Porous organic cages(POCs)as an innovative type of porous molecular materials enable multifunctional applications.Herein,a fluorescence POC(denoted as 1)has been constructed by means of 5/5'-((2/5-dimethoxy-l/4-phenylene)bis(ethyne-2/1-diyl))diiso-phthalaldehyde condensing with cyclohexanediamine enantiomer with the aid of trifluoroacetic acid.1 exhibits the permanent void and prominent fluorescence with relative quantum yield of 73%confirmed by gas sorption and emission experiments,respectively.Moreover,this organic cage is capable of sensing diavalent Pd2+according to the fluorescence response to the addition of various metal ions.The dispersion of 1 in methanol containing palladium acetate was stirred at 25℃and then 80℃to reduce Pd2+ions into nanoparticles(NPs),leading to the composite(Pd@l)composed of fine Pd NPs with the size of co.1.8 nm.The catalytic nature of Pd@l over NaBH4 promoted the degeneration of nitrobenzene and congo red.This work introduces one new case of POCs with versatile properties and functions,including ion fluorescence recognition,fine Pd NPs stabilizer,and gas adsorption,enriching the family of POC-based materials.

The increased prevalence of high temperatures （HTs） around the world is a major global concern, as they dramatically affect agronomic productivity. Upon HT exposure, plants sense the temperature change and initiate cellular and metabolic responses that enable them to adapt to their new environmental conditions.Decoding the mechanisms by which plants cope with HT will facilitate the development of molecular markers to enable the production of plants with improved thermotolerance. In recent decades, genetic, physiological, molecular, and biochemical studies have revealed a number of vital cellular components and processes involved in thermoresponsive growth and the acquisition of thermo- tolerance in plants. This review summarizes the major mechanisms involved in plant HT responses, with a special focus on recent discoveries related to plant thermosensing, heat stress signaling, and HT-regulated gene expression networks that promote plant adaptation to elevated environmental temperatures.

类固醇激素在人体新陈代谢、免疫调节、生长发育、维持生命功能等方面具有重要作用,但其同时又作为一类新型有机污染物,干扰人体正常的内分泌机能从而对人体产生危害,因此类固醇激素的研究已逐渐成为国际上环境和健康领域的研究热点。尿液具有无创收集、前处理简单、能敏感地反映生理变化等优势,更适用于定期、重复监测。色谱-质谱联用技术灵敏度高、选择性好、通量高,在尿液类固醇激素检测中得到了广泛应用。本文基于色谱-质谱联用技术综述了检测类固醇激素的尿样的采集和储存条件、前处理技术、色谱-质谱分析方法、分析软件和数据库,以及部分类固醇激素在我国不同健康人群尿液中的检出情况等研究进展,指出尿液中类固醇激素及其代谢物的检测基质复杂、种类繁多以及含量较低,仍需进行广泛而深入的研究,实现对多种类固醇激素及其代谢物的精确定量仍面临挑战。未来研究需建立标准化采集和储存方法,开发更高效和环境友好型的前处理技术以及高灵敏度、高可靠性和高通量的色谱-质谱仪器,合成更多类固醇激素及代谢物标准品和同位素,同时不断完善数据库信息以满足精确定量的需求,为尿液中类固醇激素检测及人群健康风险评估等研究提供参考。

Importance:First branchial cleft anomalies(FBCAs)are rare congenital malformations,accounting forrently known about the cause of FBCAs at the molecular level.Objective:To identify genomic alterations related to the genetic etiology of FBCAs in Chinese children.Methods:We performed whole-exome sequencing of samples from 10 pediatric patients with FBCAs.Data analysis was carried out using the Burrow-Wheeler Alignment software package,and the dbSNP database for comparisons.Rare variants were further validated by Sanger sequencing.Insertion/deletions(indels)were examined using the Genome Analysis Toolkit.Results:We identified 14 non-synonymous mutations in seven potential FBCA-susceptibility genes(TRAPPC12,NRP2,NPNT,SH3RF2,RHPN1,TENM4,and ARMCX4).We also detected 133 shared small indels in 125 genes.Gene Ontology analysis indicated that most of the identified genes played critical roles in development and differentiation pathways involved in regulating organ development.Interpretation:We characterized the mutational landscape in pathways involved in development and differentiation in Chinese children with FBCA.The results identified potential pathogenic genes and mutations related to FBCA,and provide molecular-level support for the branchial theory of FBCA pathogenesis.

为了研究S型阴离子通道蛋白(slow type anion channel,SLAC)基因家族在毛竹(Phyllostachys edulis)响应干旱胁迫中的作用,利用生物信息方法以拟南芥和水稻中的SLAC/SLAH家族蛋白序列为模板对毛竹中SLAC家族成员进行鉴定,对其基因结构、进化关系和组织表达等进行分析,并对PheSLAC1的保守功能位点以及在干旱胁迫中的功能进行初步研究。结果表明:毛竹中包含15个SLAC家族基因,其中PheSLAC1.1(PH02Gene01933.t1)和Phe SLAC1.2(PH02Gene00592.t1)在叶片中表达量最高,基因编码区(coding sequence,CDS)序列长度分别为1 716和1 677 bp,分别编码572和559个氨基酸,且包含保守的关键功能位点。亚细胞定位结果表明,PheSLAC1.1和Phe SLAC1.2均定位于细胞质膜中。PheSLAC1.1和Phe SLAC1.2在拟南芥slac1-3突变体中的表达结果表明,其能够部分减轻突变体的干旱敏感表型,表明两者均能够通过调控气孔关闭在毛竹响应干旱胁迫中发挥重要作用。

并行测试技术可以同时进行多个任务的测试,提高资源利用率,节约测试成本;并行测试调度问题是一种复杂的组合优化问题,是并行测试技术的核心要素;并行测试系统作为并行测试技术的载体,自身的性能和求解效率尤其重要;文章对并行测试完成时间极限定理进行了研究,建立了并行测试任务调度的数学模型,分析了传统元启发式算法求解并行测试问题的不足,提出了基于动态规划的递归搜索技术和人工蜂群算法相结合的混合人工蜂群算法,并采用整数规划精确算法和遗传算法对混合人工蜂群算法进行验证;得出结论采用混合人工蜂群算法进行并行测试任务的调度节约了接近50%的时间,降低了约20%的硬件资源占用,提高了测试效率,可以满足工程实际的应用。

于2021-2022年连续2年研究浅埋滴灌条件下不同氮肥基追比对甜菜叶片厚度、单位长度内的叶脉数量、株高、叶面积指数(LAI)、根冠比、单株干物质量、产量和含糖率的影响。结果表明,随氮肥追肥比例的增加,含糖率、叶片厚度和根冠比呈降低的趋势,而产量、产糖量、株高、叶面积指数和单株干物质积累量显著增加,N3处理(氮肥按基追比6:4)比CK处理(氮肥全部基施)产量提高6.93%~8.04%、产糖量提高6.12%~8.94%。相关性分析表明,甜菜产量与株高、LAI、单株干物质量呈显著正相关关系,而与叶片厚度呈显著负相关关系。因此,浅埋滴灌条件下,氮肥按基追比6:4有利于甜菜产量和产糖量的提高。

Osteoporosis is a common disease in the middle-aged and elderly population and is a global health problem that needs to be solved urgently.Vitamin D deficiency is closely related to osteoporosis,and it is of practical significance to prevent and treat kidney and strengthen bone therapy.A large number of literature have confirmed that qi and blood to represent Danggui Buxue Decoction has a reliable therapeutic effect on osteoporosis,but the mechanism is still unclear.Based on the previous work of the research group,it is believed that the vitamin D axis may be a potential target for TCM Bushen Recipe.Syndrome and vitamin D deficiency have similar pathological changes,so it is suggested that Danggui Buxue Decoction may regulate osteoporosis by regulating the vitamin d axis.

Small molecule drugs play a pivotal role in the arsenal of anticancer pharmacological agents. Nonetheless, their small size poses a challenge when directly visualizing their localization, distribution, mechanism of action (MOA), and target engagement at the subcellular level in real time. We propose a strategy for developing triple-functioning drug beacons that seamlessly integrate therapeutically relevant bioactivity, precise subcellular localization, and direct visualization capabilities within a single molecular entity. As a proof of concept, we have meticulously designed and constructed a boronic acid fluorescence drug beacon using coumarin–hemicyanine (CHB). Our CHB design includes three pivotal features: a boronic acid moiety that binds both adenosine triphosphate (ATP) and adenosine diphosphate (ADP), thus depleting their levels and disrupting the energy supply within mitochondria;a positively charged component that targets the drug beacon to mitochondria;and a sizeable conjugated luminophore that emits fluorescence, facilitating the application of structured illumination microscopy (SIM). Our study indicates the exceptional responsiveness of our proof-of-concept drug beacon to ADP and ATP, its efficacy in inhibiting tumor growth, and its ability to facilitate the tracking of ADP and ATP distribution around the mitochondrial cristae. Furthermore, our investigation reveals that the micro-dynamics of CHB induce mitochondrial dysfunction by causing damage to the mitochondrial cristae and mitochondrial DNA. Altogether, our findings highlight the potential of SIM in conjunction with visual drug design as a potent tool for monitoring the in situ MOA of small molecule anticancer compounds. This approach represents a crucial advancement in addressing a current challenge within the field of small molecule drug discovery and validation.
