目的探讨济南市2013―2019年臭氧(Ozone,O_(3))暴露对居民超额应急呼叫风险的影响。方法收集济南市2013―2019年每日的居民应急呼叫事件信息,以及同期每日的空气污染物(O_(3)、PM_(2.5)、PM_(10)、NO_(2)、SO_(2)、CO)浓度和气象因素(日均温度、日均相对湿度和日均风速)资料,采用病例交叉设计方法确定暴露-反应系数,Cox比例风险回归模型进行健康风险评估;对呼吸、循环和神经系统疾病进行分层分析,同时比较冬春季和夏秋季O_(3)暴露造成的超额应急呼叫风险是否具有差异。结果2013―2019年济南市共发生应急呼叫事件653473件,平均每日应急呼叫总量为250人次。通过总应急呼叫的影响效应分析和超额风险评估发现,O_(3)浓度每升高10μg/m3,居民总应急呼叫的风险增加0.375%(95%CI:0.288%~0.461%),造成的超额应急呼叫总量为7226人次。疾病分层分析发现,研究期间O_(3)暴露仅对增加神经系统疾病应急呼叫的超额风险差异有统计学意义(P冬春季。结论济南市O_(3)暴露增加居民的超额应急呼叫风险,且夏秋季O_(3)暴露造成的超额应急呼叫风险高于冬春季,特别是神经系统疾病受其影响更为显著。

[背景]近年来中国夏季区域性高温天气频繁出现,中暑是高温所导致的代表性气象病。分析中暑流行特征及热浪暴露对其影响,对做好中暑的监测、预警和防控工作具有重要意义。[目的]了解济南市中暑病例的流行病学特征,探讨热浪暴露对中暑的影响。收集2017-2022年济南市中暑病例的个案信息以及同期逐日的气象因素数据,描述济南市中暑病例的发生时间、人群和地区分布特征,采用时间分层的病例交叉设计并结合条件logistic回归模型,探索12种热浪定义(不同强度和持续时间组合)下热浪暴露对中暑的影响。其中,热浪强度分别使用研究期间日均温度的第90百分位数(P_(90))和P_(95)、P_(97.5)、P_(99)表示,持续时间包括≥2 d、≥3 d、≥4 d,记为Pi(k),i为温度阈值,k为持续时间,如P90(2)即表示连续至少2 d的日均温度大于等于研究期间日均温度的第90百分位数对应的温度值。以lag01表示滞后1 d的累积滞后效应,以此类推。[结果]2017-2022年济南市共报告中暑病例1394例,轻症患者581例,重症患者813例;累积报告死亡85例,累积病死率为6.10%。流行特征分析结果显示:研究期间报告的中暑病例集中分布于6-8月,且在7月出现高峰(665例,47.70%);中暑病例中男女性别比为2.02:1,病例的高发年龄段为50~89岁,且在70~79岁组和50~59岁组出现两次高峰,但最高峰出现在70~79岁;病例高发地区分布于济南市西部的中心城区(天桥区,274例,19.66%;槐荫区,223例,16.00%)和周边农村地区(平阴县,254例,18.22%)。热浪暴露影响效应分析结果显示,热浪暴露对中暑的影响具有统计学意义。对中暑影响的最大效应估计值分别出现在P_(99)(2)、P_(97.5)(3)和P_(97.5)(4)定义下的lag04、lag03、lag04,OR(95%CI)分别为9.27(4.71~14.24)、8.95(6.17~12.98)和8.22(4.91~13.78)。暴露-反应关系曲线提示,随着日均温度的升高,中暑风险呈上升趋势。[结论]每年7月是济南市居民中暑的高发时期,同时以男性病例为主,重症多,年龄集中在50~89岁。热浪的发生可进一步增加中暑的风险,且存在显著的滞后效应。

Species closely related to wheat are important genetic resources for agricultural production,functional genomics studies and wheat *** this study,a wheat gene related to regeneration,TaWOX5,was applied to establish the Agrobacterium-mediated transformation systems of Triticum monococcum,hexaploid triticale,and rye(Secale cereale L.)using their immature *** plants were efficiently *** the transformation process,the Agrobacterium infection efficiency was assessed by histochemical staining forβ-glucuronidase(GUS).Finally,the transgenic nature of regenerated plants was verified by polymerase chain reaction(PCR)-based genotyping for the presence of the GUS and bialaphos resistance(bar)genes,histochemical staining for GUS protein,and the QuickStix strip assay for bar *** transformation efficiency of *** genotype PI428182 was 94.4%;the efficiencies of four hexaploid triticale genotypes Lin456,ZS3297,ZS1257,and ZS3224 were 52.1,41.2,19.4,and 16.0%,respectively;and the transformation efficiency of rye cultivar Lanzhou Heimai was 7.8%.Fluorescence in situ hybridization(FISH)and genomic in situ hybridization(GISH)analyses indicated that the GUS transgenes were integrated into the distal or near centromere(proximal)regions of the chromosomes in transgenic *** and hexaploid triticale *** the transgenic hexaploid triticale plants,the foreign DNA fragment was randomly integrated into the AABB and RR ***,the transgene was almost stably inherited in the next generation by Mendel’s *** findings in this study will promote the genetic improvement of the three plant species for grain or forage production and the improvement of cereal species including wheat for functional genomics studies.

To identify linear epitopes on the non-structural protein 3AB of foot-and-mouth disease virus (FMDV), BABL/c mice were immunized with the 3AB protein and splenocytes of BALB/c mice were fused with myeloma Sp2/0 cells. Two hybridoma monoclonal antibodies (mAbs) cell lines against the 3AB protein of foot-and-mouth disease virus (FMDV) were obtained, named C6 and E7 respectively . The microneutralization titer was 1:1024 for mAb C6, and 1:512 for E7. Both mAbs contain kappa light chains, and were of subclass IgG2b. In order to define the mAbs binding epitopes, the reactivity of these mAbs against FMDV were examined by indirect ELISA. The results showed that both mAbs can react with FMDV, but had no cross-reactivity with Swine Vesicular Disease (SVD) antigens. The titers in abdomen liquor were 1:5×106 for C6 and 1:2×106 for E7. In conclusion, the mAbs obtained from this study are specific for the detection of FMDV, can be used for etiological and immunological researches on FMDV, and have potential use in diagnosis and future vaccine designs.

Foot-and-mouth disease (FMD),caused by foot-and-mouth disease virus (FMDV),is considered one of the most important viral diseases of cloven-hoofed animals,causing severe economic losses in affected regions of the *** serotypes (A,O,and Asia 1) of FMDV have been identified in China since *** addition,the occurrence of novel subtypes within these serotypes has made the epidemiology of FMDV more complicated over the last few *** this review,we summarize the history and the current epidemiological situation in China,genetic diversity (e.g.,quasispecies dynamics,antigenic heterogeneity,and functional constraints),intertypic recombination,and the evidence for positive selection of different FMDV *** also assess these genetic data to understand the origin,evolution,and transmission of FMDV,the findings of which may be useful in developing control measures for future epidemics.

The complete nucleotide sequence of genomic RNA of foot and mouth disease virus (FMDV) strain China/99 from infected bovine tongue epithelium is presented. The nucleotide sequence extending from the 5′ end of the genomic RNA to the 5′ end of poly (A) tail contains 8173 nucleotides (nt). Its open reading frame, which encodes a single polypeptide of 2332 amino acids, encompasses 6999 nt starting from the initiation codon AUG and terminating at the UAA codon 93 bases upstream from the 5′ end of poly (A) tract. The 5′ untranslated region (UTR) is composed of 1081nt. The consensus of the 1d gene of FMDV strain China/99 compared with that of UKG/6/2001, UKG/12/2001, China/99HN4 and China/3/Tibet is over 97%. The result showed the stains belong to the members of the Pan-Asia family. There is a remarkable differentiation in the function-unknown (FUR), p2 and p3 regions between FMDV isolates from infected cattle and swine, especially in 3a gene. No deletion was found in genes l, 1a, 1b, 2a, 2c, 3b, and 3d. These genes might be indispensable to the surviving of FMDV. The secondary structures of small (S) fragments, FUR and an internal ribosome entry site can be classified into three types, and the S fragment and 3′UTR of the positive-sense RNA fold into stem-loop structures similar to the shape of clover.

The full-length cDNA clone of swine vesicular disease virus HK/70 strain named pSVOK12 was constructed in order to study the antigenicity, replication, maturation and pathogenicity of swine vesicular disease virus. In vitro transcription RNA from pSVOK12 transfected IBRS-2 cells and the re- covered virus RNA were isolated and sequenced, then indirect hemagglutination test, indirect im- munofluorescence assays, eleectron microscope test, 50% tissue culture infecting dose (TCID50) assays and mouse virulence studies were performed to study the antigenicity and virulence of the recovered virus. The result showed that the infectious clones we ob- tained and the virus derived from pSVOK12 had the same biological properties as the parental strain HK/70. The full-length infectious cDNA clone, pSVOK12, will be very useful in studies of the anti- genicity, virulence, pathogenesis, maturation and replication of SVDV.

旨在解决如何对现有方法进行优化创新以建立口蹄疫抗体检测新方法的问题,本研究在基于原有液相阻断ELISA的基础上进行方法优化,实现口蹄疫病毒抗体的快速、灵敏检测。本研究通过对抗口蹄疫IgG抗体标记生物素技术与HRP标记链霉亲和素(HRP-SA)相结合建立新的液相阻断ELISA检测方法(SA-LPBE),在猪,牛、羊血清样品中具有较高的符合率。结合实际情况,判定优化后牛、羊血清抗体效价≥128,猪血清效价≥64时,判定为阳性,与商用检测试剂盒结果一致。并且经符合率分析,与原试剂盒具有92.3%的符合率,其批内变异系数<5%,批间变异系数<10%。该方法改变了原有的需要制备兔抗豚鼠抗血清再标记HRP技术路线,根本上提高了酶促反应的效率,保证了方法原有敏感性的同时提高了方法的特异性;利用生物素标记抗体与HRP-SA反应迅速、标记原料稳定、检测背景值等优点实现了将液相阻断ELISA试剂盒操作时间由2 d缩短为3 h,解决了原有方法操作繁琐、用时过长、操作疲劳、不稳定、易出错等问题。建立的SA-LPBE可应用于FMDV抗体检测,为FMD流行和临床检测提供了技术方法。
