Cytokinin levels and signaling respond to wounding and the perception of herbivore elicitors in Nicotiana attenuata

Nearly half a century ago insect herbivores were found to induce the formation of green islands by manipulating cytokinin （CK） levels. However, the response of the CK pathway to attack by chewing insect herbivores remains unclear. Here, we characterize the CK pathway of Nicotiana attenuata （torr. ex S. Wats.） and its response to wounding and perception of herbivoreassociated molecular patterns （HAMPs）. We identified 44 genes involved in CK biosynthesis, inactivation, degradation, and signaling. Leaf wounding rapidly induced transcriptional changes in multiple genes throughout the pathway, as well as in the levels of CKs, including isopentenyladenosine and cis-zeatin riboside; perception of HAMPs present in the oral secretions （OS） of the specialist herbivore Manduca sexta amplified these responses. the jasmonate pathway, which triggers many herbivore-induced processes, was not required for these HAMP-triggered changes, but rather suppressed the CK responses. Interestingly CK pathway changes were observed also in systemic leaves in response to wounding and OS application indicating a role of CKs in mediating long distance systemic processes in response to herbivory. Since wounding and grasshopper OS elicited similar accumulations of CKs in Arabidopsis thaliana L., we propose that CKs are integral components of wounding and HAMP-triggered responses in many plant species.

Measuring NO Production by Plant tissues and Suspension Cultured Cells

We describe an inexpensive and reliable detector for measuring NO emitted in the gas phase from plants. the method relies on the use of a strong oxidizer to convert NO to NO2 and subsequent capture of NO2 by a Griess reagent trap. the set-up approaches the sensitivity for NO comparable to that of instruments based on chemiluminescence and photoacoustic detectors. We demonstrate the utility of our set-up by measuring NO produced by a variety of well established plant sources. NO produced by nitrate reductase （NR） in tobacco leaves and barley aleurone was readily detected, as was the production of NO from nitrite by the incubation medium of barley aleurone. Arabidopsis mutants that overproduce NO or lack NO-synthase （AtNOS1） also displayed the expected NO synthesis phenotype when assayed by our set-up. We could also measure NO production from elicitor-treated suspension cultured cells using this set-up. Further, we have focused on the detection of NO by a widely used fluorescent probe 4-amino-5-methylamino-2＇,7＇-difluorofluorescein （DAF-FM）. Our work points to the pitfalls that must be avoided when using DAF-FM to detect the production of NO by plant tissues. In addition to the dramatic effects that pH can have on fluorescence from DAF-FM, the widely used NO scavengers 2-phenyl- 4,4,5, 5-tetramethylimidazoline- 1-oxyl-3-oxide （PtIO） oxide （cPtIO） can produce anomalous and unexpected to bind to dead cells and remain NO-sensitive. and 2-（4-carboxyphenyl）-4,4,5,5-tetramethylimidazoline- 1-oxyl-3- results. Perhaps the most serious drawback of DAF-FM is its ability to bind to dead cells and remain NO-sensitive.

Vulvar basal cell carcinoma: A retrospective study of 29 cases from Queensland

Objective: Review the clinical features, diagnosis, management and outcomes for 29 cases of Basal Cell Carcinoma (BCC) of the Vulva referred to Queensland Centre for Gynaecological Cancer (QCGC) between 1986 and 2010. Methods: Vulvar BCC cases from QCGC were reviewed and analysed using the computer software Statistical Package for the Social Sciences (SPSS) 11.0. Results: BCC of the vulva is uncommon with an incidence from the QCGC vulvar cancer registry of 3.2%. Of the 29 patients one died of their BCC and seven died of unrelated causes. the mean age at diagnosis was 69.5 years (range 40 to 91). All cases were Caucasian. time from onset of symptoms to diagnosis averaged 22.6 months (range 0 - 120 months). Not until a biopsy was performed was the diagnosis made. the most common presenting complaints were pruritis and a lump. Initial treatment was surgical. Conclusions: the prognosis for vulvar BCC is excellent. Histological diagnosis and long term follow-up are important management issues. the status of disease at the margins of surgical specimens does not reliably equate to patient long term outcomes. Follow up should be supervised via a gynecological oncology register to reduce the risk of patient loss to follow up.

Bartholin’s Gland Carcinomas: A 20 plus-year experience from Queensland

Objective: to review the records of cases of Bartholin’s Gland Carcinoma referred to the Queensland Centre for Gynaecological Cancer (QCGC) between mid 1993 and mid 2012. Methods: Bartholin’s Gland Carcinoma case data from QCGC were reviewed and analysed using the computer software Statistical Package for the Social Sciences 11.0. Results: Of the 12 cases four died of their disease, seven are still alive and disease free and one is alive with recurrent disease. the mean age at diagnosis was 52.8 years. time from onset of symptoms to diagnosis averaged 5.8 months. All diagnoses were confirmed histologically. Presenting symptoms included a lump and pain. the most common presenting complaint was a lump. treatment included surgical excision, occasional biopsy followed by radiotherapy with or without chemotherapy. In some cases radiation and chemotherapy was followed by vulvectomy of various extent. Conclusions: Bartholin’s Gland Carcinoma is a rare condition with outcome dependent on duration of symptoms, including delay in diagnosis, cell-type, cellular differentiation and the International Federation of Gynecology and Obstetrics (FIGO) classification. A Bartholin’s gland mass in a woman aged 40 years or more should be considered malignant until a biopsy proves *** incidence of Bartholin’s Gland Carcinoma in Queensland is less than that reported elsewhere but a higher proportion of squamous cell carcinomas was found in this small series.

Vulvar sarcomas: A 25 plus-year experience from Queensland

Objective: to review the characteristics of cases of vulvar sarcomas (VS) referred to the Queensland Centre for Gynaecological Cancer (QCGC) between mid 1984 and mid 2012. Methods: VS case data from QCGC were reviewed and analysed using the computer software Statistical Package for the Social Sciences (SPSS) 11.0. Results: Of the nine cases three died of their disease, one died of ovarian cancer and five are still alive and disease free. the mean age at diagnosis was 54.8 years. time from onset of symptoms to diagnosis averaged one month. All diagnoses were confirmed histologically. the most common presenting complaint was a lump. treatment in all but one case involved surgical excision, one case had chemotherapy alone and two cases had surgery followed by radiotherapy. Conclusions: to talk of five year survival for patients with such a rare cancer, so many histological types with variable cell differentiation plus the unknown of when sarcomatous cells metastasize from even early FIGO staged lesions is in our opinion taking generalisation to far. A multidisciplinary approach to management achieves the best results for each individual and pooling multicentre data will improve our understanding of this rare disease and benefit future patients.

Apoptosis block as a barrier to effective therapy in non small cell lung cancer

Lung cancer, is the most common cause of cancer death in men and second only to breast cancer in women. Currently, the first line therapy of choice is platinumbased combination chemotherapy. A therapeutic plateau has been reached with the prognosis for patients with advanced non-small cell lung cancer(NSCLC) remaining poor. New biomarkers of prognosis as well as new therapies focusing on molecular targets are emerging helping to identify patients who are likely to benefit from therapy. Despite this, drug resistance remains the major cause for treatment failure. In this article we review the role of apoptosis in mediating drug resistance in NSCLC. Better understanding of this fundamental biological process may provide a rationale for overcoming the current therapeutic plateau.

A new cache of Eoarchaean detrital zircons from the Singhbhum craton,eastern India and constraints on early Earth geodynamics

the dominant geodynamic processes that underpin the formation and evolution of Earth’s early crust remain enigmatic calling for new information from less studied ancient cratonic ***,we present U-Pb ages and Hf isotopic compositions of detrital zircon grains from^2.9 Ga old quartzites and magmatic zircon from a 3.505 Ga old dacite from the Iron Ore Group of the Singhbhum craton,eastern *** detrital zircon grains range in age between 3.95 Ga and 2.91 *** with the recently reported Hadean,Eoarchean xenocrystic(up to 4.24 Ga)and modem detritus zircon grains from the Singhbhum craton,our results suggest that the Eoarchean detrital zircons represent crust generated by recycling of Hadean felsic crust formed at^4.3-4.2 Ga and^3.95 *** observe a prominent shift in Hf isotope compositions at^3.6-3.5 Ga towards super-chondritic values,which signify an increased role for depleted mantle and the relevance of plate *** Paleo-,Mesoarchean zircon Hf isotopic record in the craton indicates crust generation involving the role of both depleted and enriched mantle *** infer a short-lived suprasubduction setting around^3.6-3.5 Ga followed by mantle plume activity during the Paleo-,Mesoarchean crust formation in the Singhbhum *** Singhbhum craton provides an additional repository for Earth’s oldest materials.

Aerogel materials manufactured from metal oxides have been used as components in numerous high-energy density physics targets. these aerogels have been identified to be used as a future target material in the AWE fielded campaigns at the US National Ignition Facility. A wide variety of metal oxide aerogels are required for future campaigns and therefore a versatile manufacturing route is sought; as such, an epoxide-assisted sol–gel route was investigated. Under the European Union Registration, Evaluation, Authorization and Restriction of Chemicals legislation, the most commonly used epoxide, propylene oxide, is recognized as a substance of very high concern(SVHC). this work sought to investigate suitable alternative epoxides for use in target manufacture. the outcome was the identification of synthesis routes for stable metal oxide aerogel monoliths using epoxides not subject to the above restrictions.

Semiautomated Class Attendance Monitoring Using Smartphone technology

Class attendance is *** attendance recording is often done using“roll-call”or signing attendance *** are time consuming,easy to cheat,and it is difficult to draw any information from *** are other,expensive alternatives to automate attendance recording with varying *** study experimented with a smartphone camera and different combinations of face detection and recognition algorithms to determine if it can be used to record attendance successfully,while keeping the solution *** effect of different class sizes was also *** research was done within a pragmatism philosophy,using a prototype in a field *** algorithms that were used are Viola–jones(Haar features),deep neural network and histogram of oriented gradients for detection,and eigenfaces,fisherfaces,and local binary pattern histogram for *** best combination was Viola–jones combined with fisherfaces,with a mean accuracy of 54%for a class of 10 students and 34.5%for a class of 22 *** best all over performance on a single class photo was 70%(class size 10).As is,this prototype is not accurate enough to use,but with a few adjustments,it may become a cheap,easy-to-implement solution to the attendance recording problem.
