Glypican-3 is a protein encoded by the Glypican-3 gene located on human X chromosome (Xq26), composed of two subunits, a 40 kDa N-terminal subunit, and a 30 kDa C-terminal subunit. Glypican-3 is a currently potential target molecule for liver cancer treatments because of its over-expression and growth effects on hepatocellular carcinoma (HCC). this study examined the expression and purification of a C-terminal subunit of Glypican-3 protein (GPC3-Csub) due to its application in both diagnosis and therapy for hepatocellular carcinoma. The gene encoding for GPC3-Csub was successfully cloned into plasmid pET28a fused with an affinity tag composed of six consecutive histidine residues (His-tag). Recombinant protein GPC3-Csub was expressed in Escherichia coli BL21 (DE3) in the condition of adding 3% ethanol with IPTG induction. GPC3-Csub was extracted using repeated freeze-thaw cycles with lysozyme, and inclusion bodies were solubilized by 8M Urea, SDS 10% in pH 12. His-tag fused GPC3-Csub proteins allowed it to be purified by affinity chromatography method using the Nickel-nitrilotriacetic acid (Ni-NTA) column. High expression of GPC3-Csub was confirmed by Coomassie staining and western-blot. GPC3-Csub could be isolated with a Ni-NTA column and have a purity of about 90%.

The use of the thermal power plant ashes including fly ash(FA) and bottom ash(BA) for producing unfired building bricks(UBB) using sodium hydroxide(NaOH) solution as an alkaline activator was investigated. A low applied forming pressure of 0.5 MPa and various NaOH concentrations of 5, 8, 10, and 12 M were used for the preparation of brick samples with different solution-to-binder(S/B) ratios of 0.35 and 0.40. The bricks were subjected to various test programs with reflecting the effect of both NaOH concentrations and S/B ratios on the brick’s properties. The compressive strength, unit weight, ultrasonic pulse velocity, and thermal conductivity of bricks increased with increasing NaOH concentration, whereas the contrary trend was found with increasing S/B ratio. Also, the water absorption of bricks was observed to reduce with increasing NaOH concentration and decreasing S/B ratio. As the results, the combined utilization of both low forming pressure and coal power plant ashes can produce the UBBs with low unit weight, low heat conductivity, and acceptable strength and water absorption rate as stipulated by TCVN 6477-2016. Furthermore, the outcomes of chemical analysis and microstructure observation also demonstrate that a high concentration of the Na OH promoted the geopolymerization process. Notably, the use of NaOH solution of either 10 M or above is recommended for the production of UBBs, which are classified as grade M5.0 or higher.

Objective:To assess the knowledge and attitude toward coronavirus disease-2019(COVID-19)among healthcare workers at District 2 Hospital in Ho Chi Minh City(HCMC).Methods:A cross-sectional study was performed between January 2020 and February 2020 at District 2 Hospital.A systematic random sampling strategy was carried out and the data was collected through a self-administered questionnaire of the knowledge and attitude of healthcare workers regarding *** analysis was reported to describe the demographic,mean knowledge and attitude score of healthcare *** statistics including t-test,ANOVA and Spearman’s correlation were used to evaluate the relationship between study ***:A total of 327 eligible healthcare workers had a mean score of knowledge and attitude of 8.17±1.3(range 4-10)and 1.86±0.43(range 1-5),*** showed good knowledge and a positive ***,approximately two thirds of the participants knew the mode of transmission,the isolation period and treatment(67.0%,65.8%,and 58.4%,respectively),and 82.3%and 79.8%,respectively,held positive attitude regarding the risk of personal and family members getting *** was a negative correlation between knowledge scores and attitude scores(r=-0.21,P<0.001).Additionally,healthcare workerspredominately used social media to inform themselves about COVID-19(91.1%).Conclusions:The majority of healthcare workers had good knowledge and positive attitude toward ***,the level of some knowledge and attitude lower than that expected for their position level towards the *** education interventions and campaigns are required for healthcare workers.

this paper provides a comprehensive overview of a phase-field model of fracture in solid mechanics setting. We start reviewing the potential energy governing the whole process of fracture including crack initiation, branching or merging. Then, a discretization of system of equation is derived, in which the key aspect is that for the correctness of fracture phenomena, a split into tensile and compressive terms of the strain energy is performed, which allows crack to occur in tension, not in compression. For numerical analysis, standard finite element shape functions are used for both primary fields including displacements and phase field. A staggered scheme which solves the two fields of the problem separately is utilized for solution step and illustrated with a segment of Python code.

Taxus chinensis and T. wallichiana in have been threatened in their distribution areas in recent decades because of their over-exploitation and reduction and destruction of native habitats. Determining the genetic diversity in populations of the two species will provide guidelines for their protection and preservation. Two hundred and fifteen trees from six populations of T. chinensis and150 sampled trees of T. wallichiana were sampled. Six microsatellite primer pairs selected from 16 primer pairs were used to investigate genetic variation at the population and species levels. Five yielded polymorphic alleles, and among the 13 putative alleles amplified, 11 were polymorphic（accounting for 76.33 %）.Shannon＇s information index（I） and percentage of polymorphic bands（PPB）（I = 0.202 and PPB = 67.22 % for T. chinensis; I = 0.217 and PPB = 65.03 % for T. wallichiana）. Both species had low levels of genetic diversity（mean Ho= 0.107, He= 0.121 for T. chinensis; Ho= 0.095, He= 0.109 for T. wallichiana）. Genetic differentiation among populations was higher（FST= 0.189） for T. chinensis and lower（0.156） for ***, indicating limited gene flow（Nm） among populations for T. chinensis（0.68） and T. wallichiana（0.65）.Variation among individuals of T. chinensis was 63.59 and73.12 % for T. wallichiana. Thus, the threatened status of the two conifers is related to a lack of genetic diversity. All populations are isolated in small forest remnants. An ex situ conservation site should be established with a new population for these species that comprises all the genetic groups for the best chance to improve their fitness under environmental stresses.

Application Prospects for the Innovation of Defined Fungal Starter in Rice Wine Fermentation

The feasibility of pilot-scale manufacture of defined fungal starter and its application in rice wine production from different local starchy materials were investigated. Starter consisting of Arnylomyces rouxii and Saccharomyces cerevisiae gave high performance in winemaking when prepared in conditions of rice flour 80% and cassava flour 20% with 4 incubation days. The starter level at 20% was favourably employed for manufacture with the initial amount of I0 kg mixed-flours, Dry starter granules which were vacuum packed could adapt ambient temperature （approx. 28-32℃） during 8 months of storage. The defined starter performed as superior inoculum for winemaking from different agricultural starchy resources. The undesirable bacteria were found at approx. 2 Log CFU/g of dry starter. By morphology, biochemical and physiological growth and the genetic partial 16S analyses, three bacterial isolates were characterized as Bacillus subtilis/amvloliquefaciens which may contaminate tbod but not cause food poisoning and not considered as a human pathogen.
