养的对照组并趾症小鼠(oligosyndactyly,OS)和Leaner小鼠海马神经元钙离子电流;2刺激突触前细胞,记录突触后神经元抑制性突触后电流,观察对照组OS抑制性突触后电流对钙离子浓度的依赖性。3比较对照组OS和Leaner小鼠的突触后电流中各种钙通道在突触传递中所占比例。4转染长片段P/Q型质粒以恢复Leaner小鼠细胞因突变而引起的C末端过短所造成的功能缺失。结果 1抑制性突触后电流中P/Q型钙通道与钙浓度密切相关,在正常钙浓度的条件下,细胞外钙浓度为2mmol/L时,P/Q型钙通道起主要作用,当细胞外钙浓度升高到4mmol/L后,P/Q型钙通道功能所占比例部分由N型通道代替。2对照组OS和Leaner小鼠培养的海马神经元钙通道中P/Q型钙通道有很大的不同,突变小鼠中P/Q型钙通道功能减低,由其它通道补偿代替。3在培养的海马神经元记录的突触后电流显示Leaner小鼠的P/Q功能基本丧失,由N型和其他型所取代。4通过外源性转染含长片段P/Q型通道的质粒可以基本补偿P/Q型钙通道功能。结论 Leaner小鼠海马P/Q型钙通道功能降低是导致突触传递功能减少的重要因素。

Ultrathin tunable terahertz absorber based on MEMS-driven metamaterial

The realization of high-performance tunable absorbers for terahertz frequencies is crucial for advancing applications such as single-pixel imaging and *** on the strong position sensitivity of metamaterials’electromagnetic response,we combine meta-atoms that support strongly localized modes with suspended flat membranes that can be driven *** design maximizes the tunability range for small mechanical displacements of the *** employ a micro-electromechanical system technology and successfully fabricate the *** prototype devices are among the best-performing tunable THz absorbers demonstrated to date,with an ultrathin device thickness(~1/50 of the working wavelength),absorption varying between 60%and 80%in the initial state when the membranes remain suspended,and fast switching speed(~27μs).The absorption is tuned by an applied voltage,with the most marked results achieved when the structure reaches the snap-down *** this case,the resonance shifts by >200% of the linewidth(14% of the initial resonance frequency),and the absolute absorption modulation measured at the initial resonance can reach 65%.The demonstrated approach can be further optimized and extended to benefit numerous applications in THz technology.
