One intrinsic yet critical issue that troubles the field of fluorescence microscopy ever since its introduction is the unmatched resolution in the lateral and axial directions(i.e.,resolution anisotropy),which severely deteriorates the quality,reconstruction,and analysis of 3D volume *** leveraging the natural anisotropy,we present a deep self-learning method termed Self-Net that significantly improves the resolution of axial images by using the lateral images from the same raw dataset as rational *** incorporating unsupervised learning for realistic anisotropic degradation and supervised learning for high-fidelity isotropic recovery,our method can effectively suppress the hallucination with substantially enhanced image quality compared to previously reported *** the experiments,we show that Self-Net can reconstruct high-fidelity isotropic 3D images from organelle to tissue levels via raw images from various microscopy platforms,e.g.,wide-field,laser-scanning,or super-resolution *** the first time,Self-Net enables isotropic whole-brain imaging at a voxel resolution of 0.2×0.2×0.2μm^(3),which addresses the last-mile problem of data quality in single-neuron morphology visualization and reconstruction with minimal effort and ***,Self-Net is a promising approach to overcoming the inherent resolution anisotropy for all classes of 3D fluorescence microscopy.

Nicotinamide adenine dinucleotide(NAD^(+))is indispensable for the anti-aging activity of the sirtuin(SIRT)family *** protein kinase(AMPK)upregulates NAD^(+)synthesis and SIRT activity in a nicotinamide phosphoribosyltransferase(NAMPT)-dependent ***,the molecular mechanisms that affect AMPK-driven NAMPT expression and NAD^(+)/SIRT activation remain *** this study,we tried to identify senescence-associated microRNAs(miRNAs)that negatively regulate the cascade linking AMPK and NAMPT ***-screening experiments showed that the expression of miR-146a increased in senescent cells but decreased following AMPK ***,miR-146a overexpression weakened the metforminmediated upregulation of NAMPT expression,NAD^(+)synthesis,SIRT activity,and senescence protection,whereas treatment with the miR-146a inhibitor reversed this ***,these findings were observed both in vitro and in ***,miR146a directly targeted the 3′-UTR of Nampt mRNA to reduce the expression of *** activators metformin and 5-aminoimidazole-4-carboxamide(AICAR)hindered miR-146a expression at the transcriptional level by promoting IκB kinase(IKK)phosphorylation to attenuate nuclear factor-kappaB(NF-κB)*** findings identified a novel cascade that negatively regulates the NAD^(+)/SIRT pathway by suppressing miR-146a-mediated NAMPT ***,our results showed that miR-146a impedes the anti-aging effect of *** mutual inhibitory relationship between miR-146a and AMPK enriches our understanding of the molecular connections between AMPK and SIRT and provides new insight into miRNA-mediated NAD^(+)/SIRT regulation and an intervention point for the prevention of aging and age-related diseases.

BACKGROUND Thymolipoma is a rare benign tumor arising from the anterior mediastinal thymus and is composed of mature fatty tissue and interspersed nonneoplastic thymic tissue. This tumor accounts for only a small percentage of mediastinal masses, and the majority of them are asymptomatic and found incidentally. To date, fewer than 200 cases have been published in the world literature, of which most excised tumors weighed less than 0.5 kg and the largest weighed 6 *** SUMMARY A 23-year-old man presented with a complaint of progressive breathlessness for 6mo. His forced vital capacity was only 23.6% of the predicted capacity, and his arterial partial pressure of oxygen and carbon dioxide were 51 and 60 mmHg,respectively, without oxygen inhalation. Chest computed tomography revealed a large fat-containing mass in the anterior mediastinum that measured 26 cm × 20cm × 30 cm in size and occupied most of the thoracic cavity. Percutaneous mass biopsy revealed only thymic tissue without signs of malignancy. A right posterolateral thoracotomy was successfully performed to remove the tumor along with the capsule, and the excised tumor weighed 7.5 kg, which to our knowledge, was the largest surgically removed tumor of thymic origin. Postoperatively, the patient’s shortness of breath was resolved, and the histopathological diagnosis was thymolipoma. No signs of recurrence were observed at the 6-mo *** Giant thymolipoma causing respiratory failure is rare and dangerous. Despite the high risks, surgical resection is feasible and effective.

Cerenkov luminescence imaging(CLI) has been widely investigated for biological imaging. However, the luminescence generated from Cerenkov effect is relatively weak and has poor penetration ability in biological *** limitations consequently hindered the clinical translation of CLI. In this study, we proposed an in vitro experimental study for the demonstration of quantum dots(QDs) configurations affected by the improvement of the signal intensity of CLI. Results revealed that the optimal concentrations were 0.1 mg/mL and 0.25 mg/mL for the studied CdSe/ZnS QDs with fluorescence emission peaks of 580 nm and 660 nm, respectively. The detected optical signal intensity with long-wavelength emission QDs were stronger than those with short-wavelength emission *** study illustrates an experiment to study the effects of concentrations and fluorescence emission peaks of QDs on an enhanced optical signal for the external detection of CLI.

BACKGROUND Commonly used cleaning brushes in the reprocessing of flexible endoscopes often cause damage within the working *** To develop a spray flushing system to achieving effective cleaning of the working channels while minimizing *** This prospective study included 60 used endoscopes and 60 Teflon tubes randomly divided into a control group(n=30)and an experimental group(n=30).The material of Teflon tubes was the same as that of the endoscope working *** in the control group were manually cleaned using traditional cleaning brushes,while those in the experimental group were cleaned using the newly developed spray flushing *** levels,cleanliness,and microbiological testing of the working channels were ***,Teflon tubes in the control group underwent 500 passes with a cleaning brush,while those in the experimental group were subjected to the spray flushing system,and channel damage was *** The ATP levels(RLU)in the two groups were 32.5(13-66)and 26(16-40),respectively(P>0.05).Cleanliness scores were 1.5(1-2)and 1(1-2),respectively(P>0.05).Debris was found in 73.3%of the control group,which was significantly higher than 46.7%in the experimental group(P<0.05).Microbiological tests for both groups yielded negative *** tube damage in the control group was rated at 4(4-5.25),which was significantly higher than in the experimental group 4(3-4)(P<0.01).CONCLUSION The spray flushing system demonstrated superior efficacy in removing debris and resulted in less damage to the endoscope working channels compared with traditional cleaning brushes.

A novel and fast three-dimensional reconstruction method for a Compton camera and its performance in radionuclide imaging is proposed and analyzed in this study. The conical surface sampling back-projection method with scattering angle correction(CSS-BP-SC) can quickly perform the back-projection process of the Compton cone and can be used to precompute the list-mode maximum likelihood expectation maximization(LM-MLEM). A dedicated parallel architecture was designed for the graphics processing unit acceleration of the back-projection and iteration stage of the CSS-BP-SC-based LM-MLEM. The imaging results of the two-point source Monte Carlo(MC) simulation demonstrate that by analyzing the full width at half maximum along the three coordinate axes, the CSS-BP-SC-based LM-MLEM can obtain imaging results comparable to those of the traditional reconstruction algorithm, that is, the simple back-projection-based LM-MLEM. The imaging results of the mouse phantom MC simulation and experiment demonstrate that the reconstruction results obtained by the proposed method sufficiently coincide with the set radioactivity distribution, and the speed increased by more than 664 times compared to the traditional reconstruction algorithm in the mouse phantom experiment. The proposed method will further advance the imaging applications of Compton cameras.

Simultaneous Determination of Six UV-Filters in Cosmetics by HPLC

A reversed-phase high performance liquid chromatographic method with diode-array detector was established to simultaneously determine the six UV-filters, isopentyl-4-methoxycinnamate, ethylhexyl methoxycinnamate, ethylhexyl salicylate, homosalate, butyloctyl salicylate and ethylhexyl triazone. They were separated on a Kromasil C18 column, with gradient elution by methanol and water, and detected by *** results demonstrated good linear relationship in proper mass concentration range. The limits of quantitation(LOQ) were between 0.5 and 1.25 mg/L. The recoveries of the six compounds ranged from 98.3% to 101.0% at two inrements with the relative standard deviation(RSD) no more than 1.5%.
