IgSF11-mediated phosphorylation of pyruvate kinase m2 regulates osteoclast differentiation and prevents pathological bone loss

Osteoclasts are primary bone-resorbing cells,and receptor-activated NF-k B ligand(RANkL)stimulation is the key driver of osteoclast *** late-stage differentiation,osteoclasts become multinucleated and enlarged(so-called“maturation”),suggesting their need to adapt to changing metabolic demands and a substantial increase in ***,we demonstrate that immunoglobulin superfamily 11(Ig SF11),which is required for osteoclast differentiation through an association with the postsynaptic scaffolding protein PSD-95,regulates osteoclast differentiation by controlling the activity of pyruvate kinase m isoform2(Pkm2).By using a system that directly induces the activation of Ig SF11 in a controlled manner,we identified Pkm2 as a major Ig SF11-induced tyrosine-phosphorylated *** SF11 activates multiple Src family tyrosine kinases(SFks),including c-Src,Fyn,and Hc k,which phosphorylate Pkm2 and thereby inhibit Pkm2 ***,Ig SF11-deficient cells show higher Pkm2activity and defective osteoclast ***,inhibiting Pkm2 activities with the specific inhibitor Shikonin rescues the impaired osteoclast differentiation in Ig SF11-deficient cells,and activating Pkm2 with the specific activator TEPP46 suppresses osteoclast differentiation in wild-type ***,Pkm2 activation further suppresses osteoclastic bone loss without affecting bone formation in *** together,these results show that Ig SF11 controls osteoclast differentiation through Pkm2 activity,which is a metabolic switch necessary for optimal osteoclast maturation.

Exploring the spatial arrangement of patient rooms for minimum nurse travel m hospital nursing units in korea

With in creasing dema nds on medical care services,one of the trends is the mixed patient room arrangement of single/double-bed and multi-bed rooms in a nursing unit on the same *** influences nurse-to-patient assignment and often causes an unbalanced workload and longer travel distances for *** objective of this study was to investigate how floor configuration and room density influence nurse travel in the hospitafs medical surgical urdts in *** study presented a novel approach to measure nurse travel distances in eight existingn ursing *** agent-based simulation was con ducted to model nurses'walking trails,and the distanee of one nurse travel to assigned patient rooms was measured for each *** revisions in the spatial arrangement of patient rooms,locating multibed rooms near the nurse station,symmetric room layout centering the nurse station,and planning both single/double-bed and multi-bed rooms on one side of corridors,nurse travel distanee decreased more than 15%.This study contributed to the knowledge of agent-based simulation as an evaluation framework for spatial *** from application to korea,these results are particularly of interest in countries where private patient rooms are not commonly economically feasible.

Temperature-Responsive Tensile Actuator Based on multi-walled Carbon Nanotube Yarn

many temperature indicators or sensors show color changes for materials used in food and medical ***, they are not helpful for a color-blind person or children who lack judgment. In this paper, we introduce simply fabricated and more useful low-temperature indicator(~30 °C) for devices that actuates using paraffin-infiltrated multiwalled carbon nanotube(mWCNT) coiled yarn. The density difference of mWCNT yarn provides large strain(~330 %)when heat causes the melted polymer to move. Furthermore, the mWCNT yarn decreases the melting point of *** properties allow control of the actuating temperature. In addition, mechanical strength was enhanced by mWCNT than previously reported temperature-responsive actuators based on shape memory polymers. This simply fabricated temperature indicator can be applied in latching devices for medical and biological fields.
