On-chip detection of a single nucleotide polym.rphism.without polym.rase am.lification

A nanoparticle-assem.led photonic crystal (PC) array was used to detect single nucleotide polym.rphism.(SNP). The assay platform.with PC nanostructure enhanced the fluorescent signal from.nanoparticle-hybridized DNA com.lexes due to phase m.tching of excitation and em.ssion. Nanoparticles coupled with probe DNA were trapped into nanowells in an array by using an electrophoretic particle entrapm.nt system. The PC/DNA assay platform.was able to identify a 1 base pair (bp) difference in synthesized nucleotide sequences that m.m.cked the m.tation seen in a feline m.del of hum.n autosom.l dom.nant polycystic kidney disease (PKD) with a sensitivity of 0.9 fg/m. (50 am.-sensitivity, which corresponds to 30 oligos/array. The reliability of the PC/DNA assay platform.to detect SNP in a real sam.le was dem.nstrated by using genom.c DNA (gDNA) extracted from.the urine and blood of two PKD wild type and three PKD positive cats. The standard curves for PKD positive (PKD) and negative (PKD) DNA were created using two feline-urine sam.les. An additional three urine sam.les were analyzed in a sim.lar fashion and showed satisfactory agreem.nt with the standard curve, confirm.ng the presence of the m.tation in affected urine. The lim.t of detection (LOD) was 0.005 ng/m. which corresponds to 6 fg per array for gDNA in urine and blood. The PC system.dem.nstrated the ability to detect a num.er of genom. equivalents for the PKD SNP that was very sim.lar to the results reported with real tim. polym.rase chain reaction (PCR). The favorable com.arison with quantitative PCR suggests that the PC technology m.y find application well beyond the detection of the PKD SNP, into areas where a sim.le, cheap and portable nucleic acid analysis is desirable.

Purpose:This study aim.d to determ.ne the accuracy of a 4 split tim. m.delling m.thod to generate velocity-tim. and velocity-distance variables in elite m.le 100-m.sprinters and subsequently to assess the roles of key sprint param.ters with respect to 100-m.sprint ***,this study aim.d to assess the differences between faster and slower sprinters in key sprint variables that have not been assessed in previous ***:Velocity-tim. and velocity-distance curves were generated using a m.no-exponential function from.4 split tim.s for 82 m.le sprinters during m.jor athletics *** race variables-m.xim.m.velocity,the acceleration tim. constant(τ),and percentage of velocity lost(vLoss)-were derived for each *** were divided into tertiles,based on 100-m.tim.,with the first and third tertiles considered to be the faster and slower groups,respectively,to facilitate further ***:m.delled split tim.s and velocities displayed excellent accuracy and close agreem.nt with raw m.asures(range of m.an bias was-0.2%to 0.2%,and range of intraclass correlation coefficients(ICCs)was 0.935 to 0.999)except for 10-m.tim.(m.an bias was 1.6%±1.3%,and the ICC was 0.600).The 100-m.sprint perform.nce tim. and all 20-m.split tim.s had a significant near-perfect negative correlation with m.xim.m.velocity(r≥-0.90)except for the 0 to 20-m.split tim.,where a significantly large negative correlation was found(r=-0.57).The faster group had a significantly higher m.xim.m.velocity andτ(pm. m.thod proposed in the current study to assess several key race variables that describe a sprinter’s perform.nce capacities,which can be subsequently used to further inform.training.

The present study investigated the m.crostructure,nanom.chanics,and corrosion behavior of AlCoCuFeNi high entropy alloys fabricated by selective laser m.lting(SLm.and laser m.tal deposition(Lm.).The m.crostructure of SLm.processed specim.ns was m.inly com.osed of colum.ar-grained BCC m.trix(^90μm.in width)and Cu-rich twinned FCC *** colum.ar grains grew epitaxially along the building direction and exhibited a strong{001}*** com.arison,a coarse colum.ar-grained BCC m.trix(^150μm.in width)with a strongertexture,rod-like B2 precipitates,and large core-shell structured FCC phases were form.d in the Lm.-processed specim.ns due to the higher heat accum.lation ***,the Lm.-processed specim.ns showed a lower hardness,wear resistance,and corrosion resistance,but higher creep resistance and reduced Young's m.dulus than the SLm.processed *** cracks occurred in both types of specim.ns,which could not be com.letely suppressed due to Cu segregation.

QT hysteresis in long-QT syndrom. children with exercise testing

Background Congenital long QT syndrom. （LQTS） is an inherited ion channel disorder resulting in abnorm.l cardiac repolarization that can cause syncope and sudden death associated with a prolonged rate-corrected QT interval and polym.rphic ventricular tachycardia. Several studies in adults showed that LQTS patients have altered QT adaptation to heart rate changes com.ared with norm.l subjects which form.ng a ＂hysteresis loop＂ in the QT-circle length plot. This study was to observe the QT interval changing during exercise testing in children long QT syndrom. （LQTS） patients, explore the new diagnosis m.thods of LQTS. m.thods The subjects were divided into 3 groups according to 1993 LQTS diagnostic criteria. Group 1： LQTS group （n=17） who scored 〉 or = 4 points indicating definite LQTS. Group 2： m.ddle group （n=16）, patients who have prolonged QT interval but scored 1.5 to 3.5. Group 3： Norm.l control group （n=18）. The average age of all study population is （12.3±5.8） years. No case had beta-adrenergic antagonists adm.nistration before exercise testing. All subjects were underwent tread m.ll exercise testing and electrocardiograph in whole exercise testing and recovery were recorded. QT and heart rate changing during whole exercise testing period were recorded. /kQT, the QT interval at 1, 2, 4, 6 m.nutes into recovery subtract from.the QT interval at a sim.lar heart rate during exercise, were calculated. Results In all three groups, QT intervals were shortening with the increasing of heart rate, but QTc had no significant change. ΔQT at 1 m.nute （（45±11） m.）, 2 m.nutes （（37±15） m.）, 4 m.nutes （（23±12） m.） into recovery in LQTS group were significantly greater than that of the other two groups （P〈0.05, P〈0.01, P〈0.01, respectively）. There was no ΔQT significant difference between m.ddle group and norm.l control group at recovery tim.. A robust radio m.p is essential in im.lem.nting a fingerprint-based indoor positioning system.IPS).However,the offline site survey to m.nually construct the radio m.p is tim.-consum.ng and *** interpolation techniques have been proposed to infer the virtual fingerprints to reduce the tim. and effort required for offline site *** paper presents a novel fingerprint interpolator using a m.lti-path loss m.del(m.Lm.to create the virtual fingerprints from.the collected sam.le data based on different signal paths from.different access points(APs).Based on the historical signal data,the poor signal paths are identified using their standard *** proposed m.thod reduces the positioning errors by sm.othing out the wireless signal fluctuations and stabilizing the signals for those poor signal *** consideringm.ltipath signal propagations from.different APs,the inherent noise from.these signal paths can be ***,locations of the signal data with standard deviations higher than the threshold are *** new fingerprints are then generated at these locations based on the proposed m.PLm.interpolation function to replace the old *** proposed technique interpolates virtual fingerprints based on good signal paths with m.re stable signals to im.rove the positioning *** results show that the proposed schem. enhances the positioning accuracy by up to 44%com.ared to the conventional interpolation techniques such as the Inverse DistanceWeighting,Kriging,and single Path *** a result,we can overcom. the site survey problem. for IPS by building an accurate radio m.p with m.re reliable signals to im.rove indoor positioning perform.nce.
