Molybdenum and cadmium co-induce necroptosis through Th1/Th2 imbalance-mediated endoplasmic reticulum stress in duck ovaries

Cadmium(Cd)and excess molybdenum(Mo)pose serious threats to animal health.Our previous study has determined that Cd and/or Mo exposure can cause ovarian damage of ducks,while the specific mechanism is still obscure.To further investigate the toxic mechanism of Cd and Mo co-exposure in the ovary,forty 8-day-old female ducks were randomly allocated into four groups for 16 weeks,and the doses of Cd and Mo in basic diet per kg were as follows:control group,Mo group(100 mg Mo),Cd group(4 mg Cd),and Mo+Cd group(100 mg Mo+4 mg Cd).Cadmium sulfate 8/3-hydrate(CdSO_(4)·8/3H_(2)O)and hexaammonium molybdate((NH_(4))_(6)Mo_(7)O_(24)·4H_(2)O)were the origins of Cd and Mo,respectively.At the 16th week of the experiment,all ovary tissues were collected for the detection of related indexes.The data indicated that Mo and/or Cd induced trace element disorders and Th1/Th2 balance to divert toward Th1 in the ovary,which activated endoplasmic reticulum(ER)stress and then provoked necroptosis through triggering RIPK1/RIPK3/MLKL signaling pathway,and eventually caused ovarian pathological injuries and necroptosis characteristics.The alterations of above indicators were most apparent in the joint group.Above all,this research illustrates that Mo and/or Cd exposure can initiate necroptosis through Th1/Th2 imbalance-modulated ER stress in duck ovaries,and Mo and Cd combined exposure aggravates ovarian injuries.This research explores the molecular mechanism of necroptosis caused by Mo and/or Cd,which reveals that ER stress attenuation may be a therapeutic target to alleviate necroptosis.

Conserved noncoding sequences correlate with distant gene contacts in Arabidopsis and Brassica

Physical contact between genes distant on chromosomes is a potentially important way for genes to coordinate their expressions.To investigate the potential importance of distant contacts,we performed high-throughput chromatin conformation capture(Hi-C)experiments on leaf nuclei isolated from Brassica rapa and Brassica oleracea.We then combined our results with published Hi-C data from Arabidopsis thaliana.We found that distant genes come into physical contact and do so preferentially between the proximal promoter of one gene and the downstream region of another gene.Genes with higher numbers of conserved noncoding sequences(CNSs)nearby were more likely to have contact with distant genes.With more CNSs came higher numbers of transcription factor binding sites and more histone modifications associated with the activity.In addition,for the genes we studied,distant contacting genes with CNSs were more likely to be transcriptionally coordinated.These observations suggest that CNSs may enrich active histone modifications and recruit transcription factors,correlating with distant contacts to ensure coordinated expression.This study advances our knowledge of gene contacts and provides insights into the relationship between CNSs and distant gene contacts in plants.

二次创业是制造企业持续发展的重要方式,已经成为当前学术界和业界的热点,知识性和商业性资源是制造企业二次创业的关键性资源。基于RBV理论、双元创业理论,对223个有效样本企业进行实证分析。结果表明:知识性资源和商业性资源对制造企业二次创业具有正向影响,其中知识性资源对探索式创业行为的作用更强,商业性资源对利用式创业行为的作用更强;创业导向正向调节知识性资源与两种创业行为的关系,负向调节商业性资源与探索式创业的关系,对商业性资源与利用式创业行为的调节作用不明显。

OBJECTIVE:To test the hypothesis that moxibustion may inhibit rheumatoid arthritis(RA)synovial inflammation by regulating the expression of macrophage migration inhibitory factor(MIF)/glucocorticoids(GCs).METHODS:Fifty male Sprague-Dawley rats were randomly divided into five groups(n=10 each):blank Control(CON)group,RA Model(RA)group,Moxibustion(MOX)group,MIF inhibitor(S,R)-3-(4-hydroxyphenyl)-4,5-dihydro-5-isoxazole acetic acid methyl ester(ISO-1)group,and Moxibustion+MIF inhibitor ISO-1(MOX+ISO-1)group.Rats in the ISO-1 group and ISO-1+MOX group were intraperitoneally injected with the inhibitor ISO-1.The rats in the RA group,ISO-1 group,MOX group,and ISO-1+MOX group were injected with Freund's complete adjuvant(FCA)in the right hind footpad to establish an experimental RA rat model.In the MOX group and MOX+ISO-1 group,rats were treated with Moxa.The thickness of the footpads of the rats in each group was measured at three-time points before,after modeling and after moxibustion treatment.The contents of serum MIF,corticosterone(CORT),tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were detected by enzyme-linked immunosorbent assay;and the contents of synovial MIF were detected by Western blot.Hematoxylin-eosin(HE)staining method was used to observe the pathological changes of synovial tissue under a section light microscope,and pathological scoring was performed according to the grading standard of the degree of synovial tissue disease.RESULTS:Moxibustion was found to reduce the level of MIF and alleviate inflammation in RA rats in this study.In addition,after inhibiting the expression of MIF,the level of CORT increased,and the level of TNF-α decreased.Treating RA rats with inhibited MIF by moxibustion,the level of CORT was almost unchanged,but the level of TNF-α further decreased.The correlation analysis data suggested that MIF was positively related to the expression of TNF-α and negatively correlated with the expression of CORT.CONCLUSION:Reducing MIF to increase CORT and decrease TNF-α by moxibustion

With the rapid development of information technology,IoT devices play a huge role in physiological health data detection.The exponential growth of medical data requires us to reasonably allocate storage space for cloud servers and edge nodes.The storage capacity of edge nodes close to users is limited.We should store hotspot data in edge nodes as much as possible,so as to ensure response timeliness and access hit rate;However,the current scheme cannot guarantee that every sub-message in a complete data stored by the edge node meets the requirements of hot data;How to complete the detection and deletion of redundant data in edge nodes under the premise of protecting user privacy and data dynamic integrity has become a challenging problem.Our paper proposes a redundant data detection method that meets the privacy protection requirements.By scanning the cipher text,it is determined whether each sub-message of the data in the edge node meets the requirements of the hot data.It has the same effect as zero-knowledge proof,and it will not reveal the privacy of users.In addition,for redundant sub-data that does not meet the requirements of hot data,our paper proposes a redundant data deletion scheme that meets the dynamic integrity of the data.We use Content Extraction Signature(CES)to generate the remaining hot data signature after the redundant data is deleted.The feasibility of the scheme is proved through safety analysis and efficiency analysis.

A facile approach has been developed to synthesize FeO@PAA(polyacrylic acid) nanoparticles(NPs) with carboxyl groups on the surfaces by polymerization with acrylic acid in FeO NPs aqueous suspension. Nitrilotriacetic acid(NTA) was conjugated to the magnetic NPs via well-known carboniimide chemistry using EDC and NHS. The Ni ions loaded on the surface of NPs provide abundant docking sites for immobilization of his-tagged green fluorescent proteins(his-tagged GFP). Successful surface modification was confirmed by dynamic light scattering(DLS), transmission electron microscopy(TEM), FT-IR spectroscopy, X-ray diffraction(XRD), and thermogravimetric analysis. The nanocomposites with multilayer adsorption sites, high magnetic property of FeO@PAA@NTA-Ni allows an easy separation of the NPs from solution under an external magnetic field, with high his-tagged protein binding capacity(42 μg protein/mg of NPs). These materials show great potential to separate his-tagged protein with low-cost purification at industrial scale.

Lignocellulosic biomass is a critical renewable carbon resource,but most of its utilization is inefficient,and elec-trocatalytic oxidation is a promising method of upgrading lignocellulose into value-added fuels and chemicals under mild operating conditions.Recently,efforts to enable conversion with a high efficiency and low energy con-sumption have been reported,but understanding the reaction mechanisms and realizing scaled-up applications of the electrooxidation of lignocellulosic biomass are still in their early stages.A timely overview of recently reported general reaction mechanisms,particularly the strategies developed for use in improving the reaction efficiencies,is necessary to inspire research regarding the highly efficient utilization of lignocellulose.Herein,we summa-rize the strategies developed to improve electrocatalytic performance in oxidative lignocellulose conversion.The organized summary includes strategies ranging from designing efficient electrocatalysts and adding functional co-catalysts or electrolytes to employing advanced electrolyzers.A comprehensive overview of representative examples should provide universal principles to yield insight into the reaction processes and guide the design of efficient electrocatalytic systems.Finally,the challenges and opportunities in developing the electrocatalytic oxidative upgrading of lignocellulosic biomass in the near future are proposed.

为研究华北板块西南缘中侏罗世煤层地球化学特征及其沉积环境,以义马盆地中侏罗世义马组2-3号煤层为研究对象,借助X射线荧光光谱仪、等离子体质谱仪等手段,运用地球化学理论和方法,详细研究了煤中地球化学特征及其所反映的沉积环境。研究结果表明:研究区常量元素含量与中国煤相比,SiO_(2)、P_(2)O_(5)和K_(2)O显著偏高,而Al_(2)O_(3)含量较低;微量元素中Cs富集,Sb、Nb、Ta、Zr、Hf亏损;稀土元素整体为轻微富集,δEu负异常,而δCe正异常,表明采样煤层未受岩浆、热液、海水影响,物源主要是陆源碎屑;Th/U、V/(V+Ni)、Ce异常、Sr/Cu及气候指数分析表明,采样煤层古泥炭堆积于还原环境,成煤期整体处于半干旱气候。
