In the present study,flavor profiles of Chinese spiced beef in the cooking process were comparatively analyzed by electronic nose,gas chromatography–mass spectrometry(GC–MS)with a thermal desorption system(TDS),and solid-phase microextraction(SPME).A total of 82 volatile compounds were identified,and 3-methyl-butanal,pentanal,hexanal,-xylene,heptanal,limonene,terpinene,octanal,linalool,4-terpinenol,-terpineol,and(E)-anethole were identified as the characteristic flavor compounds in Chinese spiced *** in the content of volatile components produced by different cooking processes was *** general,a cooking time of 4 h resulted in optimal flavor quality and *** indicated that the electronic nose could profile and rapidly distinguish variation among different cooking *** volatile profiling by TDS-GC–MS and responses from the electronic nose,in combination with multivariate statistical analysis,are a promising tool for control the cooking process of spiced beef.

Dear Editor,The moderate level of reactive oxygen species(ROS)contributes to cellular functions such as proliferation,differentiation,and infection resistance,but the excessive level of ROS causes oxidative damage which underlies the basic mechanism for aging and geriatric diseases.1 Therefore,precisely managing cellular ROS levels,meaning to keep redox homeostasis properly,stands as an aim for health and ***,this aim is challenging,as it asks not only to restrain excessive ROS accumulation at the right time and right place but also to guarantee proper ROS levels for fitting physiological *** ways for eliminating ROS put more attention on the efficiency and identified a variety of drugs and enzymes with antioxidant properties,known as enzymatic and non-enzymatic ROS scavengers,***,how to accurately manage the integrative effect of ROS scavengers in vivo remains a ***,the effectiveness of non-enzymatic antioxidants in vivo is controversial,explained mostly by the uncertainty to reach a given organ with the appropriate concentration or to work there persistently.2 As to enzymatic ROS scavengers,although their overexpression in a given place can be achieved,this mode is often unwieldy or harmful to intrinsic physiologic functions in cases.3 For this reason,we attempt to develop new ways for in vivo antioxidation,for establishing a high-quality balance between efficiency and *** the consequence,we innovated a gene expression system that can limit the excessive ROS accumulation but keep the physiological level of ROS.

Sludge is the by-product of wastewater treatment process. Multisource sludge can be defined as sludge from different sources. Based on the sludge properties of five typical cities in the Yangtze River basin, including Jiujiang, Wuhu, Lu’an, Zhenjiang and Wuhan, this study investigated and summarized the characteristic variations and distribution differences of multiple indicators and substances from municipal sludge, dredged sludge, and river and lake sediments. The results demonstrated pH of multisource sludge was relatively stable in the neutral range. Organic matter and water content among municipal sludge were high and varied considerably between different wastewater treatment plants. Dredged sludge had an obviously higher sand content and wider particle distribution, which could be considered for graded utilization depending on its size. The nutrients composition of river and lake sediments was usually stable and special, with lower nitrogen and phosphorus content but higher potassium levels. The sources of heavy metals and persistent organic pollutants in multisource sludge were correlated, generally much higher among municipal sludge than dredged sludge and river and lake sediments, which were the most important limitation for final land utilization. Despite various properties of multisource sludge, the final fate and destination have some overall similarities, which need to be supplemented and improved by standards and laws. The study provided a preliminary analysis of suitable technical routes for municipal sludge, dredged sludge, river and lake sediments based on their different characteristics respectively, which was of great significance for multisource sludge co-treatment and disposal in the future of China.

Comparisons of nozzle orifice processing methods using synchrotron X-ray micro-tomography

Based on the high flux synchrotron X-ray of the Shanghai Synchrotron Radiation Facility (SSRF), high precision 3D digital models of diesel nozzle tips have been established by X-ray micro-tomography technology, which reveal the internal surfaces and structures of orifices. To analyze the machining precision and characteristics of orifice processing methods, an ap- proach is presented based on the parameters of the internal structures of nozzle orifices, including the nozzle diameter, the orifice inner surface waviness, the eccentricity distance and the angle between orifices. Using this approach, two kinds of nozzle orifice processing methods, computerized numerical control drilling and electric discharge machining, have been studied and compared. The results show that this approach enables a simple, direct, and comprehensive contrastive analysis of nozzle orifice processing methods. When processing a single orifice, the electric discharge machining method has obvious advantages. However, when there are multiple orifices, the error levels of the two methods are similar in relation to the symmetry of distribution of the orifices.

Testicular nuclear receptor 4(TR4),also known as NR2C2(nuclear receptor subfamily 2,group C,member 2),is a transcriptional factor and a member of the nuclear receptor ***4 was initially cloned from human and rat hypothalamus,prostate,and testes *** almost two decades,its specific tissue distribution,genomic organization,and chromosomal assignment have been well investigated in humans and ***,it has been very difficult to study TR4's physiological functions due to a lack of specific *** knock-out animal techniques provide an alternative approach for defining the biological functions of *** vivo studies of TR4 gene knockout mice(TR4-/-) found that they display severe spinal curvature,subfertility,premature aging,and prostate prostatic intraepithelial neoplasia(PIN) *** modulators,downstream target gene regulation,feedback mechanisms,and differential modulation mediated by the recruitment of other nuclear receptors and coregulators have been identified in studies using the TR4-/-*** the establishment of a tissue-specific TR4-/-mouse model,research on TR4 will be more convenient in the future.

To date, many efforts have been made to improve the performance of paintable carbon-based （PC-based） perovskite solar cells （PSCs）. Though great progress has been achieved, their power conversion efficiencies are still relatively low compared with hole-transport-materials-based PSCs. General research on influencing factors of performance in PC-based PSCs is still insufficient. In this work, PC-based PSCs were fabricated in ambient air and four groups of controlled experi- ments were performed in which the PbI2 layers were prepared with or without antisolvent extraction treatment. These four groups of experiments were designed to find out the effect of different influencing factors on PC-based PSCs performance, for example, PbI2 residual, the surface morphology of the perovskite film, the surface roughness of the perovskite film, and the contact status of the perovskite/carbon electrode interface. With a systematic analysis, we demonstrated that the contact status of the perovskite/carbon electrode interface played a vital role in PC-based PSCs, and a fiat, smooth perovskite surface could help to improve this contact status significantly. Besides, on the precondition of a poor contact interface, no PbI2 residual and a good surface morphology only brought limited benefits to the performances of PC-based PSCs.

Neurons in the primary auditory area(AUDp)innervate multiple brain regions with long-range projections while receiving informative inputs for diverse ***,the brain-wide connections of these neurons have not been comprehensively ***,we simultaneously applied virus-based anterograde and retrograde tracing,labeled the connections of excitatory and inhibitory neurons in the mouse AUDp,and acquired whole-brain information using a dual-channel fuorescence micro-optical sectioning tomography *** results showed that the two types of neurons received inputs with similar patterns but sent heterogeneous projections to downstream *** the isocortex,functionally different areas consistently sent feedback-dominated projections to these neurons,with concomitant laterallydominated projections from the sensory and limbic cortices to inhibitory *** subcortical regions,the dorsal and medial parts of the non-lemniscal auditory thalamus(AT)were reciprocally connected to the AUDp,while the ventral part contained the most fibers of passage from the excitatory neurons and barely sent projections back,indicating the regional heterogeneity of the AUDp-AT *** results reveal details of the whole-brain network and provide new insights for further physiological and functional studies of the AUDp.

AIM： To identify the potentially pathogenic gene variants that contributes to the etiology of strabismus. METHODS： A Chinese pedigree with strabismus was collected and the exomes of two affected individuals were sequenced using the next-generation sequencing technology. The resulting variants from exome sequencing were filtered by subsequent bioinformatics methods and the candidate mutation was verified as heterozygous in the affected proposita and her mother by sanger sequencing. RESULTS： Whole exome sequencing and filtering identified a nonsynonymous mutation Co434G-T transition in paired box 3 （PAX3） in the two affected individuals, which were predicted to be deleterious by more than 4 bioinformatics programs. This altered amino acid residue was located in the conserved PAX domain of PAX3. This gene encodes a member of the PAX family of transcription factors, which play critical roles during fetal development. Mutations in PAX3 were associated with Waardenburg syndrome with strabismus. CONCLUSION： Our results report that the c.434G-T mutation （p.R145L） in PAX3 may contribute to strabismus, expanding our understanding of the causally relevant genes for this disorder.

Effect of thienorphine on intestinal transit and isolated guinea-pig ileum contraction

AIM: To evaluate the effect of thienorphine on small intestinal transit in vivo and on guinea-pig ileum (GPI) contraction in vitro . METHODS: The effects of thienorphine on intestinal transit were examined in mice and in isolated GPI. Buprenorphine and morphine served as controls. The distance traveled by the head of the charchol and the total length of the intestine were measured in vivo . Gastrointestinal transit was expressed as a percentage of the distance traveled by the head of the marker relative to the total length of the small intestine. The isolated GPI preparations were connected to an isotonic force transducer and equilibrated for at least 1 h before exposure to drugs. Acetylcholine was used for muscle stimulation. RESULTS: Thienorphine (0.005-1.0 mg/kg, ig ) or bu-prenorphine (0.005-1.0 mg/kg, sc ) dose-dependently significantly inhibited gut transit compared with saline. Thienorphine inhibited gut transit less than buprenorphine. The maximum inhibition by thienorphine on the intestinal transit was 50%-60%, whereas the maximum inhibition by morphine on gut transit was about 100%. Thienorphine also exhibited less inhibition on acetylcholine-induced contraction of GPI, with a maximum inhibition of 65%, compared with 93% inhibition by buprenorphine and 100% inhibition by morphine. Thienorphine induced a concentration-dependent decrease in the basal tonus of spontaneous movement of the GPI, the effect of which was weaker than that with buprenorphine. The duration of the effect of thienorphine on the GPI was longer than that with buprenorphine. CONCLUSION: Thienorphine had less influence, but a longer duration of action on GPI contraction and moderately inhibited intestinal transit.
