The pollen receptor kinases (PRK) are critical regulators of pollen tube growth. The Arabidopsis genome encodes eight PRK genes, of which six are highly expressed in pollen tubes. The potential functions of AtPRK1 through AtPRK5, but not of AtPRK6,in pollen growth were analyzed in tobacco. Herein, AtPRK6 was cloned, and its function was identified. AtPRK6 was expressed specifically in pollen tubes. A yeast two-hybrid screen of AtPRK6 against 14 Arabidopsis Rop guanine nucleotide exchange factors (RopGEFs) showed that AtPRK6 interacted with AtRopGEF8 and AtRopGEF12. These interactions were confirmed in Arabidopsis mesophyll protoplasts. The interactions between AtPRK6 and AtRopGEF8/12 were mediated by the C-termini of AtRopGEF8/12 and by the juxtamembrane and kinase domain of AtPRK6, but were not dependent on the kinase activity. In addition, transient overexpression of AtPRK6::GFP in Arabidopsis protoplasts revealed that AtPRK6 was localized to the plasma membrane. Tobacco pollen tubes overexpressing AtPRK6 exhibited shorter tubes with enlarged tips. This depolarized tube growth required the kinase domain of AtPRK6 and was not dependent on kinase activity. Taken together, the results show that AtPRK6,through its juxtamembrane and kinase domains (KD), interacts with AtRopGEF8/12 and plays crucial roles in polarized growth of pollen tubes.

Abscisic acid （ABA） regulates numerous physiological and developmental processes in plants. Recent studies identify intracellular ABA receptors, implicating the transport of ABA across cell membranes as crucial for ABA sensing and response. Here, we report that a DTX/Multidrug and Toxic Compound Extrusion （MATE） family member in Arabidopsis thaliana, AtDTX50, functions as an ABA efflux transporter. When expressed heterologously in both an Escherichia coli strain and Xenopus oocyte cells, AtDTX50 was found to facilitate ABA efflux. Furthermore, dtx50 mutant mesophyll cells preloaded with ABA released less ABA compared with the wild-type （WT）. The AtDTX50 gene was expressed mainly in the vascular tissues and guard ceils and its expression was strongly up-regulated by exogenous ABA. The AtDTX50：：GFP fusion protein was localized predominantly to the plasma membrane. The dtx50 mutant plants were observed to be more sensitive to ABA in growth inhibition. In addition, compared with the WT, dtx50 mutant plants were more tolerant to drought with lower stomatal conductance, consistent with its function as an ABA efflux carrier in guard cells.

Dear Editor, Root hairs create an enormous surface area to aid plants in nutrient and water acquisition, anchorage in the soil, and microbe interactions. Root hairs develop in two stages, including initiation and tip growth. Initiation occurs when a portion of genetically determined epidermal cells develop wall loosening and form a disc-shaped area of cell swelling.

Heterotrimeric GTP-binding proteins, which consist of Gα, Gβ, and Gγ subunits, play important roles in transducing extracellular signals perceived by cell surface receptors into intracellular physiological responses. In addition to a single prototypical Gα protein （GPA1）, Arabidopsis has three unique Gα-Iike proteins, known as XLG1, XLG2, and XLG3, that have been found to be localized in nuclei, although their functions and mode of action remain largely unknown. Through a transcriptomic analysis, we found that XLG2 and XLG3 were rapidly induced by infection with the bacterial pathogen Pseudomonas syringae, whereas the XLG1 transcript level was not affected by pathogen infection. A reverse genetic screen revealed that the xlg2 loss-of-function mutation causes enhanced susceptibility to P. syringae. Transcriptome profiling revealed that the xlg2 mutation affects pathogen-triggered induction of a small set of defense-related genes. However, xlgl and xlg3 mutants showed no difference from wild-type plants in resistance to P. syringae, In addition, the xlg2 xlg3 double mutant and the xlgl xlg2 xlg3 triple mutant were not significantly different from the xlg2 single mutant in the disease resistance phenotype, suggesting that the roles of XLG1 and XLG3 in defense, if any, are less significant than for XLG2. Constitutive overexpression of XLG2 leads to the accumulation of abnormal transcripts from multiple defense-related genes. Through co-immunoprecipitation assays, XLG2 was found to interact with AGB1, the sole Gβ subunit in Arabidopsis, which has previously been found to be a positive regulator in resistance to necrotrophic fungal pathogens. However, no significant difference was found between three xlg single mutants, the xlg2 xlg3 double mutant, the xlgtriple mutant, and wild-type plants in resistance to the necrotrophic fungal pathogens Botrytis cinerea or Alternaria brassicicola. These results suggest that XLG2 and AGB1 are components of a G-protein complex different from the prototypical heterotrimeric G-protein

Dendritic cells （DCs） have the potency to regulate the outcome of autoimmunity through the modulation of immune responses. The induction of antigen specific tolerance is critical for prevention and treatment of allograft rejection. In the present Study, we transfected IL-10 gene into DCs and investigated their effect on inhibition of lymphocyte activity in vitro and induction of immune tolerance on islet allograft in mice. An IDDM C57BL/6 mouse model was induced by streptozotocin. The islet cells isolated from the BALB/c mice were transplanted into the kidney capules of the model mice followed by injection of IL-10 modified DCs （mDCs）. The results showed that mDCs could significantly inhibit T lymphocyte proliferation mediated by allotype cells and induce its apoptosis, whereas, unmodified DCs （umDCs） could promote the murine lymphocyte proliferation markedly. The injection of mDCs could prolong the survival of allotype islet transplanted IDDM mice. The average plasma glucose （PG） level in mDCs treated mice returned to normal within 3 days and lasted for about 2 weeks. The rejection response in control mice occurred for 5 days after transplantation. The level of IFN-γ was lower while IL-4 was higher in mDCs treated mice than that in umDCs treated mice, which indicated that Thl/Th2 deviation occurred. Our studies suggest that IL-10 gene modified DCs can induce the immune tolerance to islet graft and prolong survival of the recipients by the inhibiting of T cell proliferation in allotype mice. Oscillations in cytosolic free calcium determine the polarity of tip-growing root hairs.The Ca2+channel cyclic nucleotide gated channel 14(CNGC14)contributes to the dynamic changes in Ca^2+concentration gradient at the root hair tip.However,the mechanisms that regulate CNGC14 are unknown.In this study,we detected a direct interaction between calmodulin 7(CaM7)and CNGC14 through yeast two-hybrid and bimolecular fluorescence complementation assays.We demonstrated that the third EF-hand domain of CaM7 specifically interacts with the cytosolic C-terminal domain of CNGC14.A two-electrode voltage clamp assay showed that CaM7 completely inhibitsCNGC14-mediated Ca2+influx,suggesting that CaM7 neg-atively regulates CNGC14-mediated calcium signaling.Fur-thermore,CaM7 overexpressing lines phenocopy the short root hair phenotype of a cngc14 mutant and this phenotype is insensitive to changes in external Ca^2+concentrations.We,thus,identified CaM7-CNGC14 as a novel interacting module that regulates polar growth in root hairs by con-trolling the tip-focused Ca2+signal.

Advanced lead-free energy storage ceramics play an indispensable role in next-generation pulse power capacitors market.Here,an ultrahigh energy storage density of~13.8 J cm^(-3)and a large efficiency of~82.4%are achieved in high-entropy lead-free relaxor ferroelectrics by increasing configuration entropy,named high-entropy strategy,realizing nearly ten times growth of energy storage density compared with low-entropy material.Evolution of energy storage performance and domain structure with increasing configuration entropy is systematically revealed for the first time.The achievement of excellent energy storage properties should be attributed to the enhanced random field,decreased nanodomain size,strong multiple local distortions,and improved breakdown field.Furthermore,the excellent frequency and fatigue stability as well as charge/discharge properties with superior thermal stability are also realized.The significantly enhanced comprehensive energy storage performance by increasing configuration entropy demonstrates that high entropy is an effective but convenient strategy to design new high-performance dielectrics,promoting the development of advanced capacitors.

目的总结并分析乳房外Paget病(extramammary Paget′s disease,EMPD)的临床及组织病理学特征,为临床诊疗提供参考。方法收集2003年9月—2023年2月就诊于本科,经临床及组织病理检查确诊的EMPD患者的临床及组织病理学资料,进行回顾性总结分析。结果共纳入EMPD患者135例,平均就诊年龄(67.3±10.2)岁,男女比例为4.6∶1,病程1个月~30年;常见受累部位男性依次为阴囊、阴茎、阴阜区及肛周,女性依次为阴唇及肛周;就诊时以红斑、斑块、丘疹结节为常见皮损;首诊误诊率达56.30%。组织病理主要表现为表皮内Paget细胞散在或呈巢状分布,75例发生真皮浸润及皮肤附属器受累,48例有肿瘤细胞间棘层松解现象。复发转移组组织病理出现真皮、附属器浸润患者数明显高于无复发转移组,两组间差异有统计学意义(P<0.05)。多因素COX比例风险模型显示,患者年龄及组织病理上是否发生棘层松解和/或真皮、附属器浸润是肿瘤复发转移的危险因素。结论EMPD好发于老年人,男性高于女性,临床误诊率高。发病年龄高、组织学上出现棘层松解、肿瘤真皮浸润及附属器受累与肿瘤的复发、转移及更差的预后相关。

Bispecific antibodies (BsAbs) of anti CD3×anti idiotype (Id) to B cell lymphocytic leukemia (CLL) were prepared by chemical conjugation and direct hybridization technique of hybridoma and hybridoma without screening markers. The specificity of BsAbs from culture supernatants or ascites was assayed by indirect ELISA and indirect immunoflurescence (IF). The results showed that BsAbs could specifically react with homologous serum IgM from patients with B CLL and cells carrying CD3 marker respectively. Cell combination test and LDH assay demonstrated that BsAb significantly increased the conjugate formation between lymphocyte activated kill (LAK) cells and Daudi cells, and enhanced the cytotoxic activity of LAK cells against Daudi cells.

High- and Ultrahigh-pressure Metamorphism and Retrogressive Textures of Gneiss in the Donghai Area——Evidence from gneisses in drillhole ZK2304

In the gneisses from the drillhole ZK2304 of the Donghai area, there have been preserved high- and ultrahigh-pressure metamorphic mineral assemblages, a series of complicated retrogressive textures and relevant metamorphic reactions. In addition to garnet, jadeititic-clinopyroxene and rutile, other peak stage (M2) minerals in some gneisses include phengite, aragonite and coesite or quartz pseudomorphs after coesite. The typical peak-stage mineral assemblages in gneisses are characterized by garnet + jadeitic-clinopyroxene + rutile + coesite, garnet + jadeitic-clinopyroxene + phengite + rutile ± coesite and garnet + jadeitic-clinopyroxene + aragonite + rutile ± coesite. The grossular content (Gro) in garnet is high and may reach 50. 1 mol%. The SiO2 content of phengite ranges from 54.37% to 54.84% with 3.54-3.57 p.f.u. Quartz pseudomorphs after coesite occur as inclusions in garnet.The gneisses of the Donghai area have been subjected to multistage recrystallization and exhibit a closewise P-T evolutional path characterized by the near-isothermal decompression. The inclusion assemblage (Hb+Ep+Bi+Pl+Qz) within garnet and other minerals has recorded a pre-peak stage (Mi) epidote amphibole fades metamorphic event. High- and ultrahigh-pressure peak metamorphism (M2) took place at T=750-860℃ and P>2.7 GPa. The symplectitic assemblages after garnet, jadeitic-clinopyroxene and rutile imply a near-isothermal decompression metamorphism (M3, M4) during the rapid exhumation. Several lines of evidence of petrography and metamorphic reactions indicate that both gneisses and eclogites have experienced ultrahigh-pressure metamorphism in the Donghai area. This research may be of great significance for an in-depth study of the metamorphism and tectonic evolution in the Su-Lu ultrahigh-pressure metamorphic belt.
