Background Ciliopath.es are a group of diseases associated with.abnormal structure or function of primary cilia. Ciliopath.es include polycystic kidney disease （PKD）, a path.logy associated with.vascular h.pertension. We previously sh.wed th.t cilia length.regulates cilia function, and cilia function is required for nitric oxide （NO） biosynth.sis in endoth.lial cells. Because patients with.PKD sh.w abnormal sensory cilia function, th. aim of our current study was to search.for a targeted th.rapy focused on primary cilia, wh.ch.we refer to as ＇cilio- th.rapy＇. Meth.ds and Results In th. present studies, our in vitro analyses refined fenoldopam as an equipotent and more specific dopa- minergic agonist to regulate cilia length.and function. Our in vivo studies indicated th.t fenoldopam increased cilia length.and serum NO th.reby reducing blood pressure in a PKD mouse model. Our crossover, multicenter, double-blind and placebo-controlled clinical study furth.r indicated th.t cilia-targeting th.rapy sh.wed an overall reduction in mean arterial pressure in PKD patients. Conclusions Overall, our studies provide th. first evidence of cilioth.rapy as an innovative intervention in patients with.abnormal primary cilia.

AIM: To investigate th. genetic constitution of an escape mutant h.N1 strain and to screen th. presence of possible amino acid signatures th.t could differentiate it from oth.r Egyptian h.N1 ***: Ph.logenetic, evolutionary patterns and amino acid signatures of th. genes of an escape mutant h.N1 influenza A virus isolated in Egypt on 2009 were analyzed using direct sequencing and multisequence ***: All th. genes of th. escape mutant h.N1 strain sh.wed a genetic pattern potentially related to Eurasian lineages. Evolution of ph.logenetic trees of different viral genes revealed th. absence of reassortment in th. escape mutant strain wh.le confirming close relatedness to oth.r h.N1 Egyptian strains from h.man and avian species. A variety of amino acid substitutions were recorded in different proteins compared to th. available Egyptian h.N1 strains. Th. strain displayed amino acid substitutions in different viral alleles similar to oth.r Egyptian h.N1 strains with.ut sh.wing amino acid signatures th.t could differentiate th. escape mutant from oth.r Egyptian ***: Th. genetic ch.racteristics of avian h.N1 in Egypt revealed evidence of a h.gh.possibility of inter-species transmission. No amino acid signatures were found to differentiate th. escape mutant h.N1 strain from oth.r Egyptian h.N1 strains.

Current efforts in sorgh.m breeding programs are exploiting genotyping-by-sequencing (GBS) data to provide full-genome scans for desired traits. Th. aim of th.s study was to utilize GBS approach.for th. identification of genomic regions associated with.stress response in sorgh.m (Sorgh.m bicolor L. Monech. accessions. DNA samples of twenty sorgh.m accessions, h.ving different response to drough., were used to prepare GBS libraries for sequencing. SNPs were called using th. TASSELGBS pipeline and th. tags th.t present at least 10 times in th. dataset were considered and aligned to th. reference genome of Sorgh.m bicolor. Th. identified SNPs were all compared with.th. publish.d sorgh.m transcript related to stress response gene activity. Overall;94.40% tags were aligned and 69,736 putative SNPs positions were identified. Blast search.revealed h.mology to annotated h.at and drough.–tolerance associated genes wh.ch.code for ATPases, Peroxidase, h.droph.bic protein LTI6A, Aquaporin SIP2-1, Aconitate h.dratase and ph.sph.tidylinositol-4-ph.sph.te-5-kinase. Th. ph.logeny of th. 20 accessions was constructed using th. generated SNPs data. Ph.logenetic analysis data sh.wed th.t th. ph.notypically tolerant line (El9)?makes a separate cluster and th. same for th. accessions h.D8653 and h.D5612 near to th. cluster th.t includes most accessions with.known post-flowering drough. tolerance (h.D7410, h.S10033, h.D8552, GESh.ISh.and h.D8849). Post-flowering drough. sensitive accessions (Tabat, Wadah.ed, h.D6468 and h.D6478) formed a separate cluster wh.le th. sensitive accession h.D9959 and th. tolerant accessions h.D8511 and h.D9566 were distributed between th. two clusters. Th.s, cluster analysis confirmed th. variation among accessions in post-flowering drough. tolerance. With.furth.r validation, th.se markers may be used for marker assisted selection for breeding new sorgh.m genotypes with.stress adaptation.

Understanding h.w different livestock species and breeds respond to consumption of brackish.water could improve usage of th.s ***,Angora,Boer,and Spanish.goat doelings and Dorper,Katah.in,and *** ewe lambs(6 animals per animal type[AT];initial age=296±2.1 days)consuming water with.varying concentrations of minerals of a natural brackish.water source(BR)and sodium ch.oride(NaCl;SL)were used to determine effects on water and feed intake,nutrient digestion,h.at energy,meth.ne emission,ruminal fluid conditions,and blood constituent *** were 6 simultaneous 6(water treatments[WT])×6(AT)Latin squares with.3-wk *** WT were fresh.FR),BR alone(100-BR),a similar total dissolved solids(TDS)concentration as 100-BR via NaCl addition to FR(100-SL),BR with.concentrations of all minerals increased by approximately 50%(150-BR),a similar TDS level as 150-BR by NaCl addition to FR(150-SL),and a similar 150 TDS level ach.eved by addition of a 1:1 mixture of BR minerals and NaCl to 100-BR(150-BR/SL).Concentrations(mg/kg)in BR were 4928 TDS,85.9 bicarbonate,224.9 calcium,1175 ch.oride,60.5 magnesium,4.59 potassium,1387sodium,1962 sulfate,and 8.3 boron,and TDS in oth.r WT were 209,5684,7508,8309,and 7319 mg/kg for FR,100-SL,150-BR,150-SL,and 150-BR/SL,*** were very few significant effects of WT or AT×WT interactions,alth.ugh.AT h.d numerous *** intake was affected by AT(P=0.02)and WT(P=0.04),with.greater water intake for 150-SL th.n for FR,100-BR,100-SL,and *** matter intake among AT was lowest(Ph.at energy differed among AT(Ph.ne emission were similar among *** aldosterone concentration was h.gh.r(Ph.n for oth.r *** conclusion,all AT seemed resilient to th.se WT regardless of mineral source and concentrations,with.TDS less th.n 8300 mg/kg,wh.ch.did not influence nutrient u
