TANK-binding kinase 1(TBK1)is an important enzyme in the regulation of cellular antiviral ***1 regulates the activity of the interferon regulatory factors IRF3 and IRF7,thereby playing a key role in type I interferon(IFN)signaling *** structure of TBK1 consists of an N-terminal kinase domain,a middle ubiquitin-like domain(ULD),and a C-terminal elongated helical *** has been reported that the ULD of TBK1 regulates kinase activity,playing an important role in signaling and mediating interactions with other molecules in the IFN *** this study,we present the crystal structure of the ULD of human TBK1 and identify several con-served residues by multiple sequence *** found that a hydrophobic patch in TBK1,containing residues Leu316,Ile353,and Val382,corresponding to the“Ile44 hydrophobic patch”observed in ubiquitin,was conserved in TBK1,IκB kinase epsilon(IKKε/IKKi),IκB kinase alpha(IKKα),and IκB kinase beta(IKKβ).In com-parison with the structure of the IKKβULD domain of Xenopus laevis,we speculate that the Ile44 hydrophobic patch of TBK1 is present in an intramolecular binding surface between ULD and the C-terminal elongated *** varying surface charge distributions in the ULD domains of IKK and IKK-related kinases may be relevant to their specificity for specific partners.

Type I interferon（IFN） can block virus replication by stimulating the expression of various antiviral genes termed interferon-stimulated genes（ISG）, including ISG56 family members（also known as IFN-induced tetratricopeptide repeats, IFITs）. The human ISG56 family includes 4 members, ISG54（IFIT2）, ISG56（IFIT1）, ISG58（IFIT5） and ISG60（IFIT3）, which have overlapping functions but distinct induction patterns. ISG56 family members not only have antiviral activities, but also can regulate cellular factors expression. They were considered to be translation inhibitors at the early stage. However, these hypotheses are not enough to explain the complex behaviors of ISG56 family proteins either for antivirus or cellular regulation. We proposed a new mechanism based on crystal structural studies of ISG54 and ISG58. The structures suggest that they belong to a new type of nucleotide-binding protein. The C-terminals of both ISG54 and ISG58 fold into super-helical structures which have extensively positively-charged amino acids. Mutagenesis studies show that the nucleotide binding ability is related to their antiviral activities. EMSA results show that ISG58 and ISG54 recognize nucleotides with high selectivity, probably because the length and inner radiuses of their helical channels are different. Based on these results, we suggest that ISG56 family proteins can recognize special nucleotide sequence, so they can selectively regulate cellular factors’ expression and inhibit virus replication.

在101.3 k Pa恒定压力下,采用改进的Rose汽液平衡釜测定了甲醇-DMM3(聚甲氧基二甲醚,聚合度为n,即DMMn)二元体系汽液平衡数据,并对汽液平衡数据进行热力学一致性检验,结果表明所测定数据符合Gibbs-Duhenm的热力学一致性。用Aspen Plus v7.1计算机软件,分别对Wilson、NRTL、UNIQUAC活度系数模型进行关联,由最大似然法对目标函数进行优化,回归出相应的二元交互作用参数。将关联结果与实验结果相比较,得到关联值与实验值的温度和汽相组成的平均绝对偏差,分别小于0.65 K和0.0065。为化工数据库增添了内容,也为含甲醇、DMM3体系的工程设计和进一步深入研究奠定了基础。

针对DV-Hop定位算法以平均跳段距离代替实际直线距离而导致定位误差较大这一问题,提出了一种改进的DV-Hop定位算法。在改进算法中,锚节点通过实际距离和估计距离的误差来修正每跳平均距离。改进DV-Hop节点坐标计算方法,摒弃传统的三边定位算法而采用新的二维双曲线定位算法计算节点坐标。最后求得包含误差修正值的最终节点坐标,它更接近实际节点坐标。仿真结果表明,与传统DV-Hop算法相比,改进算法在不需要增加节点的硬件开销的基础上能更有效地提高定位精度。

目的探讨双层光谱探测器CT能谱图像在宫颈癌检出与分期中的应用价值。方法搜集首诊宫颈癌患者48例,均行光谱CT和盆腔MRI检查,有完整组织病理学资料。由两名高年资影像学医师以双盲法分析光谱CT 40 keV及MRI图像,记录是否为阳性病灶,对阳性病灶多平面观察肿瘤部位、大小及范围,判断有无宫旁浸润、阴道浸润及淋巴结转移,并评价分期。采用χ^(2)检验分析比较上述各组参数之间的差异。选择动、静脉两期病灶强化最明显区,勾画病灶和髂动脉ROI,记录碘密度(IC),并计算标准化碘密度(NIC),采用独立样本t检验,比较宫颈癌早期(≤ⅡA期)和进展期(≥ⅡB期)之间两项定量参数的差异;绘制ROC曲线,分析IC、NIC诊断宫颈癌早期及进展期的效能。结果MRI、光谱CT对宫颈癌的检出率均高于常规CT,分别为91.67%(44/48)、81.25%(39/48)、60.42%(29/48),差异有统计学意义。光谱CT对宫颈癌ⅡB期及以上诊断符合率(100%)高于MRI(95.65%),差异无统计学意义。MRI对宫颈癌ⅠB1期及以下、ⅠB2~ⅡA期诊断符合率(80.00%、90.00%)高于光谱CT(20.00%、75.00%),差异无统计学意义。光谱CT诊断宫颈癌淋巴结转移的敏感度、特异度和准确度(90.00%、100%、94.87%)高于MRI(65.00%、87.50%、77.27%),差异有统计学意义。宫颈癌进展期动、静脉两期的IC、NIC均高于早期,动脉期IC、NIC差异均有统计学意义。ROC曲线显示,动脉期NIC诊断宫颈癌早期及进展期的曲线下面积为0.718(95%CI:0.552~0.850),高于动脉期IC的曲线下面积0.704(95%CI:0.536~0.839),差异无统计学意义(Z=0.382,P>0.05),动脉期NIC诊断效果较佳。结论光谱CT可优化显示宫颈癌病灶及其浸润范围,对评价淋巴结转移有优势,动脉期IC、NIC在宫颈癌早期与进展期鉴别中有一定的诊断价值。

四川某县城普通高中教师职业倦怠

目的：调查县城普通高中教师职业倦怠情况。方法：采用Maslach-ES（1996）编制的《教师职业倦怠量表》（MBI）,同时调查了174名县城普通高中教师的工作现状。结果：（1）县城普通高中教师职业倦怠得分情绪衰竭较为严重,去个性化程度很低,成就感很低。（2）性别、不同职称、年龄、不同学科、教龄对教师职业倦怠没有显著影响;是否班主任,婚姻状况,不同年级,工作时间是影响教师职业倦怠的重要因素。（3）班主任在情感衰竭上得分高于非班主任（t=2.55,P〈0.05）;（4）已婚教师在去个性化得分低于未婚教师（t=-3.13,P〈0.01）;已婚教师在成就感得分低于未婚教师（t=-3.13,P〈0.01）;（5）不同年级间的情感衰竭得分差异具有统计学意义（F=4.19,P〈0.05）;不同年级间的去个性化得分差异具有统计学意义（F=5.97,P〈0.01）;不同年级间的成就感得分差异具有统计学意义（F=5.97,P〈0.01）;（6）工作时间的情感衰竭得分差异具有统计学意义（F=6.71,P〈0.001）;工作时间的去个性化得分差异具有统计学意义（F=5.61,P〈0.01）;工作时间的成就感得分差异具有统计学意义（F=5.61,P〈0.01）;（7）情感衰竭在教龄第3-5年达到最高值,教龄11-15年在去个性化水平处于最低值,这一阶段也是成就感最低的阶段;教龄3-5年是教师职业倦怠最严重的阶段;（8）学校管理和制度是教师压力主要来源,近一半的教师幸福感受为一般。结论：总的来说,教师情绪衰竭较为严重,成就感极低,而教师对本工作满意。

The color and pattern of watermelon rind are crucial external traits that directly affect consumer *** with stripes having a stronger color than the background rind are ideal for studying stripe patterns in plants,while there is still limited knowledge about the genetic mechanisms underlying stripe coloration due to the lack of germplasm *** this study,we focused on a watermelon germplasm with colorless stripes,and genetic analysis revealed that the trait is controlled by a single recessive *** gene Clsc(Citrullus lanatus stripe coloration),which is responsible for the colorless stripe,was localized into a 147.6 kb region on Chr9 by linkage analysis in a large F2 mapping *** analysis revealed that the Cla97C09G175170 gene encodes the APRR2 transcription factor,plays a crucial role in determining the watermelon colorless stripe phenotype and was deduced to be related to chlorophyll synthesis and chloroplast *** experiments indicated that Cla97C09G175170 may significantly influence chloroplast development and chlorophyll synthesis in *** results of this study provide a better understanding of the molecular mechanism of stripe coloration in watermelon and can be useful in the development of marker-assisted selection(MAS)for new watermelon cultivars.

目的对2型糖尿病患者微血管病变与血浆同型半胱氨酸水平的关系进行深入探究。方法以2019年1月至2020年1月作为研究时段,选取我院接纳治疗的2型糖尿病患者100例为研究对象,依据患者是否存在微血管病变分为无微血管病变组(50例)以及微血管病变组(50例),另选同期体检健康患者50例为对照组,对所有研究对象的收缩压、血肌酐、糖化血红蛋白、同型半胱氨酸水平进行检测,并对相关数据实行对比分析。结果微血管病变组、无微血管病变组、对照组间的收缩压、血肌酐、糖化血红蛋白、同型半胱氨酸数据差异有显著统计学意义(P <0.05);且经多因素Logistic回归分析,收缩压、血肌酐、糖化血红蛋白、同型半胱氨酸水平均为患者微血管病变的独立危险因素(P <0.05)。结论在2型糖尿病患者微血管病变中,血浆同型半胱氨酸水平属独立危险因素,故可将其作为2型糖尿病病变诊断指标。
