Fast,simple,efficient Agrobacterium rhizogenes-mediated transformation system to non-heading Chinese cabbage with transgenic roots

Non-heading Chinese cabbage, a variety of Brassica campestris, is an important vegetable crop in the Yangtze River Basin of China. However,the immaturity of its stable transformation system and its low transformation efficiency limit gene function research on non-heading Chinese cabbage. Agrobacterium rhizogenes-mediated(ARM) transgenic technology is a rapid and effective transformation method that has not yet been established for non-heading Chinese cabbage plants. Here, we optimized conventional ARM approaches(one-step and two-step transformation methods) suitable for living non-heading Chinese cabbage plants in nonsterile environments. Transgenic roots in composite non-heading Chinese cabbage plants were identified using phenotypic detection, fluorescence observation, and PCR analysis. The transformation efficiency of a two-step method on four five-day-old non-heading Chinese cabbage seedlings(Suzhouqing, Huangmeigui, Wuyueman, and Sijiu Caixin) was 43.33%-51.09%, whereas using the stout hypocotyl resulted in a transformation efficiency of 54.88% for the 30-day-old Sijiu *** one-step method outperformed the two-step method;the transformation efficiency of different varieties was above 60%, and both methods can be used to obtain transgenic roots for functional studies within one month. Finally, optimized ARM transformation methods can easily,quickly, and effectively produce composite non-heading Chinese cabbage plants with transgenic roots, providing a reliable foundation for gene function research and non-heading Chinese cabbage genetic improvement breeding.

Jujube witches’broom(JWB)is caused by infection with a phytoplasma.A multi-omics approach was taken during graft infection of jujube by JWB-infected scion through the analysis of the plant transcriptome,proteome and phytohormone levels.A high number of differentially expressed genes(DEGs)were identified 37 weeks after grafting(WAG),followed by observation of typical symptoms of JWB at 48 *** 37 WAG,the majority of the upregulated DEGs and differentially expressed proteins(DEPs)were related to flavonoid biosynthesis,phenylalanine metabolism and phenylpropanoid *** of the four upregulated proteins were similar to jasmonate-induced *** the downregulated genes,the two most populated GO terms were plant–pathogen interaction and plant hormone signal transduction(mainly for tryptophan metabolism).Moreover,phytoplasma infection resulted in reduced auxin content and increased jasmonate content,indicating that auxin and jasmonic acid have important roles in regulating jujube responses during the first and second stages of phytoplasma *** 48 WAG,the two largest groups of upregulated genes were involved in phenylpropanoid biosynthesis and flavonoid *** genes and proteins involved in carbon metabolism and carbon fixation in photosynthetic organisms were downregulated,indicating that photosynthesis was affected by the third stage of phytoplasma infection.

The local Ca2+release from the heterogeneously distributed endoplasmic reticulum(ER) calcium store has a critical role in calcium homeostasis and cellular function. However, single fluorescent proteinbased ER calcium probes experience challenges in quantifying the ER calcium store in differing live cells, and intensity-based measurements make it difficult to detect local calcium microdomains in the ER. Here, we developed a genetically encoded ratiometric ER calcium indicator(GCEPIA1-SNAPER) that can detect the real-time ER calcium store and local calcium microdomains in live cells. GCEPIA1-SNAPERwas located in the lumen of the ER and showed a linear, reversible and rapid response to changes in the ER calcium store. The GCEPIA1-SNAPERprobe effectively monitored the depletion of the ER calcium store by TG or starvation treatment, and through its use we identified heterogeneously distributed calcium microdomains in the ER which were correlated with the distribution of STIM1 clusters upon ER calcium store depletion. Lastly, GCEPIA1-SNAPERcan be used to detect the ER calcium store by highthroughput flow cytometry and confers the ability to study the function of calcium microdomains of the ER.

The safety and antimicrobial efficacy of Ethyl lauroyl arginate HCI while using as a preservative have been introduced. Meanwhile the antimicrobial mechanism, the minimal inhibitory concentration and the stability to heat and pH range of ethyl lauroyl arginate HCI have been introduced. The new preservatives have been developed, they are the compounds containing ethyl lauroyl arginate HCI. The high efficacy of antimicrobial of Ethyl lauroyl arginate HCI series products ha been described while added into the formulations of amino acid shampoo and toner. It has been proved that ethyl lauroyl arginate HCI series products can be the optimized option for the alternative of traditional preservatives for cosmetic products.

In this paper,the BIW of a domestic car establishes a finite element analysis model,with the stiffness and modal of BIW as the constraints,the thickness,size and material of the BIW structure parts are optimized to achieve lightweight *** optimizing and analyzing examples,optimization results are verified through experimental comparison,the experiment results show that quality of BIW is decreased by 10.37% after optimization,while the stiffness and modal exceed the production target *** this method,quality of BIW realizes the purpose of lightweight,for the design and production of car body provides a theoretical basis and reference instance.

On August 14th,2018,a Beijing resident living in Xicheng District found a female *** tick attached to the skin at the front of his upper *** examination,the patient was afebrile and appeared *** species of the tick was identified through morphological characteristics and phylogenetic analysis based on cytochrome C oxidase subunit *** *** tick was screened for tick-borne pathogens such as viruses,bacteria and *** pathogens were screened by PCR and sequencing,while DNA pathogens were screened by metagenomic *** was found that the tick was positive for the DNA sequences of zoonotic and animal pathogens such as A phagocytophilum,Ehrlichia minasensis and *** the good health condition of the patient,we hypothesized that the pathogens originated from the tick specimen itself rather than host blood *** the first time,our study reveals the possible risk of transmission of tick-borne pathogens to human beings through tick bit in downtown *** research is needed to screen for tick-borne pathogens among unfed ticks collected from central Beijing.
