Ai.:To i.vesti.ate the effect of acetyl-L-carni.i.e(ALCAR)on cell vi.bi.i.y,morphologi.al i.tegri.y,and vascular endotheli.l growth factor(VEGF)expressi.n i. human reti.al pi.ment epi.heli.m(ARPE-19)cells usi.g a hypoxi. ***:i. the fi.st set of experi.ents,the opti.al CoCl_(2) dose was determi.ed by exposi.g ARPE-19 cell cultures to di.ferent *** evaluate the effect of ALCAR on cell vi.bi.i.y,fi.e groups of ARPE-19 cell culture were establi.hed that i.cluded a control group,a sham group(200μM CoCl_(2)),and groups that recei.ed 1,10 and 100 mM doses of ALCAR combi.ed wi.h 200μM CoCl_(2),*** cell vi.bi.i.y was measured by MTT *** morphologi.al characteri.ti.s of cells were observed by an i.verted phase contrast *** levels of VEGF and Hi.-1α secreti.n by ARPE-19 cells were detected by enzyme li.ked i.munosorbent assay(ELi.A)***:ARPE-19 cells were exposed to di.ferent doses of CoCl_(2) i. order to create a hypoxi. ***,when exposed to a concentrati.n of 200μM CoCl_(2),a notable decrease i. vi.bi.i.y to 83% was *** was found to i.crease the cell vi.bi.i.y at 1 mM and 10 mM concentrati.ns,whi.e the hi.hest concentrati.n(100 mM)di. not have an added *** cell vi.bi.i.y was found to be si.ni.i.antly hi.her i. the groups treated wi.h a concentrati.n of 1 mM and 10 mM ALCAR compared to the Sham group(P=0.041,P=0.019,respecti.ely).The cell vi.bi.i.y and morphology remai.ed unaffected by the greatest dose of ALCAR(100 mM).The admi.i.trati.n of 10 mM ALCAR demonstrated a stati.ti.ally si.ni.i.ant reducti.n i. the levels of VEGF and Hi.-1α compared wi.h the Sham group(P=0.013,P=0.033,respecti.ely).CONCLUSi.N:The fi.di.gs from the current study i.di.ate that ALCAR could represent a vi.ble therapeuti. opti.n wi.h the potenti.l to open up novel treatment pathways for reti.al di.eases,parti.ular relevance for age-related macular degenerati.n(AMD).However,to fully eluci.ate ALCAR’s appli.ati.n

Age-related memory i.pai.ments show a progressi.e decli.e across li.espan. Studi.s have demonstrated equi.ocal results i. bi.logi.al and behavi.ral outcomes of agi.g. Thus, i. the present study we exami.ed the novel object recogni.i.n task at a delay peri.d that has been shown to be i.pai.ed i. aged rats of two di.ferent strai.s. Moreover, we used a strai. of rats, Fi.her 344XBrown Norway, whi.h have publi.hed age-related bi.logi.al changes i. the brai.. Young (10 month old) and aged (28 month old) rats were tested on a standard novel object recogni.i.n task wi.h a 50-mi.ute delay peri.d. The data showed that young and aged rats i. the strai. we used performed equally well on the novel object recogni.i.n task and that both young and old rats demonstrated a ri.hthanded si.e preference for the novel object. Our data suggested that novel object recogni.i.n i. not i.pai.ed i. aged rats although both young and old rats have a demonstrated si.e preference. Thus, i. may be that geneti. di.ferences across strai.s contri.ute to the equi.ocal results i. behavi.r, and geneti. vari.nce li.ely i.fluences the course of cogni.i.e agi.g.
