An update on the molecular biology of glioblastoma,with clinical implications and progress in its treatment

Glioblastoma multiforme(GBM)is the most aggressive and common malig-nant primary brain tumor.Patients with GBM often have poor prognoses,with a median survival of∼15 months.Enhanced understanding of the molecular biology of central nervous system tumors has led to modifications in their classifications,the most recent of which classified these tumors into new categories and made some changes in their nomenclature and grading system.This review aims to give a panoramic view of the last 3 years’findings in glioblastoma characterization,its heterogeneity,and current advances in its treatment.Several molecular parameters have been used to achieve an accurate and personalized characterization of glioblastoma in patients,including epigenetic,genetic,transcriptomic and metabolic features,as well as age-and sex-related patterns and the involvement of several noncoding RNAs in glioblastoma progression.Astrocyte-like neural stem cells and outer radial glial-like cells from the subven-tricular zone have been proposed as agents involved in GBM of IDH-wildtype origin,but this remains controversial.Glioblastoma metabolism is characterized by upregulation of the PI3K/Akt/mTOR signaling pathway,promotion of the gly-colytic flux,maintenance of lipid storage,and other features.This metabolism also contributes to glioblastoma’s resistance to conventional therapies.Tumor heterogeneity,a hallmark of GBM,has been shown to affect the genetic expresion,modulation of metabolic pathways,and immune system evasion.GBM’s aggressive invasion potential is modulated by cell-to-cell crosstalk within the tumor microenvironment and altered expressions of specific genes,such as ANXA2,GBP2,FN1,PHIP,and GLUT3.Nevertheless,the rising number of active clinical trials illustrates the efforts to identify new targets and drugs to treat this malignancy.Immunotherapy is still relevant for research purposes,given the amount of ongoing clinical trials based on this strategy to treat GBM,and neoantigen and nucleic acid-based vaccines are gaining importance due

SIMULATION OF O_(2)-BLOWN CO-GASIFICATION OF WOOD CHIP AND POTATO PEEL FOR PRODUCING SYNGAS

Potato is the fifth largest agricultural crop in Canada and contributes to the generation of an abundant amount of potato peel.However,disposal/recycling this peel remains a challenge due to the stringent environmental regulations.Consequently,there is a lack of an appropriate recycling and valorization methods of potato peel.Gasification is an effective technology for producing syngas and an ecofriendly waste disposal approach.Syngas is an important industrial intermediate to produce synthetic fuels and chemicals.To develop an ecofriendly and cost-effective valorization approach for potato peel,this study used a mixture of woody biomass(i.e.,wood chips)and potato peel to produce syngas by co-gasification using O_(2) as the gasifying agent at a constant equivalence ratio of 0.3 using Aspen Plus simulation software.The influences of gasification temperature and wood chip/potato peel weight ratio on the carbon conversion efficiency(CCE),and product gas composition(molar fraction)and lower heating value(LHV)of product gas were investigated.This simulation indicated that a positive synergistic interaction occurs between wood chips and potato peel in co-gasification process in terms of an increase in CCE by comparing the arithmetic value and real value at all simulated wood chip to potato peel weight ratios(44.9%to 85.8%,46.5%to 76.2%,and 48.1%to 78.6%at ratios of 25:75,50:50,and 75:25,respectively,for wood chips to potato peel).While the molar fraction of H_(2) and CO decreased continuously with increase in the weight percentage of wood chips in the wood chip-potato peel mixture from 0 wt%to 100 wt%(H_(2),at 42.1 mol%to 41.4 mol%;and CO at 44.0 mol%to 40.4 mol%),accompanied by a decrease of the LHV of the product gas(10.3 to 9.78 MJ·Nm^(−3)).The study concluded that co-gasification for producing syngas is feasible and environmental-friendly option to recycle and valorize potato peel.

Wheat grain quality characteristics have experienced increasing attention as a central factor affecting wheat end-use products quality and human health.Nonetheless,in the last decades a reduction in grain quality has been observed.Therefore,it is central to develop efficient quality-related phenotyping tools.In this sense,one of the most relevant wheat features related to grain quality traits is grain nitrogen content,which is directly linked to grain protein content and monitorable with remote sensing approaches.Moreover,the relation between nitrogen fertilization and grain nitrogen content(protein)plays a central role in the sustainability of agriculture.Both aiming to develop efficient phenotyping tools using remote sensing instruments and to advance towards a field-level efficient and sustainable monitoring of grain nitrogen status,this paper studies the efficacy of various sensors,multispectral and visible red-greenblue(RGB),at different scales,ground and unmanned aerial vehicle(UAV),and phenological stages(anthesis and grain filling)to estimate grain nitrogen content.Linear models were calculated using vegetation indices at each sensing level,sensor type and phenological stage.Furthermore,this study explores the up-scalability of the best performing model to satellite level Sentinel-2 equivalent data.We found that models built at the phenological stage of anthesis with UAV-level multispectral cameras using red-edge bands outperformed grain nitrogen content estimation(R2=0.42,RMSE=0.18%)in comparison with those models built with RGB imagery at ground and aerial level,as well as with those built with widely used ground-level multispectral sensors.We also demonstrated the possibility to use UAV-built multispectral linear models at the satellite scale to determine grain nitrogen content effectively(R2=0.40,RMSE=0.29%)at actual wheat fields.

Heat dissipating upper body compression garment:Thermoregulatory,cardiovascular, and perceptual responses

Purpose:The aim of the present study was to determine the effects of an upper body compression garment(UBCG)on thermoregulatory responses during cycling in a controlled laboratory thermoneutral environment(~23℃).A secondary aim was to determine the cardiovascular and perceptual responses when wearing the garment.Methods:Sixteen untrained participants(age:21.3±5.7 years;peak oxygen consumption(V02 peak):50.88±8.00 mL/min/kg;mean±SD)performed 2 cycling trials in a thermoneutral environment(~23℃)wearing either UBCG or control(Con)garment.Testing consisted of a 5-min rest on a cycle ergometer,followed by 4 bouts of cycling for 14-min at ~50%VO2 peak,with 1-min rest between each bout.At the end of these bouts there was 10-min of passive recovery.During the entire protocol rectal temperature(Trec),skin temperature(Tskin),mean body temperature(Tbody),and heat storage(HS)were measured.Heart rate(HR),VO2,pH,hematocrit(Hct),plasma electrolytes,weight loss(Wloss),and perceptual responses were also measured.Results:There were no significant differences between garments for Tskin,HS,HR,VO2,pH,Hct,plasma electrolyte concentration,Wloss,and perceptual responses during the trial.Trec did not differ between garment conditions during rest,exercise,or recovery although a greater reduction in Trec wearing UBCG(p=0.01)was observed during recovery.Lower Tbody during recovery was found when wearing UBCG(36.82℃±0.30℃ vs.36.99℃±0.24℃).Conclusion:Wearing a UBCG did not benefit thermoregulatory,cardiovascular,and perceptual responses during exercise although it was found to lower Tbody during recovery,which suggests that it could be used as a recovery tool after exercise.

Survival after pulmonary metastasectomy in colorectal cancer patients:does a history of resected liver metastases worsen the prognosis?A literature review

Objective: To assess the impact of past liver metastases on the survival duration of patients who are undergoing surgery for lung metastases.Methods: We conducted a review of literature published from 2007 to 2014. The studies were identified by searching Pub Med,MEDLINE, and Embase and were supplemented by a manual search of the references listed by the retrieved studies. The following search terms were used: lung metastasectomy, pulmonary metastasectomy, lung metastases, and lung metastasis. We selected retrospective and prospective studies published from 2007 to 2014 on patients with lung metastases from colorectal cancer and were undergoing surgery with curative intent. We excluded reviews, studies that focused on surgical techniques, patients who were treated non-surgically, analyses of specific subgroups of patients, and those that did not report follow-up of the patients undergoing surgery.Results: We identified 28 papers that assessed survival after lung metastases, 21 of which were mostly retrospective studies that identified previous liver metastases to explore their impact on patient survival. In more than half of the papers analyzed(63.2%),patients with a history of resected liver metastases had a lower survival rate than those who did not have such a history, and the difference was statistically significant in eight of these studies. However, data were presented differently, and authors reported mean survival time, survival rates, or hazard ratios.Conclusions: A history of liver metastases seems to be a negative prognostic factor, but the individual data need to undergo a meta-analysis.

Electrode manufacturing process strongly impacts lithium-ion battery characteristics.The electrode slurry properties and the coating parameters are among the main factors influencing the electrode heterogeneity which impacts the battery cell performance and lifetime.However,the analysis of the impact of electrode manufacturing parameters on the electrode heterogeneity is difficult to be quantified and automatized due to the large number of parameters that can be adjusted in the process.In this work,a data-driven methodology was developed for automatic assessment of the impact of parameters such as the formulation and liquid-to-solid ratio in the slurry,and the gap used for its coating on the current collector,on the electrodes heterogeneity.A dataset generated by experimental measurements was used for training and testing a Machine Learning(ML)classifier namely Gaussian Naives Bayes algorithm,for predicting if an electrode is homogeneous or heterogeneous depending on the manufacturing parameters.Lastly,through a 2D representation,the impact of the manufacturing parameters on the electrode heterogeneity was assessed in detail,paving the way towards a powerful tool for the optimization of next generation of battery electrodes.

Wheat ear carbon assimilation and nitrogen remobilization contribute significantly to grain yield

The role of wheat ears as a source of nitrogen （N） and carbon （C） in the grain filling process has barely been studied. To resolve this question, five wheat genotypes were labeled with 15N-enriched nutrient solution. N remobilization and absorption were estimated via the nitrogen isotope composition of total organic matter and Rubisco. Gas exchange analyses showed that ear photosynthesis contributed substantially to grain filling in spite of the great loss of C due to respiration. Of the total kernel N, 64.7% was derived from the N acquired between sowing and anthesis, while the remaining 35.3% was derived from the N acquired between anthesis and maturity. In addition, 1.87 times more N was remobilized to the developing kernel from the ear than from the flag leaf. The higher yielding genotypes showed an increased N remobflization to the kernel compared to the lower yielding genotypes. In addition, the higher yielding genotypes remobilized more N from the ears to the kernel than the lower yielding genotypes, while the lower yielding genotypes remobilized more N from the flag leaf to the kernel. Therefore, the ears contribute significantly toward fulfilling C and N demands during grain filling.

Photosynthetic and Molecular Markers of CO_2-mediated Photosynthetic Downregulation in Nodulated Alfalfa

Elevated CO2 leads to a decrease in potential net photosynthesis in long-term experiments and thus to a reduction in potential growth. This process is known as photosynthetic downregulation. There is no agreement on the definition of which parameters are the most sensitive for detecting CO2 acclimation. In order to investigate the most sensitive photosynthetic and molecular markers of CO2 acclimation, the effects of elevated CO2, and associated elevated temperature were analyzed in alfalfa plants inoculated with different Sinorhizobium meliloti strains. Plants （Medicago sativa L. cv. Aragon） were grown in summer or autumn in temperature gradient greenhouses （TGG）. At the end of the experiment, all plants showed acclimation in both seasons, especially under elevated summer temperatures. This was probably due to the lower nitrogen （N） availability caused by decreased N2-fixation under higher temperatures. Photosynthesis measured at growth CO2 concentration, rubisco in vitro activity and maximum rate of carboxylation were the most sensitive parameters for detecting downregulation. Severe acclimation was also related with decreases in leaf nitrogen content associated with declines in rubisco content （large and small subunits） and activity that resulted in a drop in photosynthesis. Despite the sensitivity of rubisco content as a marker of acclimation, it was not coordinated with gene expression, possibly due to a lag between gene transcription and protein translation.
