MicroRNA-181c provides neuroprotection in an intracerebral hemorrhage model

A tracking and predicting scheme for ping pong robot

We describe a new tracking and predicting scheme applied to a lab-made ping pong robot. The robot has a monocular vision system comprised of a camera and a light. We propose an optimized strategy to calibrate the light center using the least square method. An ellipse fitting method is used to precisely locate the center of ball and shadow on the captured image. After the triangulation of the ball position in the world coordinates, a tracking algorithm based on a Kalman filter outputs an accurate estimation of the flight states including the ball position and velocity. Furthermore, a neural network model is constructed and trained to predict the following flight path. Experimental results show that this scheme can achieve a good predicting precision and success rate of striking an incoming ball. The robot can achieve a success rate of about 80% to return a flight ball of 5 m/s to the opposite court.

HUMAN DRIVING MECHANISM OF REGIONAL LAND USE CHANGE:A CASE STUDY OF KARST MOUNTAIN AREAS OF SOUTHWESTERN CHINA

Based on analysis of the change of land use and related social- economic factors in karst mountain areas of southwestern China, the index system characterizing land use change and its human driving forces in county scale is put forward. Then the relationship of land use change and the driving forces is studied by statistic analysis to identify quantitatively the contribution of human forces and their differences in driving land use change. Moreover, taking the Luodian County as the case, a model simulating the annual change of cultivated land area under the driving of human forces is built. Result of the study will supply reference for the management of the relationship of man and land in karst mountain areas of southern- western China, and accumulate research experience for further study on land use/cover change.

Crystallization and thermal fatigue of ZrTiCuNiBe bulk amorphous alloys heated by laser pulses

The crystallization and thermal fatigue behavior of Zr41Ti14Cu12.5Ni10Be22.5 bulk amorphous alloys heated by laser pulses was investigated. The results showed that crystallization occurred even below the crystallization temperature Tx in a low cycle time. Cracks appearred before or after crystallization of the heated area in the sample. The crystallization degree increased with the heating temperature and cycles. An increase in the fatigue cracks number was also observed. The plastic deformation in the heated area of the sample surface occurred, and became more pronounced with cycles and heating temperature.

Relationship between magnetic resonance imaging features and miRNA gene expression in patients with glioblastoma multiforme

Background Magnetic resonance imaging （MRI） is commonly utilized as part of the diagnostic workup for the clinical diagnosis of glioblastoma multiforme （GBM）, further guiding the clinical treatment of this aggressive cancer. Recent research has shown that microRNAs （miRNAs） may act as oncogenes, or in some cases, tumor suppressor genes that in turn may reflect the genotypic features of GBM. This study aimed to investigate the relationship between MRI features and miRNA gene expression in patients with glioblastoma multiforme. Methods In order to identify the relationship between the radiographic findings of MRI and those identified changes in miRNA gene expression of GBM, we reviewed the MRI images of GBM patients and compared them with the identified miRNA expression profiles utilizing microarray analysis of paired GBM tumor samples. We chose five MRI imaging features：（1） contrast tumor enhanced/necrosis ratio, （2） contrast tumor enhanced/T2 ratio, （3） multiple lesions, （4） hemorrhage, and （5） necrotic volume. The relationship between these five imaging features and miRNA expression was studied using significance analysis of microarrays analysis. Results We found that the expression of miRNAs such as hsa-miR-892b, hsa-miR-892a, and hsa-miR-888 was inversely correlated with an enhanced/necrosis ratio 〉1. The miRNAs such as hsa-miR-95, hsa-miR-498, and hsa-miR-1300 were associated with a contrast tumor enhanced/T2 ratio 〉-1. The miRNAs such as hsa-miR-612, hsa-miR-524-3, and hsa- miR-1282 were associated with multiple lesions identified on MRI and the expression of miR-221 was associated with hemorrhage by GBM. The expression of miR-let-7, including miR-let-7f, miR-let-7i, and miR-let-7f-l＊, was downregulated in the hemorrhage group. The gene expression of miRNAs such as hsa-miR-140-5p, hsa-miR-30e, and hsa-miR-301a was relatively low when compared with the larger necrotic volume group as identified by MRI. In order to solve a linear complementarity problem LCP(M,q),When M is an n×n nonsingular matrix,a class of generalized AOR(GAOR) methods based on GAOR methods for solving linear system,whose special case reduces generalized SOR(GSOR) methods,is proposed. Some sufficient conditions for convergence of GAOR and GSOR methods are given,when the system matrix M is an H-matrix,M-matrix and a strictly or irreducible diagonally dominant *** M is an L-matrix,their monotone convergence are discussed.

The roles of traditional Chinese medicine in gene therapy

The field of gene therapy has been increasingly studied in the last four decades, and its clinical application has become a reality in the last 15 years. Traditional Chinese medicine （TCM）, an important component of complementary and alternative medicine, has evolved over thousands of years with its own unique system of theories, diagnostics and therapies. TCM is well-known for its various roles in preventing and treating infectious and chronic diseases, and its usage in other modern clinical practice. However, whether TCM can be applied alongside gene therapy is a topic that has not been systematically examined. Here we provide an overview of TCM theories in relation to gene therapy. We believe that TCM theories are congruent with some principles of gene therapy. TCM-derived drugs may also act as gene therapy vehicles, therapeutic genes, synergistic therapeutic treatments, and as co-administrated drugs to reduce side effects. We also discuss in this review some possible approaches to combine TCM and gene therapy.

Pristimerin enhances recombinant adeno-associated virus vector-mediated transgene expression in human cell lines in vitro and murine hepatocytes in vivo

OBJECTIVE： In the present study, we systemically evaluated the ability of two bioactive compounds from traditional Chinese medicine, celastrol and pristimerin, to enhance recombinant adeno-associated virus （rAAV） serotype vector-mediated transgene expression both in human cell lines in vitro, and in murine hepatocytes in vivo. METHODS： Human cell lines were infected with rAAV vectors with either mock treatment or treatment with celastrol or pristimerin. The transgene expression, percentage of nuclear translocated viral genomes and the ubiquitination of intracellular proteins were investigated post-treatment. In addition, nonobese diabetic/severe combined immunodeficient gamma （NSG） mice were tail vain-injected with rAAV vectors and co-administered with either dimethyl sulfoxide, celastrol, pristimerin or a positive control, bortezomib. The transgene expression in liver was detected and compared over time. RESULTS： We observed that treatment with pristimerin, at as low as 1 IJmol/L concentration, significantly enhanced rAAV2 vector-mediated transgene expression in vitro, and intraperitoneal co- administration with pristimerin at 4 mg/（kg.d） for 3 d dramatically facilitated viral transduction in murine hepatocytes in vivo. The transduction efficiency of the tyrosine-mutant rAAV2 vectors as well as that of rAAV8 vectors carrying oversized transgene cassette was also augmented significantly by pristimerin. The underlying molecular mechanisms by which pristimerin mediated the observed increase in the transduction efficiency of rAAV vectors include both inhibition of proteasomal degradation of the intracellular proteins and enhanced nuclear translocation of the vector genomes. CONCLUSION： These studies suggest the potential beneficial use of pristimerin and pristimerincontaining herb extract in future liver-targeted gene therapy with rAAV vectors.

Cytotoxic genes from traditional Chinese medicine inhibit tumor growth both in vitro and in vivo

OBJECTIVE： Little effort has been made to study the protein-encoding genes isolated from traditional Chinese medicine（TCM） drugs, and the delivery of these genes into malignant cells through recombinant adeno-associated virus（r AAV） vectors has not been attempted. METHODS： We synthesized the c DNAs of five known cytotoxic proteins isolated from TCM drugs and the FLAG epitope-tagged c DNAs were subcloned into a r AAV plasmid vector. The protein expression was confi rmed by Western blot assay. Various cancer cell lines were transfected with the above plasmids and cell growth was monitored both in vitro and in vivo. The best cytotoxic gene was further packaged into r AAV vectors, under the control of a liver cancer-specifi c promoter. The liver tumor growth was then monitored following intratumor administration of the r AAV ***： The expression plasmids, encoding individual potential cytotoxic genes tagged with FLAG epitope, were successfully generated and sequenced. Among these genes, trichosanthin（TCS） gene yielded the most promising results for the inhibition of cancer cell growth in vitro. The over-expressed TCS functioned as a type I ribosome-inactivating protein, followed by inducing apoptosis that is associated with the Bcl-PARP signaling pathway. Furthermore, intratumor injection of r AAV vectors containing the TCS gene signifi cantly inhibited the growth of human hepatocellular carcinoma tumors in a murine xenograft ***： Our studies suggest that the use of TCM cytotoxic genes is a useful therapeutic strategy for treating human cancers in general, and liver tumors in particular.
