Bioinformatic and Experimental Analyses Reveal That KIF4A Is a Biomarker of Therapeutic Sensitivity and Predicts Prognosis in Cervical Cancer Patients

Objective This study aims to investigate the expression,prognostic value,and function of kinesin superfamily 4A(KIF4A)in cervical cancer.Methods Cervical cancer cell lines(Hela and SiHa)and TCGA data were used for experimental and bioinformatic analyses.Overall survival(OS)and progression free survival(PFS)were compared between patients with high or low KIF4A expression.Copy number variation(CNV)and somatic mutations of patients were visualized and GISTIC 2.0 was used to identify significantly altered sites.The function of KIF4A was also explored based on transcriptome analysis and validated by experimental methods.Chemotherapeutic and immunotherapeutic benefits were inferred using multiple reference databases and algorithms.Results Patients with high KIF4A expression had better OS and PFS.KIF4A could inhibit proliferation and migration and induce G1 arrest of cervical cancer cells.Higher CNV load was observed in patients with low KIF4A expression,while the group with low KIF4A expression displayed more significantly altered sites.A total of 13 genes were found to mutate more in the low KIF4A expression group,including NOTCH1 and PUM1.The analysis revealed that low KIF4A expression may indicate an immune escape phenotype,and patients in this group may benefit more from immunotherapy.With respect to chemotherapy,cisplatin and gemcitabine may respond better in patients with high KIF4A expression,while 5-fluorouracil etc.may be responded better in patients with low KIF4A expression Conclusion KIF4A is a tumor suppressor gene in cervical cancer,and it can be used as a prognostic and therapeutic biomarker in cervical cancer.

Cancer treatment has evolved rapidly due to major advances in tumor immunity research.However,due to the complexity,heterogeneity,and immunosuppressive microenvir-onment of tumors,the overall efficacy of immunotherapy is only 20%.In recent years,nanoparticles have attracted more attention in the field of cancer immunotherapy because of their remarkable advantages in biocompatibility,precise targeting,and controlled drug delivery.However,the clinical application of nanomedicine also faces many problems concerning biological safety,and the synergistic mechanism of nano-drugs with immunity remains to be elucidated.Our study summarizes the functional characteristics and regulatory mechanisms of nanoparticles in the cancer immune microenvironment and how nanoparticles activate and long-term stimulate innate immunity and adaptive immunity.Finally,the current problems and future development trends regarding the application of nanoparticles are fully discussed and prospected to promote the transformation and application of nanomedicine used in cancer treatment.

To address the problems of thermal damage to a workpiece surface caused by the instantaneous high temperature during grinding and the difficulty in monitoring temperature in real time,the temperature field in the case of composite surface grinding by a cup wheel is studied.In order to predict the grinding temperature,considering material removal and grinding force distribution,a nonuniform heat source model with different function distributions in the circumferential and radial directions in the cylindrical coordinate system is first proposed;then,the analytical model is deduced and the numerical model of the temperature field is established based on the heat source model.The validation experiments for grinding temperature field are carried out using a high-definition infrared thermal imager and an artificial thermocouple.Compared to the temperature field based on the uniform heat source model,the results based on the non-uniform heat source model are in better agreement with the actual temperature field,and the temperature prediction error is reduced from approximately 23% to 6%.Thus,the present study provides a more accurate theoretical basis for preventing bums in cup wheel surface grinding.

Numerical simulation study on the hygrothermal performance of building exterior walls under dynamic wind-driven rain condition

Wind-driven rain(WDR)has a significant influence on the hygrothermal performance,durability,and energy consumption of building components.The calculation of WDR loads using semi-empirical models has been incorporated into the boundary conditions of coupled heat and moisture transfer models.However,prior research often relied on fixed WDR absorption ratio,which fail to accurately capture the water absorption characteristics of porous building materials under rainfall scenarios.Therefore,this study aims to investigate the coupled heat and moisture transfer of exterior walls under dynamic WDR boundary conditions,utilizing an empirically obtained WDR absorption ratio model based on field measurements.The developed coupled heat and moisture transfer model is validated against the HAMSTAD project.The findings reveal that the total WDR flux calculated with the dynamic WDR boundary is lower than that obtained with the fixed WDR boundary,with greater disparities observed in orientations experiencing higher WDR loads.The variations in moisture flow significantly impact the surface temperature and relative humidity of the walls,influencing the calculation of cooling and heating loads by different models.Compared to the transient heat transfer model,the coupled heat and moisture transfer model incorporating dynamic WDR boundary exhibits maximum increases of 17.6%and 16.2%in cooling and heating loads,respectively.The dynamic WDR boundary conditions provide more precise numerical values for surface moisture flux,offering valuable insights for the thermal design of building enclosures and load calculations for HVAC systems.

Background：Hand injuries are very common in sports,such as skiing and ball sports.One of the major reasons causing hand and finger deformity is due to ligament and tendon injury.The aim of this study was to investigate if the high-resolution 3T magnetic resonance imaging （MRI） can demonstrate the complex anatomy of the fingers and thumb,especially the tendons and ligaments,and provide the accurate diagnosis of clinically important fingers and thumbs deformity due to ligamentous and tendinous injuries during sport activities.Methods：Sixteen fresh un-embalmed cadaveric hands were harvested from eight cadavers.A total of 20 healthy volunteers＇ hands and 44 patients with fingers or thumb deformity due to sports-related injuries were included in this study.All subjects had MR examination with T1-weighted images and proton density-weighted imaging with fat suppression （PD FS） in axial,coronal,and sagittal plane,respectively.Subsequently,all 16 cadaveric hands were sliced into 2-mm thick slab with a band saw （six in coronal plane,six in sagittal plane,and four in axial plane）.The correlation of anatomic sections and the MRI characteristics of tendons of fingers and the ulnar collateral ligament （UCL） at the metacarpal phalangeal joint （MCPJ） of thumb between 20 healthy volunteers and 44 patients （confirmed by surgery） were analyzed.Results：The normal ligaments and tendons in 16 cadaveric hands and 20 volunteers＇ hands showed uniform low-signal intensity on all the sequences of the MRI.Among 44 patients with tendinous and ligamentous injuries in the fingers or thumb,12 cases with UCL injury at MCPJ of the thumb （Stener lesion =8 and non-Stener lesion=4）,6 cases with the central slip injury,12 cases with terminal tendon injury,and 14 cases with flexor digitorum profundus injury.The ligaments and tendons disruption manifested as increased signal intensity and poor definition,discontinuity,and heterogeneous signal intensity of the involved ligaments and tendons.Conclusions：Sports injury-related fingers and thumb deformity a

Objective Obesity is a global health concern with management strategies encompassing bariatric surgery and anti-obesity drugs;however,concerns regarding complexities and side effects persist,driving research for more effective,low-risk strategies.The promotion of white adipose tissue(WAT)browning has emerged as a promising approach.Moreover,alisol B 23-acetate(AB23A)has demonstrated efficacy in addressing metabolic disorders,suggesting its potential as a therapeutic agent in obesity management.Therefore,in this study,we aimed to investigate the therapeutic potential of AB23A for mitigating obesity by regulating metabolic phenotypes and lipid distribution in mice fed a high-fat diet(HFD).Methods An obesity mouse model was established by administration of an HFD.Glucose and insulin metabolism were assessed via glucose and insulin tolerance tests.Adipocyte size was determined using hematoxylin and eosin staining.The expression of browning markers in WAT was evaluated using Western blotting and quantitative real-time polymerase chain reaction.Metabolic cage monitoring involved the assessment of various parameters,including food and water intake,energy metabolism,respiratory exchange rates,and physical activity.Moreover,oil red O staining was used to evaluate intracellular lipid accumulation.A bioinformatic analysis tool for identifying the molecular mechanisms of traditional Chinese medicine was used to examine AB23A targets and associated signaling pathways.Results AB23A administration significantly reduced the weight of obese mice,decreased the mass of inguinal WAT,epididymal WAT,and perirenal adipose tissue,improved glucose and insulin metabolism,and reduced adipocyte size.Moreover,treatment with AB23A promoted the expression of browning markers in WAT,enhanced overall energy metabolism in mice,and had no discernible effect on food intake,water consumption,or physical activity.In 3T3-L1 cells,AB23A inhibited lipid accumulation,and both AB23A and rapamycin inhibited the mammalian target of rapamycin-sterol regulato

Simulating the total ionizing dose(TID)of an electrical system using transistor-level models can be difficult and expensive,particularly for digital-integrated circuits(ICs).In this study,a method for modeling TID effects in complementary metaloxide semiconductor(CMOS)digital ICs based on the input/output buffer information specification(IBIS)was proposed.The digital IC was first divided into three parts based on its internal structure:the input buffer,output buffer,and functional area.Each of these three parts was separately modeled.Using the IBIS model,the transistor V-I characteristic curves of the buffers were processed,and the physical parameters were extracted and modeled using VHDL-AMS.In the functional area,logic functions were modeled in VHDL according to the data sheet.A golden digital IC model was developed by combining the input buffer,output buffer,and functional area models.Furthermore,the golden ratio was reconstructed based on TID experimental data,enabling the assessment of TID effects on the threshold voltage,carrier mobility,and time series of the digital IC.TID experiments were conducted using a CMOS non-inverting multiplexer,NC7SZ157,and the results were compared with the simulation results,which showed that the relative errors were less than 2%at each dose point.This confirms the practicality and accuracy of the proposed modeling method.The TID effect model for digital ICs developed using this modeling technique includes both the logical function of the IC and changes in electrical properties and functional degradation impacted by TID,which has potential applications in the design of radiation-hardening tolerance in digital ICs.

A composite material comprising a carbon layer and spherical carbon/carbon cloth(C-SC/CC)was fabricated using a hydrothermal-pyrolysis method,employing carbon cloth as the substrate and glucose as the carbon source.The C-SC/CC electrode was evaluated as an electrocatalytic electrode for hydrogen production by electrolysis of Bunsen reaction products.The electrode prepared with 4 g of glucose and annealed at 800°C showed excellent electrocatalytic activity.It requires only a potential of 185 mV(vs.SCE)to achieve a current density of 10 mA/cm2.Furthermore,the electrode demonstrated good stability with a 6%loss in current density after 1000 cycles of scanning from 0.2 V to 1.2 V.These results indicate the potential of the SC/CC electrode as an efficient and durable electrocatalyst for the electrolysis of H2SO4 and HI.

Polarized-sensitive image sensors are a kind of photodetector with great development potential due to their enhanced ability to detect and identify the target objects from the aspect of spatial,spectral and polarized information.Recently,low-dimensional anisotropic materials with inherent anisotropic properties,ultrathin thickness,tunable bandgap and feasible integration with complementary metal oxide semiconductor(CMOS) fabrication processes have attracted great interest for their facilitation of polarized photodetector devices miniaturization.Maximizing the polarized detection performance of low-dimensional materials to satisfy realistic needs stimulates the exploration of modulation of anisotropic properties.In this review,we comprehensively introduce the latest research progress in modulating the optical and optoelectronic anisotropy characteristics of low-dimensional materials.The strategy of anisotropy regulation through crystal structure engineering and coupling system is discussed emphatically.Then,the latest progress in image recognition applications using anisotropic low-dimensional materials is reviewed in detail.Finally,we summarize the challenge and propose future opportunities in the practical application of polarized-sensitive imaging photodetectors based on low-dimensional anisotropic materials.
