Cortex cinnamomi extract prevents streptozotocin- and cytokine-induced β-cell damage by inhibiting NF-κB

AIM： To clarify the mechanism underlying the antidiabetic activities of cortex cinnamomi extract （CCE）. METHODS： To induce in vivo diabetes, mice were injected with streptozotocin （STZ） via a tail vein （100 mg STZ/kg body weight）. To determine the effects of CCE, mice were administered CCE twice daily for 7 d by oral gavage starting 1 wk before the STZ injection. Blood glucose and plasma insulin concentration were measured as an index of diabetes. Also, to induce cytotoxicity of RINm5F cells, we treated with cytokines （IL-1β （2.0 ng/mL） and IFN-γ （100 U/mL））. Cell viability and nitric oxide production were measured colorimetrically. Inducible nitric oxide synthase （iNOS） mRNA and protein expression were determined by RT-PCR and Western blotting, respectively. The activation of NF-κβ was assayed by using gel mobility shift assays of nuclear extracts. RESULTS： Treatment of mice with STZ resulted in hyperglycemia and hypoinsulinemia, which was further evidenced by immunohistochemical staining of islets. However, the diabetogenic effects of STZ were completely prevented when mice were pretreated with CCE. The inhibitory effect of CCE on STZ-induced hyperglycemia was mediated through the suppression of iNOS expression. In rat insulinoma RINm5F cells,CCE completely protected against interleukin-1β and interferon-y-mediated cytotoxicity. Moreover, RINm5F cells incubated with CCE showed significant reductions in interleukin-1β and interferon-y-induced nitric oxide production and in iNOS mRNA and protein expression, and these findings correlated well with in vivo observations. CONCLUSION： The molecular mechanism by which CCE inhibits iNOS gene expression appears to involve the inhibition of NF-κβ activation, These results reveal the possible therapeutic value of CCE for the prevention of diabetes mellitus progression,

Anterior gradient-2 （AGR2） promotes tumor growth, cell migration and cellular transformation and its enhanced expression is almost completely restricted to malignant tissues, thus making AGR2 an interesting target for the development of immunotherapeutic strategies. We investigated whether the AGR2 molecule comprises human leukocyte antigen （HLA）-A 0201-binding epitopes recognized by human cytotoxic T lymphocytes （CTLs）, which could be targeted in dendritic cell （DC）-based cancer immunotherapy against colorectal cancer （CRC）. We reviewed the sequence of AGR2 for peptides that could potentially bind to HLA-A 0201 with the aid of a computer-based program. Five candidate peptides with different binding scores were synthesized and tested. These peptides were then assessed for their immunogenicity to elicit specific immune responses mediated by CTLs in vitro by means of enzyme-linked immunospot assays and CTL assays. AGR2 was highly expressed in several CRC cell lines, including DK01, DLD1, KM 12C, HCT-8 and HT-29. DCs pulsed with AGR2-P2 （aa 11-19; LLVALSYTL） or AGR2-P4 （aa 127-135; RIMFVDPSL） generated potent CTLs that could lyse T2 cells pulsed with AGR2-P2 or AGR2-P4 and HLA-A0201＋ AGR2-positive CRC cell lines in a strong dose-dependent and HLA-A 0201-restricted manner. In conclusion, these novel epitopes derived from AGR2 protein may be attractive candidates for DC-based immunotherapy for CRC.

Uhwang Chungsim Won Decreases Blood Oxygen Level-Dependent fMRI Signal Response to A Motor Stimulation Task

Objectives： TO evaluate the effect of Uhwang Chungsim Won [UC, Niuhuang Qingxin Pill （牛黄清心丸] on the blood oxygen level-dependent （BOLD） functional magnetic resonance imaging （fMRI） signal response to a motor stimulation task using a hand grasping paradigm. Methods： Ten healthy right-handed male volunteers were recruited. Their BOLD fMRI signal characteristics during hand grasping were determined before and 1 h after consuming UC. Results： With the administration of UC, BOLD responses to the right hand motor task showed a decrease in both hemispheres including the right cerebellar culmen, right and left medial frontal gyrus, right parahippocampal gyrus, right and left sublobar insula, right and left posterior cingulate, left precuneus, and right precentral gyrus. Decreased BOLD responses to the left hand motor task were shown in the right lingual gyrus, right cerebellar culmen, left superior temporal gyrus, right and left middle frontal gyrus, and right precentral gyrus after UC administration. Increased BOLD responses to the left hand motor task were shown in the left and right posterior cingulate, while there was no increased response to the right hand motor task after UC administration. Conclusions： UC administration significantly decreased fMRI BOLD signal responses to hand grasping in most of the brain areas including the primary motor cortex, but it is not clear whether these decreased responses were caused by a decrease in neuronal activities or by an increase in cerebral blood flow due to the cerebral vasodilatory effects of UC. Further studies using positron emission tomography or single photon emission computed tomography are needed to fully elucidate these findings.

Investigations on Structural Safety of Office Room Based on Fire Simulation and Transient Heat Transfer Analysis

This study aims at investigating heat propagations inside the structural members due to fire using fire simulation and transient heat transfer analysis. Toward that goal, fire simulation and transient heat transfer analysis for a 5-story building are carried out sequentially using Fire Dynamics Simulator (FDS) and ABAQUS 6.10-3, respectively. As results from fire simulation, temporal temperature information is obtained depending on various locations of the building, which is used as boundary condition for the structural elements generated in transient heat transfer analysis. Predictions from the transient heat transfer analysis show that the structural members are exposed spatially non-uniform temperatures which can cause significant eccentric deformation and aceleration of structural damages.

Comparative assessment of physicochemical properties of unripe peach(Prunus persica) and Japanese apricot(Prunus mume)

Objective:To investigate the physicochemical properties of unripe peach-Pruuns persica ***(mibaekdo) and Prunus persica *** Hakuho(Nagasawa Hakuho) as an alternative to fowl supplement while Japanese apricot(Prunus mume ***)(Backaha) was used as a control ***:The unripe fruits were analyzed for soluble solid(Brix).titratable ***,total polyphenol content,1,1-diphenyl-2-pierylhydrazyl(DPPH) radical scavenging activity,amygdalin content,free amino acid content,organic acid content,free sugar content,and α-amylase ***:Total polyphenol content of unripe peach ranged between 137.27-151.64 μg/g whereas that of apricot was 160.73 μg/*** radical scavenging activities of Rackaha was the highest189.16%.) followed by mibaekdo(85.03%) and Nagasawa Hakuho(41.30%).The highest amount of oxalic acid 1612.8 mg/100 g) was observed in mibaekdo while that of Nagasawa llakuho and Backaha were(184.6±18.1) and(334.8±16.1) mg/100 g,*** contents of *** Hakuho and Backaha were 486.61,548.60 and 174.28 μg/g,***:The results suggest that the unripe fruit of peach has a significant biochemical potential of using as a food supplement with potential health benefit for human health.

Adolescents’ Estimation of Energy Content of Standard Portion Size of Foods and Its Association with Body Mass Index

The purpose of this study is to identify the adolescents’ knowledge of the energy content of the standard portion size of foods and to investigate the association between their knowledge and energy intake and also body mass index (Bmi). A total of 251 middle school adolescents participated in this study. Participants’ knowledge was assessed based on their estimation of the energy content of the standard portion size of foods. To estimate the energy intake of the subjects, 24-hr recall was used. The percentage of participants who accurately estimated (that is within 20% of the true value) the energy content of the standard portion size was calculated for each of the 32 typical foods. The food for which the most participants revealed the accurate estimation was cooked rice (39.5%). The proportion of students who overestimated the energy contents was highest for vegetables (98%), and oils and sugar (90%). The female students were more likely than males to provide the accurate estimation of energy contents for standard portion size of foods. After adjusting for age and sex, the estimation level ([estimation value/true value] × 100) of the energy content of some foods had a significant positive relationship with Bmi, but had no significant relationship with reported energy intake. From these results, we concluded that the knowledge of energy content of food was poor among middle-school adolescents, with some gender difference, and that their estimation of the calorie contents of foods increased along with their Bmi.

microscopic characteristics of the inferior tarsal muscle and its surroundings in Korean

AIM: To investigate the detailed microscopic anatomic structures of the lower eyelid in Korean cadavers. METHODS: Eight lower eyelids of 4 formalin-fixed Asian cadavers (4 males; age range, 48-69 years; mean age, 60.2 years) were examined. Three perpendicular dissected sections with a 2mm thickness were obtained from each eyelid to investigate anatomic shapes, size and relationship with surrounding structures. One section was obtained from the midline and 2 the other sections were obtained from a 3mm apart from the lateral and medial tarsus margins. RESULTS: The inferior tarsal muscle fibers were not directly attached to the tarsus but were only linked to the tarsus with enclosed fibrous fascia. The inferior tarsal muscles connected loosely with the capsulopalpebral fascia anteriorly and the conjunctiva posteriorly. The inferior tarsal muscle runs horizontally to the tarsus according to the shape of muscle fibers. The capsulopalpebral fascia consisted of an anterior and posterior layer. The anterior layer reached the orbital septum and subcutaneous fat but the posterior layer forwarded into the tarsus. Lockwood’s ligament was separated from the inferior tarsal muscle and capsulopalpebral fascia or fused into the capsulopalpebral ***: This study suggests that the inferior tarsal muscle which runs horizontally and doesn’t insert directly into the tarsus, plays an important role in the movement and localization of the lower eyelid.

Monofilament type of polyaromatic amide(PA)and carbon nanotube(CNT)composite fibers is presented.A concept of a lyotropic liquid crystal(LLC)constructed via a spontaneous self-assembly is introduced to mitigate the extremely low com-patibility between PA and *** approaches provide an effective co-processing route of PA and CNT simultaneously to fabricate the uniform,continuous,and reliable composite fibers through a ***,the addition of a small amount PA into the dope solution of CNT governs the LLC mesophase not only in a spinneret stage but also in a coagulant ***,the developed PA/CNT composite fibers have the high uniaxial orientational order and the close interfacial packing compared to the pure CNT *** PA/CNT composite fibers achieve the outstanding tensile strength,electrical conductivity,and electrochemical response,while maintaining a *** also exhibit the chemical,mechanical,and thermal *** of these advantages can make flexible,sewable,and washable PA/CNT composite fibers ideal nanocomposite materials for use in next-generation information and energy transporting system by replacing conventional metal electrical conductors.

Free Energy Level Correction by Monte Carlo Resampling with Weighted Histogram Analysis Method

Free energy calculations may provide vital information for studying various chemical and biological *** mechanical methods are required to accurately describe interaction energies,but their computations are often too demanding for conformational *** a remedy,level correction schemes that allow calculating high level free energies based on conformations from lower level simulations have been ***,we present a variation of a Monte Carlo(MC)resampling approach in relation to the weighted histogram analysis method(WHAM).We show that our scheme can generate free energy surfaces that can practically converge to the exact one with sufficient sampling,and that it treats cases with insufficient sampling in a more stable manner than the conventional WHAM-based level correction *** can also provide a guide for checking the uncertainty of the levelcorrected surface and a well-defined criterion for deciding the extent of smoothing on the free energy surface for its visual *** demonstrate these aspects by obtaining the free energy maps associated with the alanine dipeptide and proton transfer network of the KillerRed protein in explicit water,and exemplify that the MC resampled WHAM scheme can be a practical tool for producing free energy surfaces of realistic systems.

AIM：To investigate the incidence and general characteristics of sports-related eye injuries in patients visiting the Emergency Department.
