An Approach to Quantify Endomembrane Dynamics in Pollen Utilizing Bioactive Chemicals

Tip growth of pollen tubes and root hairs occurs via rapid polar growth. These rapidly elongating cells require tip-focused endomembrane trafficking for the deposition and recycling of proteins, membranes, and cell wall materials. Most of the image-based data published to date are subjective and non-quantified. Quantitative and com- parative descriptors of these highly dynamic processes have been a major challenge, but are highly desirable for genetic and chemical genomics approaches to dissect this biological network. To address this problem, we screened for small molecules that perturbed the localization of a marker for the Golgi Ras-like monomeric G-protein RAB2：GFP expressed in transgenic tobacco pollen. Semi-automated high-throughput imaging and image analysis resulted in the identifica- tion of novel compounds that altered pollen tube development and endomembrane trafficking. Six compounds that caused mislocalization and varying degrees of altered movement of RAB2：GFP-labeled endomembrane bodies were used to generate a training set of image data from which to quantify vesicle dynamics. The area, velocity, straightness, and intensity of each body were quantified using semi-automated image analysis tools revealing quantitative differences in the phenotype caused by each compound. A score was then given to each compound enabling quantitative comparisons between compounds. Our results demonstrate that image analysis can be used to quantitatively evaluate dynamic sub- cellular endomembrane phenotypes induced by bioactive chemicals, mutations, or other perturbing agents as part of a strategy to quantitatively dissect the endomembrane network.

Calibration method to improve the accuracy of THz imaging and spectroscopy in reflection geometry

We introduce a novel method to accurately extract the optical parameters in terahertz reflection imaging. Our method builds on standard self-referencing methods using the reflected signal from the bottom of the imaging window material to further compensate for time-dependent system fluctuations and position-dependent variation in the window thickness. Our proposed method not only improves the accuracy, but also simplifies the imaging procedure and reduces measurement times.

Light-mediated therapeutics have attracted considerable attention as a method for the treatment of ophthalmologic diseases,such as age-related macular degeneration,because of their non-invasiveness and the effectiveness to ameliorate the oxidative stress of retinal ***,the current phototherapeutic devices are opaque,bulky,and tethered forms,so they are not feasible for use in continuous treatment during the patient’s daily ***,we report wireless,wearable phototherapeutic devices with red light-emitting diodes for continuous *** light-emitting diodes were formed to be conformal to three-dimensional surfaces of glasses and contact ***,fabricated light-emitting diodes had either transparency or a miniaturized size so that the user’s view is not ***,these devices were operated wirelessly with control of the light *** addition,in-vitro and in-vivo tests using human retinal epithelial cells and a live rabbit demonstrated the effectiveness and reliable operation as phototherapeutic devices.

Morphological and biochemical responses of Oryza sativa L.(cultivar MR219) to ion beam irradiation

Objective: Heavy ion beam, which has emerged as a new mutagen in the mutation breeding of crops and ornamental plants, is expected to result in the induction of novel mutations. This study investigates the morphological and biochemical responses of Oryza sativa toward different doses of carbon ion beam irradiation. Methods: In this study, the dry seeds of O. sativa were irradiated at 0, 20, 40, 60, 80, 100, and 120 Gy, followed by in-vitro germination under controlled conditions. Morphological and biochemical studies were conducted to investigate the morphological and physiological responses of O. sativa towards ion beam irradiation. Results: The study demonstrated that low doses(10 Gy) of ion beam have a stimulating effect on the height, root length, and fresh weight of the plantlets but not on the number of leaves. Meanwhile, doses higher than 10 Gy caused reductions in all the morphological parameters studied as compared to the control samples. The highest total soluble protein content [(2.11±0.47) mg/g FW] was observed in plantlets irradiated at 20 Gy. All irradiated plantlets were found to have 0.85% to 58.32% higher specific activity of peroxidase as compared to the control samples. The present study also revealed that low doses of ion beam(10 and 20 Gy) had negligible effect on the total chlorophyll content of O. sativa plantlets while 40 Gy had a stimulating effect on the chlorophyll content. Plantlets irradiated between 40 to 120 Gy were shown to be 0.38% to 9.98% higher in total soluble nitrogen content which, however, was not significantly different from the control samples. Conclusions: Carbon ion beam irradiation administered at low to moderate doses of 10 to 40 Gy may induce O. sativa mutants with superior characteristics.

Impact damage behavior of sandwich composite with aluminum foam core

Impact property of the sandwich composite with aluminum foam core was investigated by experiment and simulation analysis. Impact energies of 50, 70 and 100 J were applied to the specimens in impact tests. The results show that the striker penetrates the upper face sheet, causing the core to be damaged at 50 J test but the lower face sheet remains intact with no damage. At 70 J test, the striker penetrates the upper face sheet and the core,and causes the lower face sheet to be damaged. Finally at 100 J test, the striker penetrates both the upper face sheet and the core, and even the lower face sheet. The experimental and simulation results agree with each other. By the confirmation with the experimental results, all these simulation results can be applied on structure study of real sandwich composite with aluminum foam core effectively.

Recovery of cerium from glass polishing slurry

Cerium was extracted from rare earth slurry waste used for polishing a glass ***,glass frit and flocculant were removed by froth flotation and *** recovered rare earth slurry exhibited almost the same particle size distribution as original slurry,which could be reused as slurry for glass *** the rare earth slurry,the cerium solution was obtained by an oxidative thermal treatment and subsequent chemical *** cerium solution was further purified up to 94% by selective precipitation of rare earth species.

In this review, we examine the current theoretical and experimental status of the chiral magnetic *** discuss possible future strategies for resolving uncertainties in interpretation including recommendations for theoretical work, recommendations for measurements based on data collected in the past five years, and recommendations for beam use in the coming years of RHIC. We specifically investigate the case for colliding nuclear isobars（nuclei with the same mass but different charge） and find the case compelling. We recommend that a program of nuclear isobar collisions to isolate the chiral magnetic effect from background sources be placed as a high priority item in the strategy for completing the RHIC mission.

This paper first introduces the concept of a geogram that captures richer features to represent the objects. The spatiogram contains some moments upon the coordinates of the pixels corresponding to each bin, while the geogram contains information about the perimeter of grouped regions in addition to features in the *** we consider that a convergence process of mean shift is divided into obvious dynamic and steady states,and introduce a hybrid technique of feature description, to control the convergence process. Also, we propose a spline resampling to control the balance between computational cost and accuracy of particle filtering. Finally, we propose a boosting-refining approach, which is boosting the particles positioned in the ill-posed condition instead of eliminating the ill-posed particles, to refine the particles. It enables the estimation of the object state to obtain high accuracy. Experimental results show that our approach has promising discriminative capability in comparison with the state-of-the-art approaches.

Effect of metronome rates on the quality of bag-mask ventilation during metronome-guided 30:2 cardiopulmonary resuscitation:A randomized simulation study

BACKGROUND: Metronome guidance is a feasible and effective feedback technique to improve the quality of cardiopulmonary resuscitation(CPR). The rate of the metronome should be set between 100 to 120 ticks/minute and the speed of ventilation may have crucial effect on the quality of ventilation. We compared three different metronome rates(100, 110, 120 ticks/minute) to investigate its effect on the quality of ventilation during metronome-guided 30:2 ***: This is a prospective, randomized, crossover observational study using a Respi Trainer r. To simulate 30 chest compressions, one investigator counted from 1 to 30 in cadence with the metronome rate(1 count for every 1 tick), and the participant performed 2 consecutive ventilations immediately following the counting of 30. Thirty physicians performed 5 sets of 2 consecutive(total 10) bag-mask ventilations for each metronome rate. Participants were instructed to squeeze the bag over 2 ticks(1.0 to 1.2 seconds depending on the rate of metronome) and defl ate the bag over 2 ticks. The sequence of three different metronome rates was ***: Mean tidal volume significantly decreased as the metronome rate was increased from 110 ticks/minute to 120 ticks/minute(343±84 m L vs. 294±90 m L, P=0.004). Peak airway pressure significantly increased as metronome rate increased from 100 ticks/minute to 110 ticks/minute(18.7 vs. 21.6 mm Hg, P=0.006).CONCLUSION: In metronome-guided 30:2 CPR, a higher metronome rate may adversely affect the quality of bag-mask ventilations. In cases of cardiac arrest where adequate ventilation support is necessary, 100 ticks/minute may be better than 110 or 120 ticks/minute to deliver adequate tidal volume during audio tone guided 30:2 CPR.

Some selected medicinal plants used by Bentian tribe from Indonesia were evaluated for potential antimicrobial and antioxidant *** leaves stem bark and root of the plants were extracted with n-hexane,ethyl acetate and ethanol to give respective *** activity against Propionibacterium acnes and Candida albicans was determined by an agar well diffusion *** activity was assayed by the DPPH radical scavenging activity *** results showed that n-hexane,ethyl acetate and ethanol extracts of Cananga odorata,ethanolic extract of Chromolaena odorata,and ethanolic extracts of Hyptis capitata and Ampelocissus cinnamomeae displayed good activity against *** at 25–400μg/well of the extracts *** n-hexane and ethanol extracts of Chromolaena odorata and ***,and the n-hexane,ethyl acetate and ethanol extracts of Cananga odorata displayed more activity against *** than others at 25–400μg/well of the extracts *** most antioxidant activities against DPPH were displayed by the ethanol extracts of Ficus variegata stem bark,Leucosyke quadrinerva root and Clausena excavata leaves exhibiting 91%,91%and 86%inhibition,*** present results showed potential of some medicinal plants used by Bentian tribe from Indonesia as natural anti-microbial and anti-oxidant and potentially anti-inflammatory agents.
