Chemical, isotopic and amino acid composition of Mukundpura CM2.0(CM1) chondrite: Evidence of parent body aqueous alteration

The carbonaceous chondrites are intriguing and unique in the sense that they are the only rocks that provide pristine records of the early solar nebular processes. We report here results of a detailed mineralogical, chemical, amino acid and isotopic studies of a recently observed fall at Mukundpura, near Jaipur in Rajasthan, india. Abundance of olivines in this meteorite is low and of serpentine minerals is high. FeO/SiO_2 = 1.05 in its Poorly Characterized Phases(PCP) is similar to that observed in other CM2.0 chondrites. The water content of ～9.8 wt.% is similar to that found in many other CM *** examination of matrix shows that its terrestrial weathering grade is WO but aqueous parent body alteration is high, as reflected in low abundance of identifiable chondrules and abundant remnants of chondrules(～7%). Thus, most of the chondrules formed initially have been significantly altered or dissolved by aqueous alterations on their parent bodies. The measured bulk carbon(2.3%) and nitrogen content and their isotopic(δ^(13)C =-5.5‰, δ^(15)N = 23.6%0) composition is consistent with CM2.0 classification probably bordering CM1. Several amino acids such as Alanine, Serine, Proline, Valine, Threonine,Leucine, isoleucine, Asparagine and Histamine are present. Tyrosine and Tryptophan may occur in trace amounts which could not be precisely determined. All these data show that Mukundpura chondrite lies at the boundary of CM2.0 and CM1 type carbonaceous chondrites making it one of the most primitive chondrites.

Microsatellite markers were developed for Samadera bidwillii, a nationally listed vulnerable shrub or small tree, to enable investigation of its genetic structure and diversity within and among populations from its known distribution throughout coastal areas mainly in fragmented and disturbed lands from Mackay to Gympie, Queensland, Australia. The loci were tested for cross-amplification in related Samadera species. Ten polymorphic microsatellite markers were isolated and characterised from an enriched library of S. bidwillii, which exhibited di- and trinucleotide repeats. The mean number of alleles per locus ranged from 1.3 to 2.5 and mean expected and observed heterozygosities ranged from 0.06 to 0.33 and from 0.03 to 0.26, respectively in five populations. All loci successfully amplified in six other closely associated Samadera species also reported from Australia. Developed loci can be used in genetic diversity, population structure and gene flow studies with an emphasis on the conservation of S. bidwillii and related species.

Distribution of gymnemic acid in various organs of Gymnema sylvestre

The gymnemic acid contents in various organs of Gymnema sylvestre were investigated by High Performance Liquid Chromatography （HPLC） method. The results shows that the content of gymnemic acid in various organs, obviously different, was 54.29, 31.66, 28.82, 27.67, 25.39, 20.56 and 1.31 mg.g^-1 DW in shoot tips, flowers, nodes, leaves, internodes, roots and seeds, respectively. The highest gyrrmemic acid content （54.29 mg.g^-1 DW） was found in shoot tip, 1.96 fold higher than that in leaves （27.67 mg.g^-1DW）. Maximum quantity of gymnemic acid （35.39 mg·g^-1 DW） was observed in the young leaves, which was 1.52 times higher than that in old leaves （23.07 mg.g^-1DW）. The content of gymnemic acid in young, middle and old internodes was 26.47, 25.77 and 23.94 mg.g^-1 DW, respectively, all lower than that in leaves （27.67 mg.g^-1 DW）, whereas the content of gyirmemic acid in young, middle and old nodes was 27.96, 28.81 and 29.66 mg.g^-1 DW, respectively, all higher than that in leaves. The study provides the scientific evidences for the rational development and utilization of Gymnema sylvestre resources, since over exploitation of natural stands has caused depletion of these plants in nature.

Mode of interaction of calcium oxalate crystal with human phosphate cytidylyltransferase 1: a novel inhibitor purified from human renal stone matrix

Nephrolithiasis is a common clinical disorder, and calcium oxalate (CaOx) is the principal crystalline component in approximately 75% of all renal stones. it is widely believed that proteins act as inhibitors of crystal growth and aggregation. Acidic amino acids present in these proteins play a significant role in the inhibition process. in this study, interaction of cal-cium oxalate with human phosphate cytidylyltrans-ferase 1(CCT), a novel calcium oxalate crystal growth inhibitor purified from human renal stone matrix has been elucidated in silico and involvement of acidic amino acids in the same. As only sequence of CCT is available, henceforth its 3-D structure was modeled via Homology modeling using Prime module of Schrodinger package. Molecular dynamic simulation of modeled protein with solvation was done by mac-romodel (Schrodinger). The quality of modeled pro-tein was validated by JCSG protein structure valida-tion (PROCHECK & ERRAT) server. To analyze the interaction of modeled protein CCT with calcium oxalate along with role played by acidic amino acids, ‘Docking simulation’ was done using MOE–Dock. interaction between calcium oxalate and CCT was also studied by substituting acidic amino acid in the active sites of the protein with neutral and positively charged amino acids. The in silico analysis showed the bond formation between the acidic amino acids and calcium atom, which was further substantiated when substitution of these acidic amino acids with alanine, glycine, lysine, arginine and histidine com-pletely diminished the interaction with calcium ox-alate.

Aim: To identify, analyse and control the outbreak of Carbapenamase producing Klebsiella pneumoniae. Objectives: 1) To detect multidrug resistant K. pneumoniae at the earliest and isolate patients. 2) To find out the predisposing causes for the occurrence of this outbreak. 3) To break the chain of infection transmission. 4) To reduce the risk of Hospital acquired infections. Methods: This retrospective study along with the surveillance was conducted from January 2017 to March 2017 at HCG multispecialty cancer hospital, Bangalore, india. Results: Total 15 patients were diagnosed with KPC infection during the first month of the study period. Those affected were mostly Male patients (73%), admitted in iCU (73%) for further treatment. in our study, the incidence of KPC infection was mostly found with bloodstream infections (60%), mostly seen in those with central lines (80%) followed by patients on ventilatory support (66%). Before the outbreak of KPC infection, all the patients (100%) had already been treated with higher antibiotics including Carbapenems. in our study, only nine out of fifteen patients (60%) could be salvaged with treatment and were discharged. Conclusions: Hospital infection Control Committee’s regular screening and the training of healthcare professionals are vital for the control of the outbreak.

Human pluripotent stem cell(h PSC)-derived progenies are immature versions of cells,presenting a potential limitation to the accurate modelling of diseases associated with maturity or ***,it is important to characterise how closely cells used in culture resemble their native *** order to select appropriate time points of retinal pigment epithelium(RPE)cultures that reflect native counterparts,we characterised the transcriptomic profiles of the h PSC-derived RPE cells from 1-and 12-month *** differentiated the human embryonic stem cell line H9 into RPE cells,performed single-cell RNA-sequencing of a total of 16,576 cells to assess themolecular changes of the RPE cells across these two culture time *** results indicate the stability of the RPE transcriptomic signature,with no evidence of an epithelial–mesenchymal transition,and with the maturing populations of the RPE observed with time in *** of Gene Ontology pathways revealed that as the cultures age,RPE cells upregulate expression of genes involved in metal binding and antioxidant *** might reflect an increased ability to handle oxidative stress as cells *** with native human RPE data confirms a maturing transcriptional profile of RPE cells in *** results suggest that long-term in vitro culture of RPE cells allows the modelling of specific phenotypes observed in native mature *** work highlights the transcriptional landscape of h PSC-derived RPE cells as they age in culture,which provides a reference for native and patient samples to be benchmarked against.
