The fate of surplus embryos in the setting of assisted reproductive technology:A scoping review

Objective:To identify the attitudes of infertile couples toward their surplus frozen embryos.Methods:This study was according to PRiSMA-ScR as the guideline for scoping review.Studies that assessed the attitudes of patients or infertile couples who had surplus embryos were included.We conducted systematic searches in English studies from April 2011-April 2021 using 7 databases:PubMed,Science Direct,EBSCO,Scopus,the Cochrane Library,Sage Journals,and Google Scholar.Data were charted based on author,year of publication,country,purpose,data collection,key findings,and research focus/domain.Results:A total of 37 research articles were included in the analysis.Their attitudes encompassed:supporting the donation of the surplus embryos for both research and reproductive purposes,continuing to store the frozen embryos,and disposing of the surplus embryos.Conclusions:Most of the infertile patients support donating their surplus embryos for research and reproductive purposes.

Objective: To evaluate the zoonotic potency of Escherichia coli O157:H7 through arbitrarily primed-PCR(AP-PCR) methods as one of the DNA fingerprinting methods.Methods: A total of 14 isolates consisted of 11 isolates originated from human feces with renal failure symptoms, 2 isolates originated from cattle feces, and 1 control isolate were used in this study. DNA of each isolate was extracted, and their pro files were studied by using AP-PCR method with M13 F and M13 R arbitrary primers.Results: The results founded that all of 14 isolates had similarity range from 54.6% to88.5%. isolates KL-106(3) and KL-55(6) originated from humans showed the degree of similarity with isolates SM-25(1) and SM-7(1) originated from cattle as high as 85% and77%, respectively.Conclusions: The high degree of similarity between isolates originated from cattle and human indicated the high potency of zoonoses. The results also concluded AP-PCR method as a brie fly fingerprinting method in order to trace the epidemiological of E. coli O157:H7.

Challenges to the Conservation of Subak System as World Cultural Heritage in Bali

Subak in Bali is believed to have been existing from earlier than the ninth century. Throughout this long period, Subak has been successfully utilized water resources for irrigation. Subak has been recognized as world cultural heritage by UNESCO since 2012. Currently, however, the sustainability of the system and its existence in the future is a big concern. This research is a literature review that aims to identify the challenges and provide option of solutions to sustain the Subak. The study showed that some operational and maintenance aspects of Subak are alarming, such as some network deterioration, limited availability of maintenance facilities either from government or farmers. Despite this condition, religious tradition of Subak is still carried out, although the processions vary from one Subak to another. There are several challenges in regards to Subak conservation: (1) reduction of the Subak area, resulting in declining income to support a decent life; (2) deterioration of quality and quantity of water resources; (3) the age of farmers is above 40 years with a heavy financial burden; (4) there are no clear external regulations to support Subak conservations. This study suggests several strategic steps in keeping the sustainability of Subak. The strategic steps include:refunctioning government officials; providing government support to reduce cost levied to farmers; facilitating collaborations between farmers and other stakeholders; promoting alternative commodities which have a higher financial value; preventing and repairing damages of upstream areas; controlling pollution; forming a coordination board at a provincial, regency and river basin level; building the Subak information systems; providing supervising mechanism, and encouraging academics and students to participate in an effort to preserve Subak itself.

Background:An increasing number of valid and well-designed trials have demonstrated a positive correlation between visit-to-visit variability(VVV)in systolic blood pressure(SBP)and increased risk of stroke and coronary heart disease among hypertensive patients.Methods:A cross-sectional study was conducted that involved 74 patients who visited the outpatient clinic at the Tabanan iii Primary Health Care Service during April to May 2017.Blood pressure was retrospectively obtained from medical records.VVV was classified as low or high on the basis of the standard deviation of SBP.Antihypertensive medication adherence was expressed as the percentage of days covered,and sodium intake was measured with 24-hour food recall.Bi-variate analysis was performed,followed by multivariate analysis for significant variables.Results:Among the participants,67.6%were female,with a mean(standard deviation[SD])age of 62.70(10.00)years.Blood pressure was measured 4.82±0.78 times during the period,and the mean(SD)SBP was 139.65(10.57)mm Hg.Nonadherence and sodium intake were signifi-cantly higher in the high-VVV group than in the low-VVV group(nonadherence 13.5%vs.37.8%,P=0.033;sodium intake 1278.44±43.02 mg vs.1495.85±45.26 mg,P=0.038).After adjustment for other covariates,the differences remained significant only for nonadherence(model i exp β=3.89[95.0%confidence interval 1.23-12.34,Pii expβ=3.9[95.0%confidence interval 1.12-14.15,Pith sensitivity of 67.6%and specificity of 51.4%.Conclusion:Nonadherence to antihypertensive medication was significantly associated with higher VVV of SBP.Further study is needed to assess whether improving adherence could reduce VVV and improve cardiovascular outcomes.

Maternal inheritance of yellowfin Tuna （Thunnus albacares） in Captivity

Mitochondrial DNA introgression has been suggested to be responsible for the maternal consistent implications. Study on mt-DNA (mitochondrial DNA) variation in the yellowfin tuna (Thunnus albacares) using RFLP (restriction fragment length polymorphisms) has provided the evidence of maternal inheritance of yellowfin tuna in captivity. Eggs were collected in every spawning in 2004-2006 season. The mt-DNA genotypes of broodstock were compared with their eggs and the maternal inheritance of these females was determined from genotypes in the eggs. The result showed that six genotypes of female broodstock were observed in eggs and four of them were found to share a single female"s identity and one type was shared by two females. The same genotype was observed in almost every sampling throughout the year. The female broodstocks spawned almost daily throughout the season.
