Significance of Diffusion Tensor Imaging of Vastus Medialis Oblique in Recurrent Patellar Dislocation

Background： Numerous studies have investigated the influence of osseous factors on patellofemoral joint instability, but research on the influence of dynamic muscle factors in vivo is still in the exploratory stage. This study aimed to use magnetic resonance imaging （MRI） and diffusion tensor imaging （DTI） to evaluate vastus medialis oblique （VMO） fiber bundles in patients with recurrent patellar dislocation to explore the changes in muscle morphology and function. Methods： This prospective study involved 30 patients （7 males and 23 females; average age, 21.4 ± 3.8 years） clinically diagnosed with recurrent patellar dislocation in Peking University Third Hospital and 30 healthy volunteers matched for age, sex, and body mass index in our medical school between January 2014 and October 2014. None of the patients had a recent history of traumatic patellar dislocation or transient patellar dislocation. All patients underwent conventional MRI and DTI of the knee. The cross-sectional area of the VMO on MRI and the fractional anisotropy （FA）, apparent diffusion coefficient （ADC）, and primary （λ1）, secondary （λ2）, and three-level characteristic （λ3） values on DTI were measured. The independent-samples t-test was used to compare these parameters between the two groups. Results： Compared with the control group, the patient group showed significantly higher FA values （0.39 ± 0.05 vs. 0.33 ± 0.03） and significantly lower ADC （1.51 ± 0.13 vs. 1.58± 0.07）, λ2 （4.96 ±0.13 vs. 5.04 ± 0.07）, and λ3 values （4.44 ± 0.14 vs. 4.58 ± 0.07; t = 5.99, t = -2.58, t = 3.02, and t = -4.88, respectively： all P 〈 0.05）. Cross-sectional VMO area and λ1 values did not differ between the two groups （t = 1.82 and t = 0.22, respectively; both P 〉 0.05）. Conclusions： The functional status of the VMO is closely associated with recurrent patellar dislocation. MRI, especially DTI （FA, ADC, λ2, and ,λ3）, can detect early changes in VMO function and might lhcilitate the noninvasive monitoring of the functional status of the VMO in patients with recurre

Association of atherosclerotic plaque features with collateral circulation status in elderly patients with chronic carotid stenosis

Objective To determine the association of carotid plaque features with collateral circulation status in elderly patients with moderate to severe carotid stenosis.Methods Elderly patients(>60 years)with moderate to severe carotid stenosis were recruited and categorized into good and poor collateral circulation groups,and underwent magnetic resonance imaging and computed tomography imaging.The carotid plaque features including lipid-rich necrotic core,intraplaque hemorrhage,calcification,and fibrous cap rupture(FCR)were evaluated,and maximum wall thickness,normalized wall index(NWI),and luminal stenosis were measured.The association between these variables and collateral circulation status was analyzed.Results Of the 97 patients(78 males,mean age:69.0±6.1 years),19(19.6%)had poor collaterals.The poor collateral group had a significantly higher NWI(93.7%±5.0%vs.89.0%±7.9%,P=0.011),a greater extent of stenosis(80.0%±11.4%vs.75.3%±9.4%,P=0.036)and FCR(84.2%vs.55.1%,P=0.020)compared with good collateral group.Carotid NWI(OR=3.83,95%CI:1.36–10.82,P=0.011)and more FCR(OR=6.77,95%CI:1.35–33.85,P=0.020)were associated with poor collateral circulation after adjustment for the confounding factors.The combination of NWI,FCR,systolic blood pressure,and triglycerides had the highest area-under-the-curve(AUC=0.85)for detection of poor collaterals.Conclusions Carotid plaque features,specifically NWI and FCR,are independently associated with poor collateral circulation,and the combination of carotid plaque features and traditional risk factors has a stronger predictive value for poor collateral circulation than plaque features alone.

Seepage safety monitoring model for an earth rock dam under influence of high-impact typhoons based on particle swarm optimization algorithm

Extreme hydrological events induced by typhoons in reservoir areas have presented severe challenges to the safe operation of hydraulic structures. Based on analysis of the seepage characteristics of an earth rock dam, a novel seepage safety monitoring model was constructed in this study. The nonlinear influence processes of the antecedent reservoir water level and rainfall were assumed to follow normal distributions. The particle swarm optimization （PSO） algorithm was used to optimize the model parameters so as to raise the fitting accuracy. In addition, a mutation factor was introduced to simulate the sudden increase in the piezometric level induced by short-duration heavy rainfall and the possible historical extreme reservoir water level during a typhoon. In order to verify the efficacy of this model, the earth rock dam of the Siminghu Reservoir was used as an example. The piezometric level at the SW1-2 measuring point during Typhoon Fitow in 2013 was fitted with the present model, and a corresponding theoretical expression was established. Comparison of fitting results of the piezometric level obtained from the present statistical model and traditional statistical model with monitored values during the typhoon shows that the present model has a higher fitting accuracy and can simulate the uprush feature of the seepage pressure during the typhoon perfectly.

To the Editor:Quantitative magnetic resonance imaging(MRI)objectively evaluates the degeneration of cartilage in knee osteoarthritis(KOA)patients by measuring changes in proteoglycans,collagen fibers,and water content,thereby facilitating early clinical diagnosis and treatment.[1]Western Ontario and MacMaster Universities(WOMAC)osteoarthritis index score is used to evaluate the symptoms of KOA patients.[2]Presently,few studies have evaluated the correlation between quantitative MRI findings and clinical symptoms(eg.,study performed by Zarins et al[3]).This study explored the relationship between quantitative MRI in T1rho and T2-mapping sequence and WOMAC osteoarthritis index scores of KOA patients.

Quinoxaline-based Polymers with Asymmetric Aromatic Side Chain Enables 16.27% Efficiency for Organic Solar Cells

In recent years, due to the rapid development of high-performance small molecule acceptor (SMA) materials, the researches on p-type electron donor materials for matching with current efficient SMAs have become important. By means of asymmetric strategies to optimize the energy levels and inter/intramolecular interactions of molecules, we designed and synthesized an asymmetric aromatic side chain quinoxaline-based polymer donor TPQ-0F. Meanwhile, we took advantage of F atom which could form noncovalent interaction and strong electron-withdrawing property, to obtain the optimized quinoxaline-based polymer donors TPQ-1F, TPQ-1Fi and TPQ-2F. Eventually, the binary device based on TPQ-2F achieved an efficient power conversion efficiency (PCE) of 16.27%, which attributed to balanced hole/electron mobilities, less charge carrier recombination, and more favorable aggregation morphology. Our work demonstrates the great potential of asymmetric aromatic side chain quinoxaline-based polymer donors on optimizing the morphology of blending films, improving inter/intramolecular interactions, and subtly tuning energy level, finally for more efficient organic solar cells.

Mineralization characteristics of pellets prepared by ultrafine magnetite concentrate:influence on metallurgical property and improving method

The reducing property of pellets prepared by ultrafine magnetite concentrate(UMC)and improving method were revealed.The results show that the reduction degree of UMC pellets is only about 56%compared with that of pellets prepared from ordinary iron ore concentrate with relatively coarse particle size,which is significantly lower than the general reduction degree of about 70%.When the composite binder composed of bentonite and organic binder was added,the reduction degree was significantly increased to 69.66%.The revealed mechanism shows that the reduced pellets with common bentonite have a concentric structure,the oxidation gap between the inner and outer layers is obvious,and the outer dense oxide layer hinders the oxidation and reduction of the inner layer.After adding the composite binder,the organic components significantly improved the internal porosity of the pellets and the aggregation degree of ultrafine iron ore concentrate particles in the granulation process,forming a porous structure.The non-uniform double-layer structure is eliminated,and the increased pores are conducive to the internal diffusion of CO,and finally the reduction degree of pellets is increased to the level equivalent to that of ordinary magnetite pellets.

The key residues of OsTFIIAγ5/Xa5 protein captured by the argininerich TFB domain of TALEs compromising rice susceptibility and bacterial pathogenicity

Xanthomonas bacteria secrete transcription activator-like effector(TALE)proteins into host cells to activate plant disease susceptibility genes to cause disease,and the process is dependent on interaction between bacteria TFB domain of TALEs and host plant basal transcription factor IIA gamma subunit(TFIIAγ).The key domain or residues of plant TFIIAγand core residues of bacteria TFB domain that are indispensable for TFIIAγ-TALEs interaction in the process of TALE-carrying Xanthomonas invasion plants are unknown.Here,we showed that the thirdα-helix domain of OsTFIIAγ5/Xa5,especially the 38th,39th,40th and 42th residues were key sites for capturing by TALEs of Xanthomonas oryzae pv.oryzae(Xoo),the causal agent of rice bacterial blight disease.The latter segment of Xoo TFB domain harboring seventy-two amino acid residues was vital for TALE specific binding with host plant OsTFIIAγ5/Xa5.Substitution of some residues in this core region of TFB domain completely compromised capacity of TALEs capturing rice OsTFIIAγ5/Xa5.The rich and conserved arginine residues in this core region of TFB domain were responsible for TALE-dependent plant susceptibility gene activation and virulence of Xoo.These results provide a potential strategy for improving resistance to TALE-carrying pathogens in plants by site-specific modification of key residues of host plant TFIIAγ.

Protective Mechanism of Cordyceps sinensis Treatment on Acute Kidney Injury-Induced Acute Lung Injury through AMPK/mTOR Signaling Pathway

Objective: To investigate protective effect of Cordyceps sinensis(CS) through autophagyassociated adenosine monophosphate-activated protein kinase(AMPK)/mammalian target of rapamycin(mTOR) signaling pathway in acute kidney injury(AKI)-induced acute lung injury(ALI). Methods: Forty-eight male Sprague-Dawley rats were divided into 4 groups according to a random number table, including the normal saline(NS)-treated sham group(sham group), NS-treated ischemia reperfusion injury(IRI) group(IRI group), and low-(5 g/kg·d) and high-dose(10 g/kg·d) CS-treated IRI groups(CS1 and CS2 groups), 12 rats in each group. Nephrectomy of the right kidney was performed on the IRI rat model that was subjected to 60 min of left renal pedicle occlusion followed by 12, 24, 48, and 72 h of reperfusion. The wet-to-dry(W/D) ratio of lung, levels of serum creatinine(Scr), blood urea nitrogen(BUN), inflammatory cytokines such as interleukin-βand tumor necrosis factor-α, and biomarkers of oxidative stress such as superoxide dismutase, malonaldehyde(MDA) and myeloperoxidase(MPO), were assayed. Histological examinations were conducted to determine damage of tissues in the kidney and lung. The protein expressions of light chain 3 Ⅱ/light chain 3 Ⅰ(LC3-Ⅱ/LC3-Ⅰ), uncoordinated-51-like kinase 1(ULK1), P62, AMPK and mTOR were measured by Western blot and immunohistochemistry, respectively. Results: The renal IRI induced pulmonary injury following AKI,resulting in significant increases in W/D ratio of lung, and the levels of Scr, BUN, inflammatory cytokines, MDA and MPO(P<0.01);all of these were reduced in the CS groups(P<0.05 or P<0.01). Compared with the IRI groups, the expression levels of P62 and mTOR were significantly lower(P<0.05 or P<0.01), while those of LC3-Ⅱ/LC3-Ⅰ, ULK1, and AMPK were significantly higher in the CS2 group(P<0.05 or P<0.01). Conclusion:CS had a potential in treating lung injury following renal IRI through activation of the autophagy-related AMPK/mTOR signaling pathway in AKI-induced ALI.

Aplastic anemia(AA)is a life-threatening disease characterized by bone marrow(BM)failure and pancytopenia.As an important component of the BM microenvironment,endothelial cells(ECs)play a crucial role in supporting hematopoiesis and regulating immunity.However,whether impaired BM ECs are involved in the occurrence of AA and whether repairing BM ECs could improve hematopoiesis and immune status in AA remain unknown.In this study,a classical AA mouse model and VE-cadherin blocking antibody that could antagonize the function of ECs were used to validate the role of BM ECs in the occurrence of AA.Nacetyl-L-cysteine(NAC,a reactive oxygen species scavenger)or exogenous EC infusion was administered to AA mice.Furthermore,the frequency and functions of BM ECs from AA patients and healthy donors were evaluated.BM ECs from AA patients were treated with NAC in vitro,and then the functions of BM ECs were evaluated.We found that BM ECs were significantly decreased and damaged in AA mice.Hematopoietic failure and immune imbalance became more severe when the function of BM ECs was antagonized,whereas NAC or EC infusion improved hematopoietic and immunological status by repairing BM ECs in AA mice.Consistently,BM ECs in AA patients were decreased and dysfunctional.Furthermore,dysfunctional BM ECs in AA patients led to their impaired ability to support hematopoiesis and dysregulate T cell differentiation toward proinflammatory phenotypes,which could be repaired by NAC in vitro.The reactive oxygen species pathway was activated,and hematopoiesis-and immune-related signaling pathways were enriched in BM ECs of AA patients.In conclusion,our data indicate that dysfunctional BM ECs with impaired hematopoiesis-supporting and immunomodulatory abilities are involved in the occurrence of AA,suggesting that repairing dysfunctional BM ECs may be a potential therapeutic approach for AA patients.
