Next year will mark the 100th anniversary of the Michaelis- Menten equation [1], one of the most important and well known models in enzymology. Remarkable progress in en- zymology over the past hundred years has not only provided deep insight into biological processes, but also has dramati- cally changed our lives. However, because of limitations in available techniques, enzymologists primarily investigate the in vitro properties and catalytic mechanisms of individ- ual enzymes. Thus, they often fail to address a fundamental scientific question： how a^e biological processes, most of which are chemical reactions catalyzed by enzymes, coor- dinated in space and time to produce a livin~ organism ｜2]？

Objective: To investigate the effect and potential mechanism of dihydromyricetin(Dmy) on H9C2 cell proliferation, apoptosis, and autophagy. Methods: H9C2 cells were randomly divided into 7 groups, namely control, model, EV(empty p CDH-CMV-MCS-EF1-Cop GFP-T2A-Puro vector), IV(circHIPK3 interference), Dmy(50 μmol/L), Dmy+IV, and Dmy+EV groups. Cell proliferation and apoptosis were detected by cell counting kit-8 assay and flow cytometry, respectivley. Western blot was used to evaluate the levels of light chain 3 Ⅱ/Ⅰ(LC3Ⅱ/Ⅰ), phospho-phosphoinositide 3-kinase(p-PI3K), protein kinase B(p-AKT), and phospho-mammalian target of rapamycin(p-mTOR). The level of circHIPK3 was determined using reverse transcriptase polymerase chain reaction. Electron microscopy was used to observe autophagosomes in H9C2 cells. Results: Compared to H9C2 cells, the expression of circHIPK in H9C2 hypoxia model cells increased significantly(P<0.05). Compared to the control group, the cell apoptosis and autophagosomes increased, cell proliferation rate decreased significantly, and the expression of LC3Ⅱ/Ⅰ significantly increased(all P<0.05). Compared to the model group, the rate of apoptosis and autophagosomes in IV, Dmy, and Dmy+IV group decreased, the cell proliferation rate increased, and the expression of LC3Ⅱ/Ⅰ decreased significantly(all P<0.05). Compared to the control group, the expressions of p-PI3K, p-AKT, and p-mTOR in the model group significantly reduced(P<0.05), whereas after treatment with Dmy and sh-circHIPK3, the above situation was reversed(P<0.05). Conclusion: Dmy plays a protective role in H9C2 cells by inhibiting circHIPK expression and cell apoptosis and autophagy, and the mechanism may be related to PI3K/AKT/mTOR pathway.

Nitrogen (N) leaching in vegetable fields from December 2002 to May 2003 with equal dressings of total N for asequential rotation of Chinese flat cabbage (Brassica chinensis L. var. rosularis) and lettuce (Lactuca sativa L.) in asuburban major vegetable production base of Shanghai were examined using the lysimeter method to provide a scientificbasis for rational utilization of nitrogen fertilizers so as to prevent nitrogen pollution of water resources. Results showedthat leached N consisted mainly of nitrate N, which accounted for up to more than 90% of the total N loss and couldcontribute to groundwater pollution. Data also showed that by partly substituting chemical N (30%) in a basal dressingwith equivalent N of refined organic fertilizer in the Chinese flat cabbage field, 64.5% of the leached nitrate N was reduced,while in the lettuce (Lactuca sativa L.) field, substituting 1/2 of the chemical N in a basal dressing and 1/3 of the chemicalN in a top dressing with equivalent N of refined organic fertilizers reduced 46.6% of the leached nitrate N. In the two-year sequential rotation system of Chinese flat cabbage and lettuce, nitrate-N leaching in the treatment with the highestamount of chemical fertilizer was up to 46.55 kg ha-1, while treatment plots with the highest amount of organic fertilizerhad only 17.58 kg ha-1. Thus, partly substituting refined organic fertilizer for chemical nitrogen in the first two seasonshas a great advantage of reducing nitrate-N leaching.

Programmed cell death 1（PD-1）/programmed cell death 1 ligand（PD-L1） blockade has shown promising effects in cancer immunotherapy. Removing the so-called ＂brakes＂ on T cell immune responses by blocking the PD-1/PDL1 check point should boost anti-tumor immunity and provide durable tumor regression for cancer ***, 30%–60% of patients show no response to PD-1/PD-L1 blockade. Thus, it is urgent to explore the underlying resistance mechanisms to improve sensitivity to anti-PD-1/PD-L1 therapy. We propose that the mechanisms promoting resistance mainly include T cell exclusion or exhaustion at the tumor site,immunosuppressive factors in the tumor microenvironment（TME）, and a range of tumor-intrinsic factors. This review highlights the power of studying the cellular and molecular mechanisms of resistance to improve the rational design of combination therapeutic strategies that can be translated to the clinic. Here, we briefly discuss the development of PD-1/PD-L1 blockade agents and focus on the current issues and future prospects for potential combinatorial therapeutic strategies that include anti-PD-1/PD-L1 therapy, based upon the available preclinical and clinical data.

The pathogenic cascade of Alzheimer’s disease(AD)characterized by amyloid-β protein accumulation is still poorly understood,partially owing to the limitations of relevant models without in vivo neural tissue microenvironment to recapitulate cell-cell *** better mimic neural tissue microenvironment,three-dimensional(3D)core-shell AD model constructs containing human neural progenitor cells(NSCs)with 2% matrigel as core bioink and 2% alginate as shell bioink have been bioprinted by a co-axial bioprinter,with a suitable shell thickness for nutrient exchange and barrier-free cell interaction *** constructs exhibit cell self-clustering and-assembling properties and engineered reproducibility with long-term cell viability and self-renewal,and a higher differentiation level compared to 2D and 3D MIX *** different effects of 3D bioprinted,2D,and MIX microenvironments on the growth of NSCs are mainly related to biosynthesis of amino acids and glyoxylate and dicarboxylate metabolism on day 2 and ribosome,biosynthesis of amino acids and proteasome on day ***,the model constructs demonstrated Aβ aggregation and higher expression of Aβ and tau isoform genes compared to 2D and MIX *** model constructs will provide a promising strategy to facilitate the development of a 3D in vitro AD model for neurodegeneration research.

Shewanella putrefaciens is one of the common spoilage microorganisms in aquatic products,and its extracellular protease is the main cause of seafood *** characteristics of the extracellular protease produced by *** YZ08 and its hydrolytic properties on fish proteins were *** purification,an extracellular protease of 18.78 kDa was obtained and identified as a metalloprotease of the M23 *** gene encoding this enzyme was cloned and purified by expression in *** to obtain a recombinant ***08 protease showed optimum enzymatic activity at 40℃ and pH 9.0 and was stable at pH 7.0-10.0 and low temperature(<40℃).The kinetic parameters and thermal inactivation parameters of the enzyme were *** YZ08 protease can degrade fish myofibrillar and sarcoplasmic proteins,breaking them down into small particles and low molecular *** three-dimensional structure of the protease was simulated by homology modeling and revealed a correlation between the active site in the protein and substrate *** work provides a foundation for characterizing the properties of *** YZ08 protease and for controlling the quality and safety of seafood.
