Modification of tenascin-R expression following unilateral labyrinthectomy in rats indicates its possible role in neural plasticity of the vestibular neural circuit

we have previously found that unilateral labyrinthectomy is accompanied by modification of hyaluronan and chondroitin sulfate proteoglycan staining in the lateral vestibular nucleus of rats and the time course of subsequent reorganization of extracellular matrix assembly correlates to the restoration of impaired vestibular function. The tenascin-R has repelling effect on pathfinding during axonal growth/regrowth, and thus inhibits neural circuit repair. By using immunohistochemical method, we studied the modification of tenascin-R expression in the superior, medial, lateral, and descending vestibular nuclei of the rat following unilateral labyrin- thectomy. On postoperative day 1, tenascin-R reaction in the perineuronal nets disappeared on the side of labyrinthectomy in the superior, lateral, medial, and rostral part of the descending vestibular nuclei. On survival day 3, the staining intensity of tenascin-R reaction in perineuronal nets recovered on the operated side of the medial vestibular nucleus, whereas it was restored by the time of postoperative day 7 in the superior, lateral and rostral part of the descending vestib- ular nuclei. The staining intensity of tenascin-R reaction remained unchanged in the caudal part of the descending vestibular nucleus bilaterally. Regional differences in the modification of tena- scin-R expression presented here may be associated with different roles of individual vestibular nuclei in the compensatory processes. The decreased expression of the tenascin-R may suggest the extracellular facilitation of plastic modifications in the vestibular neural circuit after lesion of the labyrinthine receptors.

Characterization of functional biliary pain and dyspeptic symptoms in patients with sphincter of Oddi dysfunction:Effect of papillotomy

AIM： To characterize functional biliary pain and other gastrointestinal （GI） symptoms in postcholecystectomy syndrome （PCS） patients with and without sphincter of Oddi dysfunction （SOD） proved by endoscopic sphincter of Oddi manometry （ESOM）, and to assess the postendoscopic sphincterotomy （EST） outcome. METHODS： we prospectively investigated 85 cholecystectomized patients referred for ERCP because of PCS and suspected SOD. On admission, all patients completed our questionnaire. Physical examination, laboratory tests, abdominal ultrasound, quantitative hepatobiliary scintigraphy （QHBS）, and ERCP were performed in all patients. Based on clinical and ERCP findings 15 patients had unexpected bile duct stone disease and 15 patients had SOD biliary type Ⅰ. ESOM demonstrated an elevated basal pressure in 25 patients with SOD biliary-type Ⅲ. In the remaining 30 cholecystectomized patients without SOD, the liver function tests, ERCP, QHBS and ESOM were all normal. As a control group, 30 ‘asymptomatic＇ cholecystectomized volunteers （attended to our hospital for general cardiovascular screening） completed our questionnaire, which is consisted of 50 separate questions on GI symptoms and abdominal pain characteristics. Severity of the abdominal pain （frequency and intensity） was assessed with a visual analogue scale （VAS）. In 40 of 80 patients having definite SOD （i.e. patients with SOD biliary type Ⅰ and those with elevated SO basal pressure on ESOM）, an EST was performed just afl：er ERCP. In these patients repeated questionnaires were filled at each follow-up visit （at 3 and 6 too） and a second look QHBS was performed 3 mo after the EST to assess the functional response to EST. RESULTS： The analysis of characteristics of the abdominal pain demonstrated that patients with common bile duct stone and definite SOD had a significantly higher score of symptomatic agreement with previously determined biliary-like pain features than patient groups of PCS without SOD and controls. Principles of molecular self-assembly into giant hierarchical structures of hundreds of micrometers in size are studied in aggregates of meso-tetra(4-sulfonatophenyl)porphine(TPPS_(4)).The aggregates form a central tubular core,which is covered with radially protruding filamentous non-branching *** filaments cluster and orient at varying angles from the core surface and some filaments form *** to shape resemblance,the structures are termed giant sea urchin(GSU)*** resolved fluorescence microscopy reveals J-and H-bands of TPPS_(4)aggregates in both the central core and the *** fluorescence of the core is quenched while filaments exhibit strong *** drying,the filament fluorescence gets quenched while the core is less affected,showing stronger relative *** linear dichroism(FDLD)microscopy reveals that absorption dipoles corresponding to J-bands are oriented along the filament *** comparison of FDLD with scanning electron microscopy(SEM)reveals the structure of central core comprised of multilayer ribbons,which wind around the core axis forming a *** second-harmonic generation(SHG)and thirdharmonic generation microscopy exhibits strong signal from the filaments with nonlinear dipoles oriented close to the filament axis,while central core displays very low SHG due to close to centrosymmetric *** chiral nonlinear susceptibility points to helical arrangement of the *** investigation shows that TPPS_(4)molecules form distinct aggregate types,including chiral nanotubes and nanogranular aggregates that associate into the hierarchical GSU structure,prototypical to complex biological *** chiral TPPS_(4)aggregates can serve as harmonophores for nonlinear microscopy.

Nervous System and Gastrointestinal Effects of the Insecticide Esfenvalerate on the Rat:An Ex Vivo Study

Esfenvalerate belongs to the pyrethroid group of insecticides which display significant selective toxicity against insects compared to mammalian species, nevertheless, they may pose health risks, especially in case of accidental exposure. The aim of the present study was to model the effect of acute, relatively high-dose exposure of the esfenvalerate-containing formulation Sumi-Alpha?. Eventual functional alterations in the central nervous system and in the gastrointestinal tract were studied on in vitro tissue preparations at different delays after intragastric administration to rats. Neuronal effects were characterized by field potential recording in cortical and hippocampal brain slices, while gastrointestinal effects were examined by analyzing the motility and excitability of isolated ileum segments. On the brain slices originating from esfenvalerate-treated animals, changes in excitability of both inhibitory and excitatory type could be observed. Voltage thresholds necessary to evoke responses in neocortex slices were elevated, and population spike amplitudes were lower in hippocampal slices. However, epileptiform potentials with pronounced late components were also observed. A decreased long-term potentiation (LTP) could be seen in both brain areas after esfenvalerate treatment. Seizure susceptibility of the slices was not significantly altered, but tended to be somewhat higher in slices originating from treated rats. In ileum segments, both spontaneous and acetyl-choline (ACh)-elicited contractions were modified by treatment. Esfenvalerate raised the amplitude of contractions in the low ACh concentration range. However, the solvent xylene also considerably contributed to the detected changes. we can conclude that a relatively high, single oral dose of Sumi-Alpha? exerted mild and temporary effects on the elementary brain functions and intestine functions of the rat.

The horizontal pendulums of the Grotta Gigante (Giant Cave) in the Trieste Karst are long-base tiltmeters with Zllner type suspension. The instruments have been continuously recording tilt and shear in the Grotta Gigante since the date of their installation by Prof. Antonio Marussi in 1966. Their setup has been completely overhauled several times since installation, restricting the interruptions of the measurements though to a minimum. The continuous recordings, apart from some interruptions, cover thus almost 40 years of measurements, producing a very noticeable long term tiltmeter record of crustal deformation. The original recording system, still in function, was photographic with a mechanical timing and paper-advancing system, which has never given any problems at all, as it is very stable and not vulnerable by external factors as high humidity, problems in power supply, lightning or similar. In December 2003 a new recording system was installed, based on a solid-state acquisition system intercepting a laser light reflected from a mirror mounted on the horizontal pendulum beam. The acquisition frequency is 30 Hz, which turns the long-base instrument to a very-broad-band tiltmeter, apt to record the tilt signal on a broad band of frequencies, ranging from secular deformation rate through the earth tides to seismic waves. Here we describe the acquisition system, present the up to date long term recording, and the observation of a recent earthquake.

Exhaled Breath Vapor of Humans Reflects the Changes in Deuterium Concentration in the Body water

The concentration of the heavy isotope of hydrogen, deuterium (D), is not routinely measured in (human) medical laboratory tests, even though an increasing number of papers prove the pivotal role of D in tumor growth, cell cycle regulation, cell metabolism, and aging. Data from a prospective phase 2 clinical study and numerous retrospective clinical studies proved the anticancer effect of deuterium depletion achieved by replacing the regular water intake with deuterium-depleted water (DDw). In previous studies, the changes in serum D concentration of DDw-consuming patients were followed using blood samples and mass spectrometry, which was invasive, costly, and time-consuming. As future clinical trials will also require a follow-up of internal D level and the patient’s compliance, a new sampling device and procedure was developed based on condensing the exhaled breath water vapor and measuring its D content using a liquid water isotope laser analyzer. Test results showed that the device provided accurate, reliable, and reproducible data. According to the data, the internal D level in a person consuming normal water was stable. In contrast, exclusive consumption of DDw for several days resulted in a gradual decrease of D concentration in exhaled breath condensate (EBC), which was proportional to the D concentration of DDw. These data confirm that orally applied DDw equilibrates with the person’s water pool quickly, leading to a reduced internal D level reflected in the D content of EBC.

Propagation methods for black locust (Robinia pseudoacacia L.) improvement in Hungary

Black locust (Robinia pseudoacacia L.) is one of the most important stand-forming tree species in Hungary and its importance is increasing in many countries. Black locust plants are commonly produced by two methods, by seed and by root cuttings. Tissue culture propagation can be considered as a relatively new method. Growing trees from seed is a relatively sim-ple method for reliably producing seedlings on a large scale under a variety of circumstances. Mechanization of the method is easy and the production cost is relatively low. Propagation from root cuttings and tissue culture are valuable for reproduction of superior individuals or varieties. By applying these methods, superior traits of the selected trees can be preserved in the clones. Recent experiments demonstrated that micropropagated trees could be successfully transplanted into soil, hardened and grown in the field.
