DnA walker induced “signal on” fluorescence aptasensor strategy for rapid and sensitive detection of extracellular vesicles in gastric cancer

The extracellular vesicles show great potential as a noninvasive biomarker for the early detection of cancer. Hence, there is an urgent requirement to create biosensors that are time-saving, simple, and easily scalable in order to accomplish rapid, sensitive, and quantitative detection of extracellular vesicles. In this study, we present a self-propelled DnA walker powered by endonuclease *** CI, which enables the development of a “signal on” sensing platform for the rapid and highly sensitive detection of extracellular vesicles. The DnA motor employed tracks made of streptavidin magnetic beads, which consisted of substrate strands labeled with fluorescein and motor strands locked by aptamers. The aptamer recognition of the target protein on extracellular vesicles unlocked the motor strand, initiating the DnA motor process. After replacing the optimal buffer solution containing the endonuclease *** I, the motor strands autonomously moved along the streptavidin magnetic beads track, continuously releasing fluorescent molecules and producing detectable fluorescence signals. Under optimal conditions, the detection range was from 2×10～4particles/mL to 2×10～9particles/mL, with a detection limit of 2.9×10～3particles/mL,demonstrating excellent selectivity. This method has demonstrated good selectivity in different tumorderived extracellular vesicles and performs well in complex biological samples. The ability to effectively analyze surface proteins of extracellular vesicles in a short period of time gives our DnA walker a tremendous potential for developing simple and cost-effective clinical diagnostic devices.

Poly(lactic acid) (PLA) is a biodegradable and eco-friendly polymer that is increasingly being incorporated into various applications in contemporary society. However, the limited stability of PLA-based products remains a significant challenge for their broader use in various applications. In this study, poly(L-lactic acid)(PLLA)/poly(D-lactic acid) (PDLA) melt-blown nonwovens were prepared by melt spinning. The structure, thermal properties, thermal stability, biodegradability and crystalline morphology of the melt-blown nonwovens were investigated. DSC and WAXD confirmed the formation of stereocomplex (SC) crystallites in the PLLA matrix. The storage modulus (G '), loss modulus (G ''), and complex viscosity (divided by eta*divided by) of the PLLA/PDLA blend increased with an increase in SC crystallite content. The thermal degradation temperatures of PLLA/PDLA melt-blown nonwovens increased with the incorporation of SC crystallites, and the maximum rate of decomposition increased to 385.5 degrees C, thus enhancing the thermal stability. Compared with neat PLLA melt-blown nonwovens, the hydrophobicity of PLLA/PDLA melt-blown nonwovens was improved, and WCA increased to 139.7 degrees. The SC crystallites were more resistant to degradation by proteinase K compared to neat PLLA. However, the degradation rate of PLLA/PDLA melt-blown nonwovens remained at a high level. This work provides an effective strategy to obtain high-performance PLLA melt-blown nonwovens.

Histone H2B ubiquitination(H2Bub)is positively linked to transcriptional activation,but the genetic programs affected by H2Bub to enhance drought tolerance remain largely ***,we show that OsbZIP27 interacts directly with OsHUB1/2 to regulate drought tolerance in rice by binding to the promoters of OsHAK1 and OsGLn1 to achieve H2Bub and transcriptional ***,mutations in OsbZIP27 reduce transcription of OsHAK1 and OsGLn1,resulting in increased sensitivity to drought ***,loss of OsHUB1 and OsHUB2 function causes hypersensitivity to drought stress,whereas OsHUB2 overexpression enhances drought ***,our results indicate that OsbZIP27 coordinates with OsHUB1/2 to enhance rice drought tolerance by increasing H2Bub and expression of OsHAK1 and OsGLn1.

OBjECTIVE: To propose an automatic acupuncture robot system for performing acupuncture operations. METHODS: The acupuncture robot system consists of three components: automatic acupoint localization, acupuncture manipulations, and De Qi sensation detection. The OptiTrack motion capture system is used to locate acupoints, which are then translated into coordinates in the robot control system. A flexible collaborative robot with an intelligent gripper is then used to perform acupuncture manipulations with high precision. In addition, a De Qi sensation detection system is proposed to evaluate the effect of acupuncture. To verify the stability of the designed acupuncture robot, acupoints' coordinates localized by the acupuncture robot are compared with the Gold Standard labeled by a professional acupuncturist using significant level tests. RESULTS: Through repeated experiments for eight acupoints, the acupuncture robot achieved a positioning error within 3.3 mm, which is within the allowable range of needle extraction and acupoint insertion. During needle insertion, the robot arm followed the prescribed trajectory with a mean deviation distance of 0.02 mm and a deviation angle of less than 0.15°. The results of the lifting thrusting operation in the Xingzhen process show that the mean acupuncture depth error of the designed acupuncture robot is approximately 2 mm, which is within the recommended depth range for the Xingzhen operation. In addition, the average detection accuracy of the De Qi keywords is 94.52%, which meets the requirements of acupuncture effect testing for different dialects. COnCLUSIOn: The proposed acupuncture robot system streamlines the acupuncture process, increases efficiency, and reduces practitioner fatigue, while also allowing for the quantification of acupuncture manipulations and evaluation of therapeutic effects. Tandem solar cells(TSCs)such as III–V/silicon and perovskite/silicon TSCs hold the promise of overcoming the power conversion efficiency(PCE)limit of conventional single-j.nction solar cells[1–3].Direct growth and vacuum bonding have been proposed to fabricate TSCs[4].However,vacuum bonding and direct growth on textured silicon solar cells face various challenges,such as the required flat surfaces and conformational fabrication techniques[5].
