Hepatitis C virus (HCV) infects more than three million new individuals worldwide each year. In a high percent age of patients, acute infections become chronic, eventually progressing to fibrosis, cirrhosis, and hepatocellular carcinoma. Given the lack of effective prophylactic or therapeutic vaccines, and the limited sustained virological response rates to current therapies, new approaches are needed to prevent, control, and clear HCV *** into the host cell, being the first step of the viral cycle, is a potential target for the design of new antiviral compounds. Despite the recent discovery of the tight junction-associated proteins claudin-1 and occludin as HCV co-receptors, which is an important step towards the understanding of HCV entry, the precise mechanisms are still largely unknown. In addition, increasing evidence indicates that tools that are broadly employed to study HCV infection do not accurately reflect the real process in terms of viral particle composition and host cell phenotype. Thus, systems that more closely mimic natural infection are urgently required to elucidate the mechanisms of HCV entry, which will in turn help to design antiviral strategies against this part of the infection process.

Welding Effect at the Heat Affected Zone by Joining a Gray Cast Iron with Stainless Steel E308-16

Different investigations of the union of dissimilar materials such as stainless steel and different castings have been carried out, but rapid cooling immediately after welding has not been considered, in this work it was investigated how rapid cooling affects the metallurgical microstructure and consequently the mechanical properties. The effect of welding parameters on the microstructure and mechanical properties of the joint between dissimilar metals, an E-308-16 austenitic stainless steel and Gray Cast Iron was also analyzed. Gray cast iron samples (GCI) were fabricated, welded and cooled. The main welding parameters studied in this work are the welding technique and the type of filler electrodes. Flux-coated electrode E-308-16 was applied for this different joint. An experimental study was carried out for the analysis of welded joints of similar and dissimilar steels. The microstructure of the welded joints was analyzed using an optical microscope, in the base metals, heat affected zone (HAZ) and filler metal. The mechanical properties of the welded joints were evaluated by Vickers microhardness and tensile strength tests. The hardness profile showed differences in hardness between the base metals, the heat affected zone and the filler metal. The metallurgical microstructures observed along the welded areas corresponded to the profile. The hardness differences determined the effect on the mechanical and metallurgical characteristics of the welded samples as a result of the cooling rate differences. This research work is important because it allows us to analyze the possibility of reworking pieces of dissimilar materials by welding or, failing that, to determine if this may or may not be possible.

Depth of closure:New calculation method based on sediment data

Obtaining depth of closure(DoC) in an accurate manner is a fundamental issue for coastal engineering,since good results for coastal structures and beach nourishment depend mainly on DoC. Currently, there are two methods for obtaining the DoC, mathematical formulations and profile surveys. However, these methods can incur important errors if one does not take into account the characteristics and morphology of the area, or if one does not have a sufficiently long time series. In this work the DoC is obtained from the break in the trend of the sediment with the depth, that is, in general with the increase of the depth a decrease in the size of the sediment takes place. However, at one point this tendency changes and the size increases, and then decreases again. When comparing the point where the minimum sediment size occurs before the increase, it is observed that the error incurred is small compared to other methods. If the Standard Deviation of Depth Change(SDDC) method is considered as the most accurate method, the error incurred by the proposed method is less than 7%. In addition, it can be seen that the dispersion of the sediment method always occurs outside the zone of bar movement. Whereas in the methods of profiles survey(using 2 cm precision profiles), sometimes the DoC is obtained within the active zone of bar movement. In addition, where the relative minimum of the median sediment size is found, and the sizes of 0.063 and 0.125 mm predominate in the composition of the sample. Therefore, this new method allows the precise location of the DoC to be obtained in a fast and simple way. Furthermore, this method has the advantage that it is not affected by the modifications that may be experienced by both the study area and the cross-shore beach profile.

T follicular helper expansion and humoral-mediated rejection are independent of the HVEm/BTLA pathway

The molecular pathways contributing to humoral-mediated allograft rejection are poorly defined. In this study, we assessed the role of the herpesvirus entry mediator/B- and T-lymphocyte attenuator （HVEm/BTLA） signalling pathway in the context of antibody-mediated allograft rejection. An experimental setting was designed to elucidate whether the blockade of HVEm/BTLA interactions could modulate de novo induction of host antidonor-specific antibodies during the course of graft rejection. To test this hypothesis, fully allogeneic major histocompatibility complex-mismatched skin grafts were transplanted onto the right flank of recipient mice that were treated with isotype control, anti-CD40L or modulatory antibodies of the HVEm/BTLA signalling pathway. The frequencies of CD4 T follicular helper （Tfh） cells （B220-, CD4＋ CXCR5＋ PD-lhigh）, extrafollicular helper cells （B220-, CD4＋ CXCR5- PD-1＋ and PD-1-） and germinal centre （GC） B cells （B220＋Fas＋ GL7＋） were analysed by flow cytometry in draining and non-draining lymph nodes at day 10 post transplantation during the acute phase of graft rejection. The host antidonor isotype-specific humoral immune response was also assessed. Whereas blockade of the CD40/CD40L pathway was highly effective in preventing the allogeneic humoral immune response, antibody-mediated blockade of the HVEm/BTLA-interacting pathway affected neither the expansion of Tfh cells nor the expansion of GC B cells. Consequently, the course of the host antidonor antibody-mediated response proceeded normally, without detectable evidence of impaired development. In summary, these data indicate that HVEm/BTLA interactions are dispensable for the formation of de novo host antidonor isotype-specific antibodies in transplantation.

Z-line examination by the PillCam^(Tm) SB:Prospective comparison of three ingestion protocols

AIm:To evaluate the Z-line visualization by the PillCamTm SB2 using three different ingestion protocols. mETHODS:Ninety consecutive patients undergoing small bowel capsule endoscopy(SBCE)between January and may 2008 were included in the study. They swallowed the capsule in the standing(Group A= 30),supine(Group B=30)and right supine positions (Group C=30).Baseline patient characteristics, difficulties in capsule ingestion,esophageal transit times(ETT)and Z-line visualization were noted. RESULTS:No significant differences were found between the groups with regard to baseline patient characteristics,ingestion difficulties and complete SB examinations(P>0.05).At least 1 frame of the Z-line was detected in 15.8%,46.7%and 90%of patients in groups A,B and C,respectively(Pmber of Z-line images was 0.21±0.53,3.23 ±6.59 and 5.53±7.55 and the mean%of the Z-linedetected was 71.3,25.1 and 8.3,in groups A,B and C, respectively(both Pmes were longer in the supine group followed by the right supine and the standing groups(median of 237 s vs 64 s and 39 s, respectively;Pmost cases when the capsule is swallowed in the right supine position.

Using 172 plates taken with the 40-cm astrograph of the Sternberg Astronomical Institute （Lomonosov moscow University） in 1976-1994 and digitized with a resolution of 2400dpi, we dis- covered and studied 275 new variable stars. We present the list of our new variables with all necessary information concerning their brightness variations. As in our earlier studies, the new discoveries show a rather large number of high-amplitude Delta Scuti variables, predicting that many stars of this type re- main not detected in the whole sky. We also performed automated classification of the newly discovered variable stars based on the Random Forest algorithm. The results of the automated classification were compared to traditional classification and showed that automated classification was possible even with noisy photographic data. However, further improvement of automated techniques is needed, which is especially important when considering the very large numbers of new discoveries expected from all-sky surveys.
