Ubiquitously transcribed tetratricopeptide repeat on chromosome X(UTX),also known as lysine(K)-specific demethylase 6A(KDM6A),functions as a tumor suppressor gene or oncogene depending on the tumor type and ***,its tumor-suppressive mechanisms remain largely ***,we investigated the clinical significance and biological effects of UTX expression in pancreatic ductal adenocarcinoma(PDA)and determined the potential mechanisms of its *** expression and its association with clinicopathologic characteristics of PDA patients were analyzed using *** mRNA and protein expression and their regulation in PDA cell lines were measured using quantitative polymerase chain reaction and Western blot *** biological functions of UTX in PDA cell growth,migration,and invasion were determined using gain-and loss-of-function assays with both in vitro and in vivo animal *** expression was reduced in human PDA cell lines and *** UTX expression was associated with poor differentiation and prognosis in *** UTX expression inhibited PDA proliferation,migration,and invasion in vitro and PDA growth and metastasis in vivo,whereas knockdown of UTX expression did the ***,UTX expression was trans-activated by GATA6 ***6-mediated PDA progression could be blocked,at least partially,by silencing UTX *** conclusion,loss of GATA6-mediated UTX expression was evident in human PDA and restored UTX expression suppressed PDA growth and ***,UTX is a tumor suppressor in PDA and may serve as a prognostic biomarker and therapeutic target.

Knowledge about the seismic elastic modulus dispersion,and associated attenuation,in fluid-saturated rocks is essential for better interpretation of seismic observations taken as part of hydrocarbon identification and time-lapse seismic surveillance of both conventional and unconventional reservoir and overburden performances.A Seismic Elastic Moduli Module has been developed,based on the forced-oscillations method,to experimentally investigate the frequency dependence of Young's modulus and Poisson's ratio,as well as the inferred attenuation,of cylindrical samples under different confining pressure *** with three standard samples showed that the measured elastic moduli were consistent with the published data,indicating that the new apparatus can operate reliably over a wide frequency range of f∈[1-2000,10^(6)]*** Young's modulus and Poisson's ratio of the shale and the tight sandstone samples were measured under axial stress oscillations to assess the frequency-and pressure-dependent *** dry condition,both samples appear to be nearly frequency independent,with weak pressure dependence for the shale and significant pressure dependence for the *** particular,it was found that the tight sandstone with complex pore microstructure exhibited apparent dispersion and attenuation under brine or glycerin saturation conditions,the levels of which were strongly influenced by the increased effective *** addition,the measured Young's moduli results were compared with the theoretical predictions from a scaled poroelastic model with a reasonably good agreement,revealing that the combined fluid flow mechanisms at both mesoscopic and microscopic scales possibly responsible for the measured dispersion.

A novel acidophile community populating waste ore deposits at an acid mine drainage site

Waste ore samples （pH 3.0） were collected at an acid mine drainage （AMD） site in Anhui, China. The present acidophillc microbial community in the waste ore was studied with 16S rRNA gene clone library and denaturing gradient gel electrophoresis （DGGE）. Eighteen different clones were identified and affiliated withActinobacteria, low G ＋ C Gram-positives, Thermomicrobia, Acidobacteria, Proteobacteria, candidate division TM7, and Planctomycetes. Phylogenetic analysis of 16S rRNA gene sequences revealed a diversity of acidophiles in the samples that were mostly novel. It is unexpected that the moderately thermophilic acidophiles were abundant in the acidic ecosystem and may play a great role in the generation of AMD. The result of DGGE was consistent with that of clone library analysis. These findings help in the better understanding of the generation mechanism of AMD and in developing a more efficient method to control AMD.

Osteoid osteoma of the patella： report of two cases

Osteoid osteoma is very rarely located in the patella, and can represent a significant diagnostic challenge, resulting in a delay of treatment. Patients with osteoid osteoma of the patella often present with knee pain that is also a typical symptom of trauma or of other diseases such as arthritis, which are much more common than osteoid osteoma. We present two young male patients diagnosed with osteoid osteoma of the patella. Each of these patients had a history of intense knee pain; however, accurate diagnosis of osteoid osteoma in the patella had been delayed for more than one year. Computed tomography （CT） scans or magnetic resonance imaging （MRI） showed a circumscribed lesion of the patella in both patients, whereas X-ray examination （posteroantedor projection） was not able to detect the tumor. Different surgical procedures were performed in these patients for resection of the tumors, and the pathology findings confirmed the diagnosis of osteoid osteoma. Both patients recovered completely from surgery.

Influence on Cellular Signal Transduction Pathway in Dairy Cow Mammary Gland Epithelial Cells by Galactopoietic Compound Isolated from Vaccariae segetalis

The galactopoietic mechanism of Vaccaria segetalis is still unknown. Understanding dibutyl phthalate （DBP） separated from Vaccaria segetalis on the expression of lactation signal transduction genes of mammary gland epithelial cells, including prlr, erα, akt1, socs2, pparγ and elf5, will be helpful to reveal the molecular mechanism. Western blot and qRT- PCR were used to study the change of prlr, erα, akt, socs2, pparγ, and elf5 expression at mRNA and protein level. Co- localization expression of prolactin receptor （PRLR） and estrogen receptor α （ERα） was observed by immunofluorescence; the expression changes of miRNAs （21, 125b, 143, and 195） and the secretion of β-casein and lactose were detected by qRT-PCR and RP-HPLC. The results showed that Vaccaria segetalis active compound had similar fuctions as estrogen and/or prolactin （PRL） in dairy cow mammary gland epithelial cells （DCMECs）, increased the expressions of prlr, erα, akt1, and elf5 genes, while repressed pparγ expressions. DBP promoted socs2 mRNA expression, but its protein expressions were repressed. Furthermore, both DBP and PRL could repress the expressions of miRNA-125b, miRNA-143 and miRNA- 195 in DCMECs. DBP could repress the expression of miRNA-21, while the influence of PRL on miRNA-21 was not certain. DBP could promote the lactation ability of DCMECs by regulating the ER and PRLR cellular signal transduction pathway.

Subchondral bone grafting reduces degenerative change of knee joint in patients of giant cell tumor of bone

Background Giant cell tumors （GCTs） most commonly occur around the *** most beneficial procedure for this disease has been extensive curettage with ***,since many GCTs may compromise the subchondral bone,surgery can further jeopardize the articular cartilage and result in secondary *** this study,we aimed to determine the factors associated with the development of degenerative arthritis and the effect of bone grafting on the prevention of secondary *** We retrospectively analyzed 76 patients with GCT around the *** mean age at first diagnosis was 31.1 *** treatments included extensive curettage and cementation with or without bone grafting in the subchondral *** follow-up was a median duration of 35 months,ranging from 18 to 113 *** The local recurrence rate was 5.3％（4/76）.Secondary degenerative changes occurred in 30.3％（23/76） of the *** than 10 mm of the residual thickness of the remaining subchondral bone was correlated with secondary degenerative changes in 57 patients （P ＜0.001）.Of these 57 patients,56.5％（13/23） treated with bone cement reconstruction alone developed secondary degenerative changes; following bone grafting,the rate decreased to 29.4％（10/34）,with a statistically significant difference （P=0.041）.Conclusions GCT patients with less residual thickness of the subchondral bone are more likely to develop degenerative arthritis after *** grafting in the subchondral bone area is recommended when the residual thickness of the subchondral bone is less than 10 mm.

Impact of migraine attacks on the blood-brain barrier

Background Cortical spreading depression can cause migraine attack, and up-regulate matrix metalloproteinase-9 （MMP-9） expression in animal. This study aimed to determine the impact on the structure and function of the blood-brain barrier by measuring plasma MMP-9 levels in patients at the acute and late stages of migraine attacks in order to elucidate the pathological mechanisms involved. Methods We recruited a case-control cohort of 38 adult migraine patients and 20 age- and gender-matched healthy control subjects. Five milliliter blood samples were collected at the acute and late stages of migraine （days 1-7）, and also from the control subjects. Solid phase double antibody sandwich enzyme-linked immunosorbent assay was used to determine plasma MMP-9 levels. Statistical analysis was performed using the SAS version 9.1. Results Initial plasma MMP-9 levels of migraine patients were significantly higher than those of controls （（12.612±0.016） μg/L vs. （6.069±0.023）μg/L, respectively, P 〈0.05）. High MMP-9 expression was observed during days 1-6 of migraine attacks, with highest expression occurring on day 3 （（17.524±0.035） μg/L）. During attacks, MMP-9 levels were similar in migraine patients with and without aura （P 〉0.05）; in addition, levels were not correlated with degree of headache pain （P 〉0.05）. Conclusions We hypothesize that migraine could lead to increased plasma MMP-9 levels resulting in blood-brain barrier damage. MMP-9 levels increase during days 1-6 of migraine attacks, peaking on day 3. Therefore, MMP-9 could be used as a biological marker to guide treatment of migraine attacks.
