Rno_circ_0005139 regulates apoptosis by targeting Wnt5a in rat anorectal malformations

BACKGROUND The molecular mechanisms underlying anorectal malformations(ARM)are not fully established.Circular RNAs(circRNAs)are new born non-coding RNAs,and their role in ARM is unclear.We assumed that rno_circ_0005139 influences apoptosis and proliferation by acting as a miR-324-3p sponge,and downregulating Wnt5a in ARM.AIM To identify the differential expression of circRNAs and mRNAs in a rat ARM model.METHODS Sixty-six pregnant Wistar rats were randomly divided into two groups:ARM group(2-imidazolidinethione-induced)and control groups.Embryos were harvested by cesarean delivery,and anorectal tissue was taken on embryonic days 16(E16),17(E17),19(E19),and 21(E21).RNA sequencing and gene microarray analysis was used to identify differentially expressed circRNAs and mRNAs in the ARM in a rat model.We selected 6 circRNAs and 3 mRNAs in the Wnt signal pathway from the result of the RNA sequencing and gene microarray analysis,and quantitative reverse transcription polymerase chain reaction was performed to evaluate their tissue expression.According to bioinformatics prediction,rno_circ_0005139 acted as a miR-324-3p sponge to regulate the expression of Wnt5a.We chose rno_circ_0005139 and Wnt5a as the final candidates.We tested the function of rno_circ_0005139 and the binding sites between rno_circ_0005139 and miR-324-3p,miR-324-3p and Wnt5a by luciferase assays.Co-transfection of rno_circ_0005139 and miR-324-3p was to verify their functional consistency.RESULTS We identified 38 upregulated and 42 downregulated circRNAs on E17(P2.0).We found that rno_circ_0006880 and rno_circ_0011386 were upregulated,whereas rno_circ_0000436,rno_circ_0005139,rno_circ_0009285,rno_circ_0014367,Wnt5a,Wnt10b,and Wnt2b were downregulated in ARM tissues.According to bioinformatics prediction,rno_circ_0005139 acted as a miR-324-3p sponge to regulate the expression of Wnt5a.We chose rno_circ_0005139 and Wnt5a as the final candidates.Because the rol

Red bayberry(Morella rubra)is an evergreen fruit tree found in southern China whose whole-genome sequence has recently been published.We updated the linkage map of the species by adding 118 SSR markers and the femalespecific marker MrFT2_BD-SEX.The integrated map included eight linkage groups and spanned 491 cM.Eleven sexassociated markers were identified,six of which were located in linkage group 8,in agreement with the previously reported location of the sex-determining region.The MrFT2_BD-SEX marker was genotyped in 203 cultivated accessions.Among the females of the accessions,we found two female-specific alleles,designated W-b(151 bp)and W-d(129 bp).We previously found that‘Dongkui’,a female cultivar,could produce viable pollen(we refer to such plants‘Dongkui-male’)and serve as the paternal parent in crosses.The genotypes of the MrFT2_BD-SEX marker were W-b/Z in‘Biqi’and W-d/Z in‘Dongkui-male’.The progeny of a cross between these parents produced a 3:1 female(W-)to male(ZZ)ratio and the expected 1:1:1:1 ratio of W-b/W-d:W-b/Z:W-d/Z:Z/Z.In addition,the flowering and fruiting phenotypes of all the F1 progeny fit their genotypes.Our results confirm the existence of ZW sex determination and show that the female phenotype is controlled by a single dominant locus(W)in a small genomic region(59 kb and less than 3.3 cM).Furthermore,we have produced a homozygous“super female”(WW)that should produce all-female offspring in the F2 generation,providing a foundation for commercial use and presenting great potential for use in modern breeding programs.

Psychological interventions for depression in children and adolescents:A bibliometric analysis

BACKGROUND Depression has gradually become a common psychological disorder among children and adolescents.Depression in children and adolescents affects their physical and mental development.Psychotherapy is considered to be one of the main treatment options for depressed children and adolescents.However,our understanding of the global performance and progress of psychological interventions for depression in children and adolescents(PIDCA)research is limited.AIM To identify collaborative research networks in this field and explore the current research status and hotspots through bibliometrics.METHODS Articles and reviews related to PIDCA from January 2010 to April 2023 were identified from the Web of Science Core Collection database.The Charticulator website,CiteSpace and VOSviewer software were used to visualize the trends in publications and citations,the collaborative research networks(countries,institutions,and authors),and the current research status and hotspots.RESULTS Until April 16,2023,1482 publications were identified.The number of documents published each year and citations had increased rapidly in this field.The United States had the highest productivity in this field.The most prolific institution was the University of London.Pim Cuijpers was the most prolific author.In the context of research related to PIDCA,both reference co-citation analysis and keywords co-occurrence analysis identified 10 research hotspots,including thirdwave cognitive behavior therapy,short-term psychoanalytic psychotherapy,cognitive behavioral analysis system of psychotherapy,family element in psychotherapy,modular treatment,mobile-health,emotion-regulation-based transdiagnostic intervention program,dementia risk in later life,predictors of the efficacy of psychological intervention,and risks of psychological intervention.CONCLUSION This bibliometric study provides a comprehensive overview of PIDCA from 2010 to present.Psychological intervention characterized as psychological-process-focused,short,family-involved,modular,inte

Background:Yiqi Peiyuan(YQPY)prescription,a composite prescription of traditional Chinese medicine,has been used to prevent or delay the continued deterioration of renal function after acute kidney injury(AKI)in some institutions and has shown considerable efficacy.Objective:This is the first randomized controlled trial to assess efficacy and safety of YQPY for improving short-term prognosis in adult patients with AKI.Design,setting,participants and interventions:This is a prospective,double-blind,multicenter,randomized,and placebo-controlled clinical trial.A total of 144 enrolled participants were randomly allocated to two groups according to a randomization schedule.Participants,caregivers and investigators assessing the outcomes were blinded to group assignment.Patients in the YQPY group received 36 g YQPY granules twice a day for 28 days.Patients in the placebo group received a placebo in the same dose as the YQPY granules.Main outcome measures:The primary outcome was the change in the estimated glomerular filtration rate(eGFR)between baseline and after 4 and 24 weeks of treatment.The secondary outcomes were the change of serum creatinine(Scr)level between baseline and after treatment,and the incidence of endpoint events,defined as eGFR increasing by more than 25%above baseline,eGFR>75 mL/min per1.73 m2or the composite endpoint,which was defined as the sum of patients meeting either of the above criteria.Results:Data from a total of 114 patients(59 in the YQPY group and 55 in the control group)were analyzed.The mean changes in eGFR and Scr in weeks 4 and 24 had no difference between the two groups.In further subgroup analysis(22 in the YQPY group and 31 in the control group),the mean change in eGFR after treatment for 4 weeks was 27.39 mL/min per 1.73 m2in the YQPY group and 5.78 mL/min per1.73 m2in the placebo group,and the mean difference between groups was 21.61 mL/min per 1.73 m2(P<0.001).Thirteen(59.1%)patients in the YQPY group and 5(16.1%)in the placebo group reached the composite endpoints(P=0.002).Du

Nasopharyngeal carcinoma(NPC),a highly invasive and metastatic cancer,is widely prevalent in South China,Southeast Asia,and North Africa.NPC patients with stages I and II are often asymptomatic.Therefore,NPC patients are usually diagnosed with advanced-stage(stages III and IV)at their first visit.Detection of NPC at early stage is the key to improve the survival of patients.Epstein-Barr virus(EBV)plays an essential role in NPC occurrence in endemic areas.EBV-based biomarkers,including EBV antibody in serum and EBV DNA in plasma,have been specifically proposed for NPC early detection.Herein,we summarize recent advances in biomarkers for the early detection of NPC.

Role of phosphorus on the biogeochemical behavior of cadmium in the contaminated soil under leaching and pot experiments

Phosphorus(P)is involved in various biochemical reactions in plant growth,so it is beneficial to plants growing in soils contaminated by metals,including cadmium(Cd).However,few studies have reported on the mechanistic roles of P in mitigating Cd toxicity to ryegrass root,and especially in alleviating the disruption of the mitochondrial function of living cells.In this study,the physiological and biochemical mechanisms associated with ryegrass growth under various Cd and P treatments were investigated using leaching and pot systems.The concentration of Cd in soil leachates showed a significantly positive relationship with redox potential(Pminated soil.Compared to the control(without P addition),the cumulative Cd content was reduced by 53.3%and 64.5%in the soil leachate with exogenous P application(20 mg/L and 80 mg/L),respectively.Notably,application of P decreased the Cd concentrations in the symplastic fractions and increased the Cd concentrations in the apoplastic fractions in root tips,which may help to alleviate Cd stress to the protoplast.Moreover,exogenous P was found to play a positive role in mitochondrial function and Ca^(2+)variation in root cells under Cd stress,which provides novel insights into the mechanisms of exogenous P in alleviating plant Cd injury.

Diabetic kidney disease is a leading cause of end-stage renal disease,making it a global public health concern.The molecular mechanisms underlying diabetic kidney disease have not been elucidated due to its complex pathogenesis.Thus,exploring these mechanisms from new perspectives is the current focus of research concerning diabetic kidney disease.lon channels are important proteins that maintain the physiological functions of cells and organs.Among ion channels,potassium channels stand out,because they are the most common and important channels on eukaryotic cell surfaces and function as the basis for cell excitability.Certain potassium channel abnormalities have been found to be closely related to diabetic kid-ney disease progression and genetic susceptibility,such as KATp,Kca,Kir,and Kv.In this review,we summarized the roles of different types of potassium channels in the occurrence and devel-opment of diabetic kidney disease to discuss whether the development of DKD is due to potas-sium channel dysfunction and present new ideas for the treatment of DKD.

On the Difference Between the Skew-rank of an Oriented Graph and the Rank of Its Underlying Graph

Let G be a simple graph and G~σ be the oriented graph with G as its underlying graph and orientation σ.The rank of the adjacency matrix of G is called the rank of G and is denoted by r(G).The rank of the skew-adjacency matrix of G~σ is called the skew-rank of G~σ and is denoted by sr(G~σ).Let V(G)be the vertex set and E(G) be the edge set of G.The cyclomatic number of G,denoted by c(G),is equal to |E(G)|-|V(G)|+ω(G),where ω(G) is the number of the components of G.It is proved for any oriented graph G~σ that-2c(G)≤sr(G~σ)-r(G)≤2c(G).In this paper,we prove that there is no oriented graph G~σwith sr(G~σ)-r(G)=2c(G)-1,and in addition,there are infinitely many oriented graphs G~σ with connected underlying graphs such that c(G)=k and sr(G~σ)-r(G)=2c(G)-l for every integers k,l satisfying 0 ≤l≤4k and l≠1.

Alzheimer's disease(AD) is a progressive and degenerative neurological disease characterized by the deterioration of cognitive functions. While a definitive cure and optimal medication to impede disease progression are currently unavailable, a plethora of studies have highlighted the potential advantages of exercise rehabilitation for managing this condition. Those studies show that exercise rehabilitation can enhance cognitive function and improve the quality of life for individuals affected by AD. Therefore, exercise rehabilitation has been regarded as one of the most important strategies for managing patients with AD. Herein, we provide a comprehensive analysis of the currently available findings on exercise rehabilitation in patients with AD, with a focus on the exercise types which have shown efficacy when implemented alone or combined with other treatment methods, as well as the potential mechanisms underlying these positive effects. Specifically, we explain how exercise may improve the brain microenvironment and neuronal plasticity. In conclusion, exercise is a cost-effective intervention to enhance cognitive performance and improve quality of life in patients with mild to moderate cognitive dysfunction. Therefore, it can potentially become both a physical activity and a tailored intervention. This review may aid the development of more effective and individualized treatment strategies to address the challenges imposed by this debilitating disease, especially in low-and middle-income countries.
