Low temperature-mediated repression and far-red light-mediated induction determine morning FLOWERING LOCUS T expression levels

In order to flower in the appropriate season,plants monitor light and temperature changes and alter downstream pathways that regulate florigen genes such as Arabidopsis(Arabidopsis thaliana)FLOWERING LOCUS T(FT).In Arabidopsis,FT messenger RNA levels peak in the morning and evening under natural long-day conditions(LDs).However,the regulatory mechanisms governing morning FT induction remain poorly understood.The morning FT peak is absent in typical laboratory LDs characterized by high red:far-red light(R:FR)ratios and constant temperatures.Here,we demonstrate that ZEITLUPE(ZTL)interacts with the FT repressors TARGET OF EATs(TOEs),thereby repressing morning FT expression in natural environments.Under LDs with simulated sunlight(R:FR=1.0)and daily temperature cycles,which are natural LD-mimicking environmental conditions,FT transcript levels in the ztl mutant were high specifically in the morning,a pattern that was mirrored in the toe1 toe2 double mutant.Low night-to-morning temperatures increased the inhibitory effect of ZTL on morning FT expression by increasing ZTL protein levels early in the morning.Far-red light counteracted ZTL activity by decreasing its abundance(possibly via phytochrome A(phyA))while increasing GIGANTEA(GI)levels and negatively affecting the formation of the ZTL-GI complex in the morning.Therefore,the phyA-mediated high-irradiance response and GI play pivotal roles in morning FT induction.Our findings suggest that the delicate balance between low temperature-mediated ZTL activity and the far-red light-mediated functions of phyA and GI offers plants flexibility in fine-tuning their flowering time by controlling FT expression in the morning.

Lignocellulosic nanofibers(LCNFs),implying lignin-containing cellulose fibers,maintain the prop-erties of both lignin and cellulose,which are hydrophobic and hydrophilic,respectively.The pres-ence of hydrophobic lignin in LCNFs is expected to be an economical and attractive option that can improve the thermal and mechanical properties of polymers.Thus,this study was conducted to produce lignin-rich LCNFs from sugar-rich waste obtained from rice husks after acidic pretreat-ment.The LCNFs were produced from the lignin-rich solid fractions obtained after pretreatment and enzymatic hydrolysis,which were then incorporated as an additive into a chitosan-based film.The variations in lignin content in the range of approximately 50.6%-66.8%in differently obtained LCNFs gave significantly different optical strengths and mechanical properties.These controllable processes may allow for customized film formation.Additionally,the glucose-rich liquid fractions obtained after pretreatment and enzymatic hydrolysis were used as a substrate for ethanol fermentation to achieve total utilization of rice husk biomass waste.In conclusion,the lignin-rich biomass fraction holds promise as a suitable material for chitosan-LCNF film and has the potential to increase the economic feasibility of the biomaterial industry.

Repeated ventricular bigeminy by trigeminocardiac reflex despite atropine administration during superficial upper lip surgery: A case report

BACKGROUND The trigeminocardiac reflex(TCR)is usually caused by an increased parasympathetic tone when pressure or traction is applied to the surrounding tissue of the trigeminal nerve.However,the inexperienced anesthesiologists may have challenges on the management of TCR patients.CASE SUMMARY This is the case of an 18-year-old woman diagnosed with hemangioma of the upper lip.During the operation,about 1 h after surgery started,a constant 1:1 premature ventricular complex was detected,and blood pressure was decreased when approaching the deeper part with more strong traction for exposure of the part.Although the management of arrhythmias,such as lidocaine and atropine,was injected,arrhythmia induced by surgical stimulation could not be eliminated completely.As the traction repeated,bradycardia was also repeated,despite injecting additional atropine.Therefore,the anesthesiologist and the surgeon decided to perform the operation only to the extent that the vascular tissue was selectively removed only at the site without the reflex.CONCLUSION With TCR,anesthesiologists should perform appropriate monitoring.In addition to proper drug administration,surgeons should be consulted to cope with stopping the surgery and setting the scope of the surgery even if the site is superficial.

Recurrent intramuscular lipoma at extensor pollicis brevis:A case report

BACKGROUND This report describes and discusses recurrent intramuscular lipoma(IML)of the extensor pollicis brevis(EPB).An IML usually occurs in a large muscle of the limb or torso.Recurrence of IML is rare.Recurrent IMLs,especially those with unclear boundaries,necessitate complete excision.Several cases of IML in the hand have been reported.However,recurrent IML appearing along the muscle and tendon of EPB on wrist and forearm has not been reported yet.CASE SUMMARY In this report,the authors describe clinical and histopathological features of recurrent IML at EPB.A 42-year-old Asian woman presented with a slowgrowing lump in her right forearm and wrist area six months ago.The patient had a history of surgery for a lipoma of the right forearm one year ago with a scar of 6cm on the right forearm.magnetic resonance imaging confirmed that the lipomatous mass,which had attenuation similar to subcutaneous fat,had invaded the muscle layer of EPB.Excision and biopsy were performed under general anesthesia.On histological examination,it was identified as an IML showing mature adipocytes and skeletal muscle fibers.Therefore,surgery was terminated without further resection.No recurrence occurred during a follow-up of five years after surgery.CONCLUSION Recurrent IML in the wrist must be examined to differentiate it from sarcoma.Damage to surrounding tissues should be minimized during excision.

Improving toughness of medium-Mn steels after friction stir welding through grain morphology tuning

This work demonstrated the viability of friction stir welding for the welding of medium-Mn steels when used as cryogenic vessel materials for liquefied gas storage.We used an intercritically annealed Fe-7 Mn-0.2 C-3 Al(wt.%)steel with a dual-phase(α'martensite andγ_(R) retained austenite)nanolaminate structure as a base material and systematically compared its microstructure and impact toughness after friction stir and tungsten inert gas welding.The friction stir welded specimen exhibited a large amount ofγ_(R) phase owing to a relatively low temperature during welding,whereas the tungsten inert gas welded specimen comprised only theα'phase.Furthermore,the friction stir welded steel exhibited a tuned morphology of nanoscale globular microstructure at the weld zone and did not exhibit any prior austenite grain boundary due to active recrystallization caused by deformation during welding.The preserved fraction ofγ_(R) and morphological tuning in the weldment improved the impact toughness of the friction stir welded steel at low temperatures.In the steel processed by tungsten inert gas welding,the notch crack propagated rapidly along the prior austenite grain boundaries-weakened by Mn and P segregations-resulting in poor impact toughness.However,the friction stir welded steel exhibited a higher resistance against notch crack propagation due to the slow crack propagation along the ultrafine ferrite/ferrite(α/α)interfaces,damage tolerance by the active transformation-induced plasticity from the large amount ofγR,and enhanced boundary cohesion by suppressed Mn and P segregations.

GC-MS analysis of volatile compounds of Perilla frutescens Britton var. Japonica accessions: morphological and seasonal variability

Objective: To investigate the composition of volatile compounds in the different accessions of Perilla frutescens(P. frutescens) collected from various habitats of China and Japan. Methods: In the present study, the essential oil from the leaves of P. frutescens cultivars from China and Japan was extracted by hydro-distillation and the chemical composition and concentration of the volatile components present in the oils were determined by gas chromatography–mass spectrometry(GC–MS) analysis. Results: Among the volatile components, the major proportion was of perilla ketone, which was followed by elemicin and beta-caryophyllene in the Chinese Perilla cultivars. The main component in the oil extracted from the Japanese accessions was myristicin, which was followed by perilla ketone and beta-caryophyllene. We could distinguish seven chemotypes, namely the perilla ketone(PK) type, perilla ketone, myristicin(PM) type, perilla ketone, unknown(PU) type, perilla ketone, beta-caryophyllene, myristicine(PB) type, perilla ketone, myristicin, unknown(PMU) type, perilla ketone, elemicine, myristicin, beta-caryophyllene(PEMB) type, and the perilla ketone, limonene, betacryophyllene, myristicin(L) type. Most of the accessions possessed higher essential oil content before the flowering time than at the flowering stage. The average plant height, leaf length, leaf width of the Chinese accessions was higher than those of the Japanese accessions. Conclusion:The results revealed that the harvest time and geographical origin caused polymorphisms in the essential oil composition and morphological traits in the Perilla accessions originating from China and Japan. Therefore, these chemotypes with desirable characters might be useful for industrial exploitation and for determining the harvest time.

BACKGROUND Primary non-Hodgkin’s lymphoma of the frontal sinus is extremely rare.In addition,Epstein-Barr virus(EBV)has been reported to play a role in the development of human immunodeficiency virus(HIV)-related malignant lymphomas.To the best of our knowledge,there is no report for the HIVassociated,EBV-positive primary diffuse large B-cell lymphoma(DLBCL)in the frontal sinus.CASE SUMMARY We present a unique case of HIV-associated,EBV-positive DLBCL in the frontal sinus in a 46-year-old man.Computed tomography of paranasal sinuses revealed dense opacification of the right frontal sinus with combined soft tissue swelling.Based on the clinical and radiological findings,the initial diagnosis was complicated frontal sinusitis,presenting Pott’s puffy tumor.Unexpectedly,HIV testing was positive on preoperative laboratory test,and the frontal sinus lesion was confirmed as EBV-positive DLBCL on biopsy.CONCLUSION Through this article,we suggest that EBV-positive DLBCL should be considered as possible diagnosis for patients with nonspecific space-occupying lesion of the paranasal sinuses.We also highlight an importance of clinical suspicion in diagnosing HIV infection because HIV serology is not routinely tested in patients with paranasal sinus problem.

Lemierre's syndrome caused by Klebsiella pneumoniae:A case report

BACKGROUND Lemierre's syndrome is a disease that causes anaerobic sepsis,internal jugular vein thrombosis,and septic embolism in the lungs and other organs after acute oropharyngeal infection.It was named after André-Alfred Lemierre in 1936.CASE SUMMARY Here,we have reported a case of Lemierre’s syndrome in a 56-year-old female patient who presented with a sore throat.The patient had septic shock,had not voided,and had severe hyperglycemia at the time of her visit.Imaging tests revealed bilateral pneumonia,pleural effusion,pulmonary embolism,and renal vein thrombosis.The patient was admitted to the intensive care unit and placed on mechanical ventilation due to acute respiratory distress syndrome.Continuous renal replacement therapy was administered to treat renal failure with anuria.Klebsiella pneumoniae was cultured from blood and sputum samples.After reviewing various results,the patient was ultimately diagnosed with Lemierre’s syndrome.The patient was treated with appropriate antibiotics and thrombolytic agents.She was discharged from the hospital after recovery.CONCLUSION Lemierre’s syndrome is associated with a high mortality rate.Therefore,clinicians should be familiar with the signs and symptoms of this disease as well as the preemptive examinations,procedures,and treatments.

Orostachys japonicus ethyl acetate fraction suppresses MRSA biofilm formation

Objective:To investigate the effect of Orostachys(O.)japonicus,a perennial herbaceous plant of the Family Crassulaceae,on biofilm formed by methicillin-resistant Staphylococcus aureus(MRSA).Methods:Powdered O.japonicus was extracted by 95%methanol,concentrated,and then,systematically fractionated with n-hexane,dichloromethane(DCM),ethyl acetate(EtOAc),n-butanol,and H2 O according to polarity.Among them,the flavonoid-rich EtOAc fraction demonstrated the highest antibacterial activity and was used in this study.Using the biofilm inhibition assay,cell-surface attachment assay,confocal laser scanning microscopy,latex agglutination assay,and real time qRT-PCR,we examined whether the EtOAc fraction inhibited the formation of MRSA biofilm.Results:The EtOAc fraction exhibited distinct activity against biofilm formation and cell-surface attachment of MRSA up to 1 mg/m L through down-regulating the expression of mec A gene and the production and agglutination of penicillin-binding protein 2 a as solidly observed in biofilm inhibition assay,cell-suface attachment assay,confocal laser scanning microscopy,latex agglutination assay,and real time qRT-PCR analysis.Conclusions:These results suggest that O.japonicus could be utilized as a potential resource for the development of new antibiofilm formation of MRSA and antibacterial agents in the future.

mitochondrial-targeting moieties to anticancer drugs, antioxidants and sensor molecules. Among them, the most widely applied mitochondrial targeting moiety is triphenylphosphonium(TPP), which is a delocalized cationic lipid that readily accumulates and penetrates through the mitochondrial membrane due to the highly negative mitochondrial membrane potential. Other moieties, including short peptides,dequalinium, guanidine, rhodamine, and F16, are also known to be promising mitochondrial targeting agents. Direct conjugation of mitochondrial targeting moieties to anticancer drugs, antioxidants and sensors results in increased cytotoxicity, anti-oxidizing activity and sensing activity, respectively,compared with their non-targeting counterparts, especially in drug-resistant cells. Although many mitochondria-targeted anticancer drug conjugates have been investigated in vitro and in vivo, further clinical studies are still needed. On the other hand, several mitochondria-targeting antioxidants have been analyzed in clinical phases I, II and III trials, and one conjugate has been approved for treating eye disease in Russia. There are numerous ongoing studies of mitochondria-targeted sensors.
