Early-life EV-A71 infection augments allergen-induced airway inflammation in asthma through trained macrophage immunity

Virus-induced asthma is prevalent among children,but its underlying mechanisms are unclear.Accumulated evidence indicates that early-life respiratory virus infection increases susceptibility to allergic asthma.Nonetheless,the relationship between systemic virus infections,such as enterovirus infection,and the ensuing effects on allergic asthma development is unknown.Early-life enterovirus infection was correlated with higher risks of allergic diseases in children.Adult mice exhibited exacerbated mite allergen-induced airway inflammation following recovery from EV-A71 infection in the neonatal period.Bone marrow-derived macrophages(BMDMs)from recovered EV-A71-infected mice showed sustained innate immune memory(trained immunity)that could drive naïve T helper cells toward Th2 and Th17 cell differentiation when in contact with mites.Adoptive transfer of EV-A71-trained BMDMs induced augmented allergic inflammation in naïve recipient mice,which was inhibited by 2-deoxy-D-glucose(2-DG)pretreatment,suggesting that trained macrophages following enterovirus infection are crucial in the progression of allergic asthma later in life.

AIM:To investigate the etiology of esophageal cancer among Taiwan Residents women.METHODS:This is a multi-center,hospital-based,case-control study.Case patients consisted of women who were newly diagnosed and pathology-proven to have esophageal squamous cell carcinoma(ESCC) from three large medical centers(one from Northern and two from Southern Taiwan,respectively)between August 2000 and December 2008.Each ESCC patient was matched with 4 healthy women based on age(within 3 years)and hospital of origin,from the Department of Preventive Medicine in each hospital.A total of 51 case patients and 204 controls,all women,were studied.RESULTS:Frequencies of smokers and drinkers among ESCC patients were 19.6%and 21.6%,respectively,which were significantly higher than smokers(4.4%) and drinkers(4.4%)among controls(OR=4.07,95%CI:1.36-12.16,P=0.01;OR=3.55,95%CI:1.03-12.27,P=0.04).Women who drank an amount of alcohol more than 158 g per week had a 20.58-fold greater risk(95%CI:1.72-245.62,P=0.02)of ESCC than those who never drank alcohol after adjusting for other covariates,although the sample size was small.CONCLUSION:Cigarette smoking and alcohol drinking,especially heavy drinking,are the major risks for developing ESCC in Taiwan Residents women.

Clinical and pathological correlation of the microdeletion of Y chromosome for the 30 patients with azoospermia and severe oligoasthenospermia

Aim: To review the accumulated 30 patients with different area of Y chromosome microdeletions, focusing on their correlation with the clinical and pathological findings. Methods: A total of 334 consecutive infertile men with azoospermia (218 patients) and severe oligoasthenospermia (116 patients) were screened. Complete physical and endocrinological examinations, general chromosome study and multiplex polymerase chain reaction assay to evaluate the Y chromosome microdeletion were performed. Ten patients received testicular biopsy. Then the clinical and pathological findings were analyzed with reference to the areas of Y chromosome microdeletion. Results: There is a decline of the percentage of sperm appearing in semen in the group that the gene deletion region from AZFc to AZFb. The clinical evidence of the impairment (decreased testicular size and elevated serum FSH) is also relevantly aggravated in this group. However, the pathology of testicular biopsy specimen was poorly correlated with the different deletion areas of the Y chromosome, which may be due to the limited number of specimens. Conclusion: The clinical correlation of spermatogenic impairment to the different AZF deletion regions may provide the information for the infertile couples in pre-treatment counseling.

Prognostic potential of the small GTPase Ran and its methylation in hepatocellular carcinoma

Background: Hepatocellular carcinoma(HCC) is a common malignant tumor with high mortality. The prognostic significance of Ran, a member of Ras superfamily, remains unclear in HCC patients. Methods: Based on The Cancer Genome Atlas(TCGA) database and Tumor Immune Estimation Resource(TIMER), we analyzed the correlations among Ran expression, promoter methylation and immune cell infiltration. We also investigated the Ran expression levels in HCC tissues and normal tissues by using quantitative real-time PCR. Results: Ran m RNA expression was significantly increased in HCC tissues compared with the normal tissues( P < 0.001). Time-dependent receiver operating characteristic(ROC) curves showed that Ran expression had predictive value of the 1-, 3-and 5-year overall survival for HCC patients, and the areas under the curves(AUC) were 0.747, 0.634 and 0.704, respectively. Cox regression analysis showed that Ran expression was an independent prognostic factor for HCC patients(HR = 1.492, 95% CI: 1.129-1.971, P = 0.005). We also found a negative relationship between Ran m RNA expression and its promoter methylation( r =-0.36, P < 0.001). High Ran expression and promoter hypomethylation predicted worse overall survival and progression-free survival( P < 0.05) and were involved in the progression of HCC. Ran expression exhibited significant correlations with immune infiltrates and prognostic immune-related genes. Conclusions: The present study provides further insight into the prognosis of HCC, and Ran could serve as a biomarker for predicting the survival of HCC patients.

Tetrasphaera have been recently identified based on the 16S ribosomal RNA(rRNA)gene as among the most abundant polyphosphate-accumulating organisms(PAOs)in global full-scale wastewater treatment plants(WWTPs)with enhanced biological phosphorus removal(EBPR).However,it is unclear how Tetrasphaera PAOs are selectively enriched in the context of the EBPR microbiome.In this study,an EBPR microbiome enriched with Tetrasphaera(accounting for 40%of 16S sequences on day 113)was built using a top-down design approach featuring multicarbon sources and a low dosage of allylthiourea.The microbiome showed enhanced nutrient removal(phosphorus removal~85%and nitrogen removal~80%)and increased phosphorus recovery(up to 23.2 times)compared with the seeding activated sludge from a local full-scale WWTP.The supply of 1 mg·L^(-1)allylthiourea promoted the coselection of Tetrasphaera PAOs and Microlunatus PAOs and sharply reduced the relative abundance of both ammonia oxidizer Nitrosomonas and putative competitors Brevundimonas and Paracoccus,facilitating the establishment of the EBPR microbiome.Based on 16S rRNA gene analysis,a putative novel PAO species,EBPR-ASV0001,was identified with Tetrasphaera japonica as its closest relative.This study provides new knowledge on the establishment of a Tetrasphaera-enriched microbiome facilitated by allylthiourea,which can be further exploited to guide future process upgrading and optimization to achieve and/or enhance simultaneous biological phosphorus and nitrogen removal from high-strength wastewater.

Tunable photoredox-catalyzed chlorosulfonylation and oxysulfonylation ofα-trifluoromethylstyrenes with sulfonyl chloride were facilely achieved by simply manipulating the photocatalyst and solvent.This strategy makes full use of the structural features of sulfonyl chloride to afford diverse bifunctional products ofα-trifluoromethylstyrenes in up to 95%yield with excellent functional group tolerance.

Visualizing Aβdeposits in live young AD model mice with a simple red/near-infrared-fluorescent AIEgen

Precise and early detection ofβ-amyloid(Aβ)deposits in situ and in real time is pivotal to the diagnosis and early intervention of Alzheimer’s disease(AD).Optical imaging stands out to be a promising technique for such a task;however,it still remains a big challenge,due to the lack of high-performance imaging contrast agent.Restricted by poor blood-brain barrier(BBB)penetrability,short-wavelength excitation and emission,as well as the aggregation-caused quenching effect,the widely used goldstandard probes cannot be used for early in-vivo imaging of Aβdeposits.Herein,we integrate the Aβdeposits-favored geometry,amphiphilic and zwitterionic molecular structure,extended D-π-A electronic structure,and 3 D conformation into one molecule,facilely establishing a simple and economic imaging contrast agent that enjoys high specificity and affinity to Aβdeposits,good BBB penetrability,bright red/near-infrared fluorescence,low interference from autofluorescence,aggregation-induced emission(AIE)feature,high signal-to-noise ratio(SNR),and high contrast.In-vitro,ex-vivo,and in-vivo experiments with different strains of mice indicate that AIE-CNPy-AD holds the universality to Aβdeposits identification.Noteworthily,AIE-CNPy-AD is even able to precisely trace the small and sparsely-distributed Aβdeposits in AD model mice as young as 4-month-old APP/PS1 mice,the youngest having Aβdeposits.Moreover,the present probe could clearly reveal the increase and enlargement of Aβdeposits as the mice grow.Therefore,AIE-CNPy-AD might be an ideal alternative for early AD diagnosis and highly reliable monitoring of AD progression.

The extract of Celtis choseniana Nakai alleviates testosterone-in-duced benign prostatic hyperplasia through inhibiting 5αreductase type 2 and the Akt/NF-κB/AR pathway

Benign prostatic hyperplasia(BPH)is a chronic male disease characterized by the enlarged prostate.Celtis choseniana Nakai(C.choseniana)is medicinally used to alleviate pain,gastric disease,and lung abscess.In this study,the effect of C.choseniana extract on BPH was investigated using testosterone-induced rats.Sprague Dawley rats were divided into five groups:control,BPH(testosterone 5 mg·kg^(−1)),Fina(finasteride 2 mg·kg^(−1)),and C.choseniana(50 and 100 mg·kg^(−1)).After four weeks of TP treatment with finasteride or C.choseniana,prostate weights and DHT levels were measured.In addition,the prostates were histopathologically examined and measured for protein kinase B(Akt)/nuclear factor-κB(NF-κB)/AR signaling,proliferation,apoptosis,and autophagy.Pro-state weight and epithelial thickness were reduced in the C.choseniana groups compared with that in the BPH group.The extract of C.choseniana acted as a 5αreductase inhibitor,reducing DHT levels in the prostate.Furthermore,the extract of C.choseniana blocked the activation of p-Akt,nuclear NF-κB activation and reduced the expression of AR and PSA compared with BPH.Moreover,the ex-pression of Bax,PARP-1,and p53 increased,while the expression of bcl-2 decreased.The present study demonstrated that C.choseni-ana extract alleviated testosterone-induced BPH by suppressing 5αreductase and Akt/NF-κB activation,reducing AR signaling and in-ducing apoptosis and autophagy in the prostate.These results suggested that C.choseniana probably contain potential herbal agents to alleviate BPH.

Revisiting the variations of precipitation and water vapour budget over the Tibetan Plateau

Owing to the scarcity of observation data in the western Tibetan Plateau(TP),the knowledge of precipitation changes over the entire plateau based only on the limited data in eastern TP is not reliable.Therefore,the alternative high-resolution precipitation data of the China Meteorological Forcing Dataset(CMFD)are used for the comprehensive analysis of precipitation changes over the whole TP(including western and northern TP)to fill in the lack of understanding of precipitation in the western TP.Compared with observations,CMFD can broadly capture the spatial distributions and identify the temporal variabilities of precipitation over the TP.Results with CMFD data suggested that the annual precipitation over the whole TP did not show a uniform humidification trend in 1979-2018 and featured wetting and drying trends in the northern(NTP)and southern TP(STP),respectively.Additionally,the four seasonal regimes of precipitation over the northern TP(NTP,including most areas of western TP)all experienced a noticeable interdecadal shift around the late 1990s,followed by above-normal precipitation.Except for spring,the seasonal precipitation over the southern TP(STP)showed interannual variations.Spring precipitation over the STP has undergone moistening since the late 1990s,which was consistent with that over the NTP.Four different reanalysis datasets,namely JRA55,MERRA2,ERA5 and CRA40,were used to compare the water vapour budget of each boundary over the TP.The increase in spring precipitation over the NTP and STP was found to be related to the decrease in water vapour outflow from the north boundary.The interdecadal increase in summer precipitation over the NTP was mainly due to the reduction of outflow from the east boundary.Finally,the increase in autumn precipitation was related to the increase in inflow from the west boundary.

Refractive index and temperature optical fiber sensor based on thin core S-taper and spherical structure

A Mach-Zehnder interferometer(MZI)for simultaneously measuring refractive index(RI)and temperature is proposed and verified in this paper.The sensor head is composed of thin core fiber(TCF)S-taper structure and spherical structure.By monitoring two interference dips,experimental results show that the RI sensitivities are-70.392 nm/RIU and-60.08 nm/RIU in the RI range of 1.3384—1.3500,respectively.And the temperature sensitivities are 0.05072 nm/℃and 0.0717 nm/℃in the temperature range of 30—70℃,respectively.The simultaneous measurement of the temperature and external RI is demonstrated based on the sensitive matrix.The sensor also has the advantages of low cost,simple structure and high sensitivity.
