SARS-CoV-2 Pooled Testing Methodology for PCR Testing Applied in Private Laboratory in Armenia

Since the beginning of COVID-19 pandemics many countries were facing challenges with testing capacity recourse limitations. Throughout the waves of the pandemic countries were trying to address the existing constrains exploring solutions to increase the testing capacity with more cost-effective approaches. Pooled methodology was one of the methods which many have validated and used. It is evident that in case of pooled sample testing the sensitivity becomes lower, however the variation highly depends on the pool size as well as the incidence rate at the certain point. Armenia as well as many other countries has adopted regulations for mandatory COVID-19 PCR testing for all the travelers. Current study aimed to explore the efficiency of COVID-19 pooled PCR testing for nasopharyngeal swabs of individuals with no symptoms in a time period with good epidemiological state of the infection. Nasopharingeal swab samples from individuals were collected. The manual extraction of RNAs of samples was performed after pooling up to 5 samples. The pools with Cycle Threshold (CT) of < 37 were considered positive and were retested individually. In total 28,015 samples were grouped in 667 pools of which 57 were positive. The total number of positive samples was 65. The median difference (CT-pool–CT samples) was 2.4 (ranging from–3.0 to 8.9). The correlation of CT of pools and positive samples was positive. The correlation coefficient r = 0.84, P < 0.000, 95% CI range 0.7423 to 0.9243). The total economic saving when using pools compared to the individual testing was 72%. The minor difference between CT values of pools and samples can be explained by the dilution effect in the pool. However, the positive correlation between the values as well as the amount of cost saving demonstrate that pooling on nasopharyngeal samples for COVID-19 PCR testing can be a good method for efficient screening with significant resource saving. Introduction: Sleep disorders among professional drivers are a major road safety problem. They cause fatigue and drowsiness at the wheel, which can lead to road traffic accidents and even accidents at work. The aim of this study was to assess sleep disorders among professional drivers of oil tankers in Ouagadougou, Burkina Faso. Methods: This was a descriptive and analytical cross-sectional study carried out from 11 April 2020 to 11 September 2020 in the national hydrocarbon company’s fuel depot. All the drivers present at the time were included and accomplished a questionnaire incorporating the commonly scales using to assess sleep disorders, sleep quality and sleep apnoea. Data analysis using R 3.6.1 software enabled to perform univariate and multivariate analyses to identify associated factors. Adjusted odd ratios were used to measure the strength of association. The significance level chosen was p ≤ 0.05. Results: All the 339 respondents were male. A proportion of 33.6% suffered from insomnia, including 12.7% with moderate to severe insomnia, and 18.58% were at risk of sleep apnoea. On the Epworth scale, 26.6% of drivers showed sleep debt and 4.1% excessive daytime sleepiness. The risk factors associated with insomnia were the use of psychostimulants. A history of diabetes and insomnia were risk factors associated with sleep apnoea syndrome. Conclusion: The prevalence of sleep disturbance and insomnia among the respondents were 52.2% and 33.6% respectively. Use of psychostimulants was the identified risk factor. The prevalence of SAS was 4.72%, with diabetes and insomnia as associated factors.

Optic nerve sheath diameters in healthy adults measured by computer tomography

AIMTo measure optic nerve sheath diameters (ONSD) in different locations by computer tomography (CT) and to recommend the best location for cases when ONSD is used for intracranial pressure *** a prospective cohort study, CT data of 300 healthy adults were analyzed (600 eyes). In all cases, the CT investigation was performed at the Emergency Department because of the various conditions that proved not to be connected with ophthalmological or neurological pathology. The ONSD were measured at 3 mm and 8 mm distance from the globe, and 3 mm from the anterior opening of the optic canal. The correlation analysis was performed with gender, age, and ethnic *** right/left ONSD are 4.94±1.51/5.17±1.34 mm at 3 mm, 4.35±0.76/4.45±0.62 mm at 8 mm from the globe, and 3.55±0.82/3.65±0.7 mm at 3 mm from the optic canal. No significant differences correlated with gender of the patients, their age, and ethnic background were *** healthy persons, the ONSD varies from 5.17±1.34 mm to 3.55±0.82 mm in different locations within the intraorbital space. The most stable results with lesser standard deviation can be obtained if it is measured 8-10 mm from the globe.

New GATEWAY vectors for High Throughput Analyses of Protein-Protein Interactions by Bimolecular Fluorescence Complementation

Complex protein interaction networks constitute plant metabolic and signaling systems. Bimolecular fluorescence complementation （BiFC） is a suitable technique to investigate the formation of protein complexes and the localization of protein-protein interactions in planta. However, the generation of large plasmid collections to facilitate the exploration of complex interaction networks is often limited by the need for conventional cloning techniques. Here, we report the implementation of a GATEWAY vector system enabling large-scale combination and investigation of candidate proteins in BiFC studies. We describe a set of 12 GATEWAY-compatible BiFC vectors that efficiently permit the combination of candidate protein pairs with every possible N-or C-terminal sub-fragment of S（CFP）3A or Venus, respectively, and enable the performance of multicolor BiFC （mcBiFC）. We used proteins of the plant molybdenum metabolism, in that more than 20 potentially interacting proteins are assumed to form the cellular molybdenum network, as a case study to establish the functionality of the new vectors. Using these vectors, we report the formation of the molybdopterin synthase complex by interaction of Arabidopsis proteins Cnx6 and Cnx7 detected by BiFC as well as the simultaneous formation of Cnx6/Cnx6 and Cnx6/Cnx7 complexes revealed by mcBiFC. Consequently, these GATEWAY-based BiFC vector systems should significantly facilitate the large-scale investigation of complex regulatory networks in plant cells.

Hepatocellular carcinoma （HCC） is the fish most common cancer in the world. This case documents an unusual metastatic presentation of HCC in the humerus. Preoperative palliative arterial embolization of the tumor was performed to arrest severe tumor bleeding caused by the biopsy. Embolization turned out to be useful also in limiting/preventing potential uncontrolled bleeding during subsequent amputation.

Serological Investigation of COVID-19 Antibodies in Armenia

Background: In December 2019 in Wuhan China the new coronavirus outbreak emerged and quickly spread in all parts of the world resulting to more than 500,000,000 infection cases and around 6,200,000 deaths. The global incidence of the infection is still growing as well as number of deaths. COVID-19 is a new virus, therefore not much is known about the immune response of infected organism, which is crucial not only for vaccination policy development, but also for identification of public health strategies. Aim: Current research aims to describe COVID-19 IgG levels depending on symptoms, antibiotic and antiviral medications intake history, existing chronic condition and smoking status during March-December of 2020 in Armenia. Furthermore, the study aims to help elucidate the fraction of asymptomatic or presymptomatic/sub- clinical infections in the population and understand the main risk factors for infection complication. Methodology: The cross-sectional study with convenience sampling of individuals who turned to “EcoSense” laboratories to be tested for COVID-19 IgG were examined. The NovaTec SARS-CoC-2 (COVID-19) IgG COVG940 96 Determinations ELISA test kits were used. The questionnaire was filled regarding the COVID-19 status, symptoms, exposure history, disease history, pre-existing chronic conditions, medication and vaccination history. The descriptive as well as multivariate analysis was performed. Results: Overall 1573 testing was performed 837 of subjects agreed to participate in the interview. 24.1% of participants had laboratory confirmed COVID-19 but by the time of interview were already recovered. 212 (25.3%) participants had positive IgG levels, among 126 (15.1%) participants IgG levels were in the grey zone. Out of PCR confirmed cases only 58.7% had positive IgG levels and 3.9% IgG level was in the grey zone. First-Eutectic Features of Polar Metals in Double Systems

It is known that the dense part of any liquid metal consists of ramified clusters of almost regular tetrahedrons (triangular pyramids with atoms in their vertexes) that are connected into chains by faces. Any metal additive as a second component of liquid alloy can be both beyond these clusters as separated atoms and into them as inherent clusters. The liquid-metal alloy transfers into the second state, at the first eutectic of the solvent. This polymorphic transition of liquid matrix is discovered in the systems, Pb-K and Na-Pb, by molecular-dynamic simulating their microstructure and in experiments on scattering slow neutrons by these alloys of different compositions. In the first system, the obtained results identify both the homogeneous alloy at low concentrations of potassium in liquid lead and the alloy clustering, (Pb4K)n, at potassium concentrations following the eutectic, Pb0.91K0.09. In the second one at the concentrations of lead more than 2%, just the second state is discovered with the clusters, (Na4Pb)n. One can expect the same polymorphic transition in the eutectic, Na0.93Tl0.07, with the micro-inhomogeneity, (Na6Tl)n, and with the melting point of 64 C. This eutectic maintained by the oxygen-free technology and enriched by the isotope, 205Tl, can become the best coolant for fast nuclear reactors due to the depressed chemical activity of sodium and composition stability.
