Dendritic cell-mimicking scaffolds for ex vivo T cell expansion

We propose an ex vivo T cell expansion system that mimics natural antigen-presenting cells(APCs)for adoptive cell therapy(ACT).Microfiber scaffolds coated with dendritic cell(DC)membrane replicate physicochemical properties of dendritic cells specific for T cell activation such as rapid recognition by T cells,long duration of T cell tethering,and DC-specific co-stimulatory *** DC membrane-coated scaffold is first surface-immobilized with T cell stimulatory ligands,anti-CD3(αCD3)and anti-CD28(αCD28)antibodies,followed by adsorption of releasable interleukin-2(IL-2).The scaffolds present both surface and soluble cues to T cells ex vivo in the same way that these cues are presented by natural APCs in *** demonstrate that the DC-mimicking scaffold promotes greater polyclonal expansion of primary human T cells as compared toαCD3/αCD28-func-tionalized *** importantly,major histocompatibility complex molecules derived from the DC membrane of the scaffold allow antigen-specific T cell expansion with target cell-specific killing *** addition,most of the expanded T cells(~97%)can be harvested from the scaffold by density gradient ***,the DC-mimicking scaffold offers a scalable,modular,and customizable platform for rapid expansion of highly functional T cells for ACT.

A dynamic infiltration technique to synthesize nanolayered cathodes for high performance and robust solid oxide fuel cells

Solution infiltration is a popular technique for the surface modification of solid oxide fuel cell(SOFC)***,the synthesis of nanostructured SOFC cathodes by infiltration is a tedious process that often requires several infiltration and high temperature(≥500℃)calcination ***,fabricating large-area nanostructured cathodes via infiltration still requires serious ***,we propose a facile and scalable urea assisted ultrasonic spray infiltration technique for nanofabrication of SOFC *** is demonstrated that by using urea as a precipitating agent,the calcination after each infiltration cycle can be omitted and the next infiltration can be performed just after a drying step(≤100℃).Finally,the precipitates can be converted into a desired catalyst phase in single calcination thus,a nanostructured cathode can be fabricated in a much faster *** is also shown that the low calcination temperature of the cathode(≤900℃)can produce highly durable SOFC performance even without employing a Ce_(0.9)Gd_(0.1)O_(2)(GDC)diffusion barrier layer which provides the ease of SOFC *** coupling with an ultrasonic spray technique,the urea assisted infiltration can be scaled up for any desired cathode ***_(0.6)Sr_(0.4)Co_(0.2)Fe_(0.8)O_(3) nanolayered cathode was fabricated and it was characterized by scanning electron microscope(SEM),X-ray diffraction(XRD),and transmission electron microscopy(TEM)*** showed the formation of a nanolayer cathode just after 5 cycles of the urea assisted infiltration while the XRD and TEM confirmed the phase and stoichiometric uniformity of the 100 nm cathode *** effectiveness of the newly developed technique was further verified by the stable operation of a GDC buffer layer free SOFC having an active cathode area of 25 cm^(2) during a 1200 h durability *** research outcomes propose urea assisted ultrasonic spray infiltration as a facile,scalable,and commercially viable method

A critical challenge hindering the practical application of lithium–oxygen batteries(LOBs)is the inevitable problems associated with liquid electrolytes,such as evaporation and safety *** study addresses these problems by proposing a modified polyrotaxane(mPR)-based solid polymer electrolyte(SPE)design that simultaneously mitigates solvent-related problems and improves ***-SPE exhibits high ion conductivity(2.8×10^(−3)S cm^(−1)at 25℃)through aligned ion conduction pathways and provides electrode protection ability through hydrophobic chain *** this mPR-SPE into solid-state LOBs resulted in stable potentials over 300 *** situ Raman spectroscopy reveals the presence of an LiO_(2)intermediate alongside Li_(2)O_(2)during oxygen *** situ X-ray diffraction confirm the ability of the SPE to hinder the permeation of oxygen and moisture,as demonstrated by the air permeability *** present study suggests that maintaining a low residual solvent while achieving high ionic conductivity is crucial for restricting the sub-reactions of solid-state LOBs.

Curative resection is undoubtedly the most important prognostic factor in perihilar cholangiocarcinoma(1).The current consensus of surgical resection of perihilar cholangiocarcinoma consists of hemihepatectomy,including caudate lobectomy and aggressive lymphadenectomy,depending on the extent of bile duct *** decisions on whether to resect the right or left liver and whether to perform an extended hepatectomy or trisectionectomy are all aimed at obtaining negative radial and longitudinal margins(2,3).

BACKGROUND Clostridioides difficile(***)colitis is one of the most common infections in hospitalized patients,characterized by fever and *** usually improves after appropriate antibiotic treatment;if not,comorbidities should be ***(CMV)colitis is a possible co-existing diagnosis in patients with *** infection with poor treatment ***,compared with immunocompromised patients,CMV colitis in immunocompetent patients is not well *** SUMMARY We present an unusual case of co-existing CMV colitis in an immunocompetent patient with *** *** 80-year-old female patient was referred to the infectious disease department due to diarrhea,abdominal discomfort,and fever for 1 wk during her hospitalization for *** toxin B polymerase chain reaction on stool samples was *** *** infection was diagnosed,oral vancomycin treatment was *** symptoms including diarrhea,fever and abdominal discomfort improved for ten ***,the symptoms worsened again with bloody diarrhea and ***,a sigmoidoscopy was performed for evaluation,showing a longitudinal ulcer on the sigmoid *** biopsy confirmed CMV colitis,and the clinical symptoms improved after using *** Co-existing CMV colitis should be considered in patients with aggravated *** infection on appropriate treatment,even in immunocompetent hosts.

Effect of ZnO Electrodeposited on Carbon Film and Decorated with Metal Nanoparticles for Solar Hydrogen Production

In this study, we prepared horn-like ZnO structures on carbon films（ZnO/CF） by electrodeposition and decorated the ZnO horns with different metals（Ag, Au, and Pt） via photodeposition（M-ZnO/CF）. Using M-ZnO/CF as photocatalysts, we examined ways to enhance solar hydrogen production from various points of view, such as modifying the intrinsic physical properties and thermodynamics of the materials, and varying the chemical environment during M-ZnO/CF fabrication. In particular, we focused on the effects of the carbon film and metals in M-ZnO/CF hybrid photocatalysts on solar hydrogen production. The type of metal nanoparticles is an important factor in solar hydrogen production because the deposition rate and electrical conductivity of each metal affect the proton-water reduction ability.

Noninvasive Parameters and hepatic fibrosis scores in children with nonalcoholic fatty liver disease

AIM:To evaluate the noninvasive parameters and hepatic fibrosis scores in obese children with nonalcoholic fatty liver disease(NAFLD).METHODS:A total of 77 children diagnosed with NAFLD via liver biopsy were included and divided into 2 subgroups according to the histopathologic staging of hepatic fibrosis:mild(stage 0-1)vs significant fibrosis(stage 2-4).Clinical and laboratory parameters were evaluated in each *** aspartate aminotransferase(AST)/alanine aminotransferase(ALT)ratio,AST/platelet ratio index(APRI),PGA index,Forns index,FIB-4,NAFLD fibrosis score,and pediatric NAFLD fibrosis index(PNFI)were ***:No clinical or biochemical parameter exhibited a significant difference between patients with mild and significant *** noninvasive hepatic fibrosis scores,only APRI and FIB4 revealed a significant difference between patients with mild and significant fibrosis(APRI:0.67±0.54 vs 0.78±0.38,P=0.032 and FIB4:0.24±0.12 vs 0.31±0.21,P=0.010).The area under the receiving operating characteristic curve of FIB4 was 0.81,followed by Forns index(0.73),APRI(0.70),NAFLD fibrosis score(0.58),AST/ALT ratio(0.53),PGA score(0.45),and PNFI(0.41).CONCLUSION:APRI and FIB4 might be useful noninvasive hepatic fibrosis scores for predicting hepatic fibrosis in children with NAFLD.

Effect of citrulline on post-exercise rating of perceived exertion,muscle soreness,and blood lactate levels:A systematic review and meta-analysis

Background:Citrulline is one of the non-essential amino acids that is thought to improve exercise performance and reduce post-exercise muscle *** conducted a systematic review and meta-analysis to determine the effect of citrulline supplements on the post-exercise rating of perceived exertion(RPE),muscle soreness,and blood lactate ***:A random effects model was used to calculate the effect sizes due to the high variability in the study design and study populations of the articles included.A systematic search of PubMed,Web of Science,and *** was *** for study inclusion was limited to studies that were randomized controlled trials involving healthy individuals and that investigated the acute effect of citrulline supplements on RPE,muscle soreness,and blood lactate *** supplementation time frame was limited to 2 h before *** types and number of participants,types of exercise tests performed,supplementation protocols for L-citrulline or citrulline malate,and primary(RPE and muscle soreness)and secondary(blood lactate level)study outcomes were extracted from the identified ***:The analysis included 13 eligible articles including a total of 206 *** most frequent dosage used in the studies was 8 g of citrulline *** supplementation significantly reduced RPE(n=7,p=0.03)and muscle soreness 24-h and 48-h after post-exercise(n=7,p=0.04;n=6,p=0.25,respectively).However,citrulline supplementation did not significantly reduce muscle soreness 72-h post-exercise(n=4,p=0.62)or lower blood lactate levels(n=8,p=0.17).Conclusion:Citrulline supplements significantly reduced post-exercise RPE and muscle soreness without affecting blood lactate levels.
