分级存储系统通常根据访问频率将数据分层:将访问频率最高的数据放在快速存储设备中(热数据层),将访问频率最低的数据存储在慢速存储设备中(冷数据层),将访问频率介于两者之间的数据放在中速存储设备中(温数据层)。分布式文件系统通常使用多副本技术来保证系统的可靠性和可用性,比如在集群的不同节点上保存数据的三个副本,多副本技术通常在热数据层被使用。类似于Reed Solomon的纠删码技术(Erasure Code)正越来越多被使用,因为纠删码技术能够进一步地减小存储开销,但是其提供了较低性能的I/O和可用性。所以纠删码技术被更多地用来存储冷数据(比如冷存储层)。现有系统使用三副本复制,纠删码技术或将两者融合合来实现异构存储系统。在这些实现中,多副本技术和纠删码技术的融合表明:两种技术在I/O性能和存储成本方面存在着巨大差距,这导致将数据从一种形式迁移到另一种形式将产大巨大的系统开销。因此,现有的方法不适用于温数据层实现,特别是在分层存储系统中。为了解决这个问题,在本文中我们引入了Warm Cache,一种用于存储温数据的新数据层:将一个副本通过纠删码技术存储,另一个副本则存储在主存储数据层中。在纠删码数据层中使用一个副本可确保数据可靠性,而另一个副本放在主存储数据层可提供高效的I/O性能。为了展示Warm Cache的性能,我们在Alluxio和Hadoop系统中实现了我们的方法,并使用TestDFSIO来测量系统的I/O性能和存储开销。结果表明,与传统方法(例如多副本,纠删码,两者组合)相比,我们的方案在存储开销方面的性能比纠删码技术高出10.2倍,比多副本技术高出3.63倍。

Distributed Hybrid Testing（DHT） is an experimental technique designed to capitalise on advances in modern networking infrastructure to overcome traditional laboratory capacity limitations. By coupling the heterogeneous test apparatus and computational resources of geographically distributed laboratories, DHT provides the means to take on complex, multi-disciplinary challenges with new forms of communication and collaboration. To introduce the opportunity and practicability afforded by DHT, here an exemplar multi-site test is addressed in which a dedicated fibre network and suite of custom software is used to connect the geotechnical centrifuge at the University of Cambridge with a variety of structural dynamics loading apparatus at the University of Oxford and the University of Bristol. While centrifuge time-scaling prevents real-time rates of loading in this test, such experiments may be used to gain valuable insights into physical phenomena, test procedure and accuracy. These and other related experiments have led to the development of the real-time DHT technique and the creation of a flexible framework that aims to facilitate future distributed tests within the UK and beyond. As a further example, a real-time DHT experiment between structural labs using this framework for testing across the Internet is also presented.

A metagenomic fosmid library of approximately 52 000 clones was constructed to identify functional genes encoding cold-adapted enzymes. metagenomic DNA was extracted from a sample of glacial meltwater, collected on the Antarctic Peninsula during the ANTARKOS XXIX Expedition during the austral summer of 2012-2013. Each clone contained an insert of about 35-40 kb, so the library represented almost 2 Gb of genetic information from metagenomic DNA. Activity-driven screening was used to detect the cold-adapted functions expressed by the library. Fifty lipase/esterase and two cellulase-producing clones were isolated, and two clones able to grow on Avicel as the sole carbon source. Interestingly, three clones formed a brown precipitate in the presence of manganese （II）. Accumulation of manganese oxides was determined with a leucoberbelin blue assay, indicating that these three clones had manganese-oxidizing activity. To the best of our knowledge, this is the first report of a manganese oxidase activity detected with a functional metagenomic strategy.

It is estimated that more than three quarters of the Earth＇s biosphere is in perennially cold environments. Despite the extreme environmental conditions of desiccation and freezing, microbes can colonize these habitats through the adaptation of metabolic functions and the synthesis of structurally adapted enzymes. Enzymes within psychrophilic microbes exhibit high specific activity at low and moderate temperature, with low thermostability. In this study we used a classic microbiological approach to isolate Antarctic bacteria with cellulolytic, lipolytic, and ligninolytic activities. From 15 different environmental samples, we generated a collection of approximately 800 bacterial isolates that could grow on R2A or marine medium at 4℃. This collection was then screened for the presence of the three types of activity at 4℃. We found that 47.7% of the isolates displayed lipolytic activity, 10.2% had cellulase/xylanase activity, and 7.7% showed guaiacol oxidase activity. Of these, 10% displayed two different types of activity, while 0.25% displayed all three types of activity. Our results indicate that cold environments represent outstanding resources for bioprospecting and the study of enzymatic adaptation.

Optimization of large-scale supply chain planning models requires the application of decomposition strategies to reduce the computational expense. Two major options are to use either spatial or temporal Lagrangean decomposition. In this paper, to further reduce the computational expense a novel decomposition scheme by products is presented. The decomposition is based on a reformulation of knapsack constraints in the problem. The new approach allows for simultaneous decomposition by products and by time periods, enabling the generation of a large number of subproblems, that can be solved by using parallel computing. The case study shows that the proposed product decomposition exhibits similar performance as the temporal decomposition, and that selecting different orders of products and aggregating the linking constraints can improve the efficiency of the algorithm.

We studied the involvement of different types of Ca2+ channels, cyclic nucleotides and different kinases in the regulation of human umbilical artery (HUA) contractility. The elucidation of the precise mechanisms regulating the contractility of this artery could be very important to reveal potential therapeutic targets to treat HUA disorders such as preeclampsia. The relevancy of different types of Ca2+ channels on the regulation of HUA tonus was analyzed. Among the different Ca2+ channel inhibitors used, only the L-type calcium channels (LTCC) inhibition induced relaxation of HUA in Ca2+ containing medium. The inhibition of T-type calcium channels (TTCC) or TRP channels did not significantly affect HUA contractility. In presence of Ca2+, the intracellular increase of a cyclic nucleotide (cAmP or cGmP) induces relaxation of HUA, which was almost complete in histamine-con- tracted HUA, and lower effect was observed in arteries contracted by KCl and serotonin (5-HT). Inhibition of PKA and PKG weakly reduced the relaxations induced by the increase of cAmP and cGmP respectively, suggesting that the relaxation induced by these nucleotides is not totally mediated by the activation of their respective kinases and that other mechanisms are involved. In calcium containing solution, PP2A inhibition produces relaxation of contracted HUA. In KCl contracted arteries, the OA and nifedipine relaxant effects are similar and not additive, suggesting that PP2A could activate LTCC. Besides, the increase of cyclic nucleotides significantly increased the OA effect, suggesting that the effect of PP2A inhibition is independent of the cyclic nucleotide pathways. The contractions induced by KCl, histamine and 5-HT in presence of Ca2+ were not significantly affected by ROCK, ERK1/2 or p38mAPK inhibitors. The building as a model for conceiving a project： The future of the construction industry will be a science of what to do with the built environment rather than the classical problem of what is needed to be built. The necessary areas for living, working and entertainment are probably already built, but we need to update them to comply with the new codes, the new needs and the new uses of our society. For this reason we are looking at new ways of addressing our profession： Firstly we have to decide whether a new way of living requires a new space or if there is already an existing space available. Secondly, we have to adapt the chosen space to the new requirements of society and the new codes and regulations. Thirdly, we have to think about the sustainable and energetic needs, in order to update the building to the new efficiency requirements in terms of energy. Life cicle： We want to add a vision of life cycle to a building in terms of changing and updating. With this vision we are looking for ＂quality existing buildings＂ and for a way of designing ＂versatile frameworks＂ and layouts allowing for future renovation and refurbishments, Therefore our research is two pronged：（1） Identify examples of transformable buildings; （2） Looking for a design strategy that allows for easy changes of use.
