Comparison of dissimilarity measures for cluster analysis of X-ray diffraction data from combinatorial libraries

Machine learning techniques have proven invaluable to manage the ever growing volume of materials research data produced as developments continue in high-throughput materials simulation,fabrication,and *** particular,machine learning techniques have been demonstrated for their utility in rapidly and automatically identifying potential composition-phase maps from structural data characterization of composition spread libraries,enabling rapid materials fabrication-structure-property analysis and functional materials discovery.A key issue in development of an automated phase-diagram determination method is the choice of dissimilarity measure,or kernel *** desired measure reduces the impact of confounding structural data issues on analysis *** issues include peak height changes and peak shifting due to lattice constant change as a function of *** this work,we investigate the choice of dissimilarity measure in X-ray diffraction-based structure analysis and the choice of measure’s performance impact on automatic composition-phase map *** dissimilarity measures are investigated for their impact in analyzing X-ray diffraction patterns for a Fe-Co-Ni ternary alloy composition *** cosine,Pearson correlation coefficient,and Jensen-Shannon divergence measures are shown to provide the best performance in the presence of peak height change and peak shifting(due to lattice constant change)when the magnitude of peak shifting is *** prior knowledge of the maximum peak shifting,dynamic time warping in a normalized constrained mode provides the best *** work also serves to demonstrate a strategy for rapid analysis of a large number of X-ray diffraction patterns in general beyond data from combinatorial libraries.

Characterization of NADase-Inactive NAD<sup>+</sup>Glycohydrolase in <i>Streptococcus pyogenes</i>

Background: Streptococcus pyogenes secretes NAD+ glycohydrolase (NADase, also known as SPN or Nga). All S. pyogenes strains examined to date possess the gene that encodes SPN (spn), but some strains produce SPN that lacks detectable NADase activity. Although there is much evidence to support that SPN’s NADase activity contributes to virulence, there is very little evidence that NADase-inactive SPN has detectable functions. Results: In order to characterize the NADase-inactive SPN, we firstly attempted to clone the NADase-inactive spn allele in Escherichia coli. Although we obtained recombinants which were shown to have the correct size insert, all had some mutations in the spn allele. Therefore, we attempted to change the mutated nucleotides back to the original nucleotides. While a nucleotide mutagenesis (inverse PCR method) easily changed a target nucleotide of control genes back to the original nucleotides, the mutations of NADase-inactive spn allele were never successfully converted back to the original nucleotides. Finally the mutant spn alleles were sub-cloned into another vector (pLZ12-Km2), which is maintained in both E. coli and S. pyogenes. The resultant plasmids were subjected to nucleotide mutagenesis using inverse PCR;the resultant mutagenized plasmid DNAs were used to transform both E. coli and S. pyogenes strains. We observed successful nucleotide substitutions back to the original spn nucleotide sequence in S. pyogenes transformants, but not in E. coli transformants. Thus, the NADase-inactive spn allele was successfully cloned in S. pyogenes, but not in E. coli. However, we could not find an association with NADase-inactive spn allele and virulence in a mouse infection model. Conclusions: These results suggest that NADase-inactive spn allele has some toxic effect to E. coli, but not S. pyogenes. This effect may due to an as of yet unknown function attributable to NADase-inactive SPN.

Dysregulation of Circular Antigen-Specific T Cells Anergy in Autoimmune Vitiligo

Background: Vitiligo is an autoimmune disorder related to melanocyte loss;however, the exact interplay between antigen-specific autoimmunity and local oxidative stress remains unclear. Recently, the migration ability and number of Foxp3-expressing regulatory T cells (Tregs) in lesional skin was found to be reduced in vitiligo patients. Objectives: We aimed to clarify the T cell anergy status of melanocytes by focusing on the impaired equivalence of peripheral melanocyte-specific cytotoxic T cells and functional Tregs in patients with progressive vitiligo. Materials and methods: Ten progressive vitiligo patients and 10 age-matched healthy individuals were enrolled in this study. We analyzed the number of functional Tregs in progressive vitiligo patients and compared the findings with those of controls. Next, to assess the suppressive activity of Tregs on melanocyte-specific T lymphocytes, we strictly purified the functional Tregs fraction and Melan-A-specific CD8+ T cells and co-cultured these cells with each other. The number of Melan-A-specific CD8+ T cells was then counted by FACS. In addition, the expression of the representative exhaustion markers PD-1 and CTLA-4 on functional Tregs was assessed in vitiligo patients and normal controls. Results: The number of functional Tregs itself was not significantly decreased in the blood of vitiligo patients compared to healthy controls. However, the cytotoxic T cell (CTL) proliferation was significantly decreased after cultivation with Tregs from healthy individuals (p < 0.01), and this decrease in CTLs was less marked after cultivation with Tregs from vitiligo patients. Conclusions: We demonstrated a reduced suppressive function of activated Tregs on Melan-A-specific CTLs in the circulating cells of vitiligo patients compared with healthy controls. This result suggests that T cell anergy with Tregs dysfunction may participate in the immune response to melanocytes in vitiligo patients.

Synthesis of powdered silver nanoparticles using hydrogen in aqueous medium

Environmentally benign one-pot protocol is used in the synthesis of silver nanoparticles （AgNPs） in powder form, involving the reduction of silver nitrate in the presence of fully hydrolyzed polyvinyl alcohol （PVA） as stabilizer by hydrogen （H2） as reducing agent in aqueous medium. Fully hydrolyzed biodegradable PVA has extremely low cytotoxicity, making it a favorable stabilizer from green perspective. In the present methodology, essentially naked silver nanoparticles are obtained in good yield. The prepared sil- ver nanoparticles were characterized by TEM, XRD, EDAX, UV-Vis spectroscopy and DLS histogram, and studied for its activity as a recyclable catalyst for synthesis of different enaminones.

A New Therapy for Human Endometriosis: The Therapeutic Value of Leukotriene Receptor Antagonist for Endometriosis

Some drugs that regulate estrogen are currently used as therapy for endometriosis. However, these medical treatments have significant side effects and patients who hope to become pregnant cannot overcome infertility. To compare morphological alterations and clinical symptoms, revised American Fertility Society (r-AFS) scores were compared between patients with and without leukotriene receptor antagonist (LTR-A) treatment. LTR-A-treated cases showed significantly decreased r-AFS score. Furthermore, a significant correlation was seen between r-AFS score and LTR-A treatment period. In treated cases, clinical symptoms decreased and some patients achieved pregnancy. Morphologically, lesions in LTR-A-treated cases showed apoptotic fibroblasts and degeneration of collagen fibers. Our findings revealed that LTR-As had significant therapeutic value for the treatment of human endometriosis. Anti-LT therapy appears efficacious not only in the treatment of clinical symptoms and lesions, but also in improving fertility.

A significant gap in the middle Palaeozoic apparent polar wander(APW) path precludes polarity definition of the early Palaeozoic palaeopole for North *** problem can in part be resolved by the intercontinental correlation of magnetic polarity patterns across small time intervals.A magnetostratigraphic study was carried out on upper Cambrian to lower Ordovician sediments near Zhaogezhuang(long.118.5°E,lat.39.7°N),North *** stepwise thermal or thermal and alternating field demagnetizations,a characteristic magnetic component with normal and antipodal reversed directions was *** data,drawn from 49 samples,yield a north palaeopole at long.294.6°E,lat.32.9°N(dp=3.0°,dm=5.3°).A concordant magnetic polarity pattern around the Cambrian-Ordovician boundary and lowest Ordovician obtained from different continents favours a Southern Hemisphere origin(□17.) formation *** the Cambrian-Ordovician APW paths between North China and Gondwana,we suggest that the North China block(NCB) was part of Gondwana during the Cambro-lowest Ordovician,and started breaking away from Gondwana in the lower *** finding is contrary to some palaeomagnetic models where the NCB was quite separate from Gondwana in the late Proterozoic, and was attached to the‘Pacific’ side of Antarctica.

Joint Operation of Atomic Force Microscope and Advanced Laser Confocal Microscope for Observing Surface Processes in a Protein Crystal

We demonstrated the insitu observation of a moving atomic force microscope (AFM) cantilever using a laser confocal microscope combined with a differential interference microscope (LCM-DIM). The AFM cantilever scanned or indented the {110} surface of a hen egg-white lysozyme crystal in a supersaturated solution. Using a soft cantilever, we could observe the step growth with high time resolution by LCM-DIM and perform quantitative measurements of the step height by AFM simultaneously. In addition, a hard cantilever was used with LCM-DIM to observe the dynamics of crystal surface scratching and indentation. In the supersaturated solution, the small steps generated from the scratched line aggregated to macro steps, and subsequently flattened the surface.
