Inhibitory effect of maspinon neovascularization in diabetic retinopathy

BACKGROUND Diabetic retinopathy(DR)is a serious and potentially blinding complication of diabetes *** neovascularization is one of the main pathological features of proliferative DR,and inhibiting retinal neovascularization is a research *** The aim was to evaluate the effect of intravitreal injection of recombinant human maspin on neovascularization in *** An oxygen-induced retinopathy(OIR)mouse model was used to simulate neovascularization in *** born C57BL/6J mice were randomly divided to a normal control group,a maspin injection OIR group,and an OIR *** mice in the maspin injection OIR group were injected with recombinant human maspin in the bilateral vitreous cavity on postnatal day P12,and those in the OIR group were injected with sterile phosphate buffered *** protein expression of vascular endothelial growth factor(VEGF)and hypoxia-inducible factor 1-alpha(HIF-1α)in the retina was measured by western blotting,and the mRNA expression of VEGF and HIF-1αwas measured by real-time polymerase chain *** vascular cell nuclei that broke through the inner limiting membrane(ILM)were counted in haematoxylin-eosin stained retinal *** It was found that the number of vascular cell nuclei breaking through the ILM was 31.8±8.75 in the OIR group,which was significantly more than that in the normal control group(P<0.001).The number of vascular cell nuclei breaking through the ILM was 6.19±2.91 in the maspin injection OIR group,which was significantly less than that in OIR group(P<0.01).The relative protein and mRNA expression of VEGF and HIF-1αwas significantly lower in the retinas in the maspin injection OIR group than in those in the OIR group(P<0.01).CONCLUSION Maspin inhibited neovascularization in DR by modulating the HIF-1α/VEGF pathway,which provides a potential and effective strategy for the treatment of DR.

Metal-organic frameworks with dinuclear metal centers for synergistically boosting CO2 photoreduction

Dinuclear metal synergistic catalysis （DMSC） has been proved powerful in boosting CO2reduction,while this has not been achieved in heterogeneous catalysts with long-range order *** this study,we,by introduction of metal catalytic centers into an indium-based metal-organic framework （MOF）,obtained a series of isostructural In-MOFs,including three MOFs with mononuclear metal catalytic center （M-In）,three MOFs with dinuclear homometallic catalytic centers （MM-In）,and three MOFs with dinuclear heterometallic catalytic centers (MM’-In,M/M’=Co（II）,Ni（II）,Zn（II）).It was found that all these MOFs can serve as catalysts for photochemical CO2reduction to *** with M-In,the catalytic activities of MM-In were obviously enhanced,and that of MM’-In was further significantly *** example,the CO yield of Co Zn-In reaches a record value of 12,700μmol g-1h-1,which is 4.0,8.1,9.4,and 18.6-fold higher than that of Co Co-In,Zn Zn-In,Co-In,and Zn-In,***,the catalytic activities of Co Ni-In and Ni Zn-In also show different fold increase than those of corresponding dinuclear homometallic and mononuclear metallic *** results of control experiments and theoretical calculation reveal that the higher catalytic activities of MM-In over M-In can be ascribed to the DMSC effect,and the higher catalytic activity of MM’-In over MM-In can be ascribed to the strengthened DMSC *** work reveals the relationship between DMSC effect and bimetal centers in CO2reduction,which provides new insights into the DMSC and paves a new way to design highly active catalysts for photochemical CO2reduction.

Small extracellular vesicles derived from human induced pluripotent stem cell-differentiated neural progenitor cells mitigate retinal ganglion cell degeneration in a mouse model of optic nerve injury

Several studies have found that transplantation of neural progenitor cells(NPCs)promotes the survival of injured ***,a poor integration rate and high risk of tumorigenicity after cell transplantation limits their clinical *** extracellular vesicles(sEVs)contain bioactive molecules for neuronal protection and *** studies have shown that stem/progenitor cell-derived sEVs can promote neuronal survival and recovery of neurological function in neurodegenerative eye diseases and other eye *** this study,we intravitreally transplanted sEVs derived from human induced pluripotent stem cells(hiPSCs)and hiPSCs-differentiated NPCs(hiPSC-NPC)in a mouse model of optic nerve *** results show that these intravitreally injected sEVs were ingested by retinal cells,especially those localized in the ganglion cell *** with hiPSC-NPC-derived sEVs mitigated optic nerve crush-induced retinal ganglion cell degeneration,and regulated the retinal microenvironment by inhibiting excessive activation of *** analysis further revealed that hiPSC-NPC derived sEVs transported neuroprotective and anti-inflammatory miRNA cargos to target cells,which had protective effects on RGCs after optic nerve *** findings suggest that sEVs derived from hiPSC-NPC are a promising cell-free therapeutic strategy for optic neuropathy.

Developing a cost-effective and environmentally friendly process for the production of valuable chemicals from abundant herbal biomass receives great attentions in recent ***,taking advantage of the“lignin first”strategy,corn straw is converted to valuable chemicals including lignin monomers,furfural and 5-methoxymethylfurfural via a two steps *** key of this research lies in the development of a green and low-cost catalytic process utilizing magnetic Raney Ni catalyst and high boiling point ethylene *** utilization of neat ethylene glycol as the sole slovent under atmospheric conditions obviates the need for additional additives,thereby facilitating the entire process to be conducted in glass flasks and rendering it highly convenient for scaling *** the initial step,depolymerization of corn straw lignin resulted in a monomer yield of 18.1 wt%.Subsequently,in a dimethyl carbonate system,the carbohydrate component underwent complete conversion in a one-pot process,yielding furfural and 5-methoxymethylfurfural as the primary products with an impressive yield of 47.7%.

Pronounced impact of size and shape effects on creep rupture life of a K439B superalloy combustion chamber casting simulator

The size and shape effect(SSE) of components has become a critical issue for mechanical properties, application reliability, and processing. In this study, the creep rupture life(CRL) of components with different wall thicknesses and positions in a combustion chamber casing simulator made of K439B superalloy was investigated. The intrinsic mechanisms of the SSE were explored from the dendrite structure, volume fraction and size of the γ' phase, and element segregation, etc. It is shown that this casting exhibits a strong SSE of creep rupture life, characterized by a significant difference in the CRL values up to 60% with the variation of wall thickness and position in the casing. In terms of casting technology, the influence of SSE on CRL is actually determined by the cooling rate. The SSE on the creep rupture life originates from the dendrite structure(such as the secondary dendrite arm spacing), volume fraction size of the γ' phase in the dendrite trunk, and elements segregation rate. This work may have implications for the design and application of engineering components with large sizes and complex structures.
