Apoptosis induction of colorectal cancer cells HTL-9 in vitro by the transformed products of soybean isoflavones by Ganoderma lucidum

Soybean isoflavones have been one of the potential preventive candidates for antitumor research in recent years. In this paper, we first studied the transformation of soybean isoflavones with the homogenized slurry of Ganoderma lucidum. The resultant transformed products（TSI） contained（703.21±4.35） mg/g of genistein, with transformed rates of 96.63% and 87.82% of daidzein and genistein, respectively, and TSI also could enrich the bioactive metabolites of G. lucidum. The antitumor effects of TSI on human colorectal cancer cell line HTL-9, human breast cancer cell line MCF-7, and human immortalized gastric epithelial cell line GES-1 were also studied. The 3-（4,5-dimethyl-2-thiazolyl）-2,5-diphenyl-2-H-tetrazolium bromide（MTT） assay showed that TSI could dramatically reduce the viability rates of HTL-9 cells and MCF-7 cells without detectable cytotoxicity on GES-1 normal cells when the TSI concentration was lower than 100 μg/ml. With 100 μg/ml of TSI, HTL-9 cells were arrested in the G1 phase, and late-apoptosis was primarily induced, accompanied with partial early-apoptosis. TSI could induce primarily earlyapoptosis by arresting cells in the G1 phase of MCF-7 cells. For HTL-9 cells, Western-blot and reverse-transcriptase polymerase chain reaction（RT-PCR） analysis showed that TSI（100 μg/ml） can up-regulate the expression of Bax, Caspase-3, Caspase-8, and cytochrome c（Cyto-c）, indicating that TSI could induce cell apoptosis mainly through the mitochondrial pathway. In addition, the expression of p53 was up-regulated, while the expression of Survivin and nuclear factor κB（NF-κB） was down-regulated. All these results showed that TSI could induce apoptosis of HTL-9 cells by the regulation of multiple apoptosis-related genes.

Submerged fermentation production and characterization of intracellular triterpenoids from Ganoderma lucidum using HPLC-ESI-MS

As the main bioactive metabolites of Ganoderma lucidum, triterpenoids have various pharmacological effects. In this paper, the nutritional requirements and culture conditions of a submerged culture of G. lucidum were optimized using the response surface methodology; maximum mycelia biomass and intracellular triterpenoid production reached 1.87 g/100 ml and 93.21 mg/100 ml, respectively, for a culture consisting of wort 4.10% （0.041 g/ml） and yeast extract 1.89% （0.0189 g/ml）, pH 5.40. For the first time, we established that wort, which is cheap and abundant, can replace the more commonly used glucose as the sole source of carbohydrate. Using high-performance liquid chromatography-electrospray ionization-mass spectrometry （HPLC-ESI-MS）, 10 major ganoderic acids were tentatively identified based on the predominant fragmentation pathways with the elimination of H20 and CO2, as well as cleavage of the D-ring.

Atmospheric chemistry of 2-nitrobenzaldehyde:Initiated by photo-excitation,OH-oxidation,and small TiO_(2) clusters adsorption catalysis

The fate of 2-nitrobenzaldehyde(2-NBA)is of interest in atmospheric chemistry as it is a semi-volatile organic compound with high *** study presents a quantum chemical study of the gas-phase reactions of 2-NBA photo-excitation and OH-oxidation in the absence and presence of small TiO_(2) *** further understand the unknown photolysis mechanism,the photo-reaction pathways of ground singlet state and the lying excited triplet state of 2-NBA were investigated including the initial and subsequent reactions of proton transfer,direct CO,NO_(2),and HCO elimination routes in the presence of O_(2) and ***,the OH-mediated degradation of 2-NBA proceeded via five H-extraction and six OH-addition channels by indirect mechanism,which follows a succession of reaction steps initiated by the formation of weakly stable intermediate *** H-extraction from the-CHO group was the dominant pathway with a negative activation energy of-1.22 kcal/*** calculated rate coefficients at 200–600 K were close to the experimental data in literature within 308-352 K,and the kinetic negative temperature independence was found in both experimental literature and computational ***,2-NBA was favored to be captured onto small TiO_(2) clusters via six adsorption configurations formed via various combination of three types of bonds of Ti…O,Ti…C,and O…H between the molecularly adsorbed 2-NBA and TiO_(2) *** indicted that the chemisorptions of aldehyde oxygen have largest *** results suggested adsorption conformations have a respectable impact on the catalysis *** study is significant for understanding the atmospheric chemistry of 2-nitrobenzaldehyde.

Potassium-ion hybrid capacitors(PIHCs)are widely regarded as highly promising energy storage devices,due to their exceptional energy density,impressive power density,and abundant potassium ***,restricted by the inherent capacitive storage mechanism,the carbon cathodes possess a much lower specific capacity than battery-type ***,designing high-performance carbon cathodes is extremely urgent for the development of ***,N,O codoped porous carbon(NOPC)was fabricated through the NaCl hard template method and combined KOH/melamine chemical activation technique,displaying the characteristics of abundant N/O content(4.7 at%/16.9 at%),ultrahigh specific surface area(3092 m^(2)g^(-1))and hierarchical pore *** designed NOPC cathode delivers a high specific capacity(164.4 mAh.g^(-1)at 0.05 A.g^(-1))and superior cyclability(95.1%retention ratio at 2 A·g^(-1)over 2500 cycles).Notably,the adjustable ratio of micropores to mesopores facilitates the achievement of the optimal bal-ance between capacity and rate ***,the pseudocapacitance can be further augmented through the incorporation of N/O functional *** expected,the graphite//NOPC based PIHC possesses a high energy density of 113 Wh·kg-at 747 W·kg^(-1)and excellent capacity retention of 84.4% fter 400 cycles at 1.0 A·g^(-1).This work introduces a novel strategy for designing carbon cathodes that enhances the electrochemical performance of PIHCs.

Objective To investigate the effects and mechanisms of olfactory three-needle(OTN)electroacupuncture(EA)stimulation of the olfactory system on cognitive dysfunction,synaptic plasticity,and the gut microbiota in senescence-accelerated mouse prone 8(SAMP8)*** Thirty-six SAMP8 mice were randomly divided into the SAMP8(P8),SAMP8+OTN(P8-OT),and SAMP8+nerve transection+OTN(P8-N-OT)groups according to a random number table(n=12 per group),and 12 accelerated senescence-resistant(SAMR1)mice were used as the control(R1)*** was performed at the yintang(GV 29)and bilateral yingxiang(LI 20)acupoints of SAMP8 mice for 4 *** Morris water maze test,transmission electron microscopy,terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling(TUNEL)staining,Nissl staining,Golgi staining,Western blot,and 16S rRNA sequencing were performed,*** Compared with the P8 group,OTN improved the cognitive behavior of SAMP8 mice,inhibited neuronal apoptosis,increased neuronal activity,and attenuated hippocampal synaptic dysfunction(P<0.05 or P<0.01).Moreover,the expression levels of synaptic plasticity-related proteins N-methyl-D-aspartate receptor 1(NMDAR1),NMDAR2B,synaptophysin(SYN),and postsynaptic density protein-95(PSD95)in hippocampus were increased by OTN treatment(P<0.05 or P<0.01).Furthermore,OTN greatly enhanced the brain-derived neurotrophic factor(BDNF)/cAMP-response element binding(CREB)signaling and phosphatidylinositol 3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/AKT/mTOR)signaling compared with the P8 group(P<0.05 or P<0.01).However,the neuroprotective effect of OTN was attenuated by olfactory nerve *** with the P8 group,OTN had a very limited effect on the fecal microbial structure and composition of SAMP8 mice,while specifically increased the genera Oscillospira and Sutterella(P<0.05).Interestingly,the P8-N-OT group showed an abnormal fecal microbiota with higher microbialα-diversity,Firmicutes/Bacter

Pogostemon cablin essential oil as feed additive promotes the repair of the rumen epithelial barrier in heat-stressed beef cattle

Pogostemon cablin essential oil(PEO),extracted from ***,has anti-oxidant,anti-inflammatory,and anti-stress properties,as well as the ability to improve gastrointestinal *** study aims to evaluate the effects of PEO on the performance,rumen epithelial morphology,and barrier function in heat-stressed beef ***-six male Jingjiang cattle at 18 months old were randomly assigned into four groups and fed a diet containing PEO at 0(control),50,100,or 150 mg/kg in the feed concentrate(n=9).All experimental cattle were fed under high temperature and humidity in summer for 60 *** results indicated that 50 mg/kg of PEO treatment enhanced the average daily gain of beef cattle compared with the control group(P=0.032).All PEO treatments reduced the diamine oxidase activity(P=0.004)and malondialdehyde content(P=0.008)in *** addition,the content of 70 kDa heat shock protein in the 100 mg/kg group was increased,and the activity of glutathione peroxidase and total antioxidant capacity in both 100 mg/kg and 150 mg/kg groups were enhanced compared to the control group(P<0.05).More importantly,PEO treatment with 50 mg/kg enhanced the mRNA relative expressions of occludin in ruminal epithelia but decreased the mRNA relative expressions of c-Jun N-terminal kinase,P38 mitogen-activated protein kinases,caspase-3,Beclin1(P<0.05),and extremely significant declined the mRNA relative expressions of extracellular regulated protein kinases and ubiquitin-binding protein in contrast to the control group(P<0.01).These findings indicated that dietary PEO supplementation might be favorable to improve growth performance and repairing damaged rumen epithelium of heat-stressed cattle by down-regulating the mitogen-activated protein kinase signaling pathway.

Fluorescent carbon dots synthesized in solid phase and air for application in LEDs

Carbon dots(CDs)have been widely adopted as optical materials because of their excellent luminescent ***,most of the reported synthetic methods are conducted in solvents,especially hydrothermal/solvothermal reactions,leading to intractable problems such as toxic and flammable solvents,complex and inseparable by-products,and dangerously high pressures and ***-phase synthesis of CDs in air is an effective solution to overcome the above issues,but solid reactions always result in uncontrolled growth and agglomeration of *** this study,some inorganic salts are selected as catalysts for synthesizing CDs in solid states and air,which also play as dispersants to hinder CDs *** the meantime,some aromatic derivatives containing hydroxyl and amino groups are chosen as carbon sources,ground with the optimized catalyst,and then heated together in *** production yields are affected by the reaction time and reactant ratio,while the graphitization degrees of the CDs are determined by the reaction *** IG/ID value of their Raman spectra increases from 0.59 to 0.85,and the particle size decreases from 2.5 to 1.4 nm when the synthesis temperature is increased from 200 to 280℃.The as-prepared CDs show emission peaks ranging from 366 to 606 nm,with the photoluminescence(PL)quantum yield up to 53%.Their emission color variation mainly results from different carbon sources,which can be ascribed to the differences in the element composition,functional groups,and graphitic nitrogen content of these *** dispersing CDs of different concentrations into polyvinyl alcohol(PVA)and combining them with blue LEDs,cold,standard,and warm white light emitting devices(WLEDs)are prepared,with a color rendering index(CRI)up to *** the as-prepared CDs have antioxidant ability at high temperature,the as-prepared WLEDs have long lifespans,remaining the effective white luminescence after 72 h continuous work.

A regression approach for seismic first-break picking

The picking efficiency of seismic first breaks(FBs)has been greatly accelerated by deep learning(DL)***,the picking accuracy and efficiency of DL methods still face huge challenges in low signal-to-noise ratio(SNR)*** address this issue,we propose a regression approach to pick FBs based on bidirectional long short-term memory(Bi LSTM)neural network by learning the implicit Eikonal equation of 3D inhomogeneous media with rugged topography in the target *** employ a regressive model that represents the relationships among the elevation of shots,offset and the elevation of receivers with their seismic traveltime to predict the unknown FBs,from common-shot gathers with sparsely distributed *** from image segmentation methods which automatically extract image features and classify FBs from seismic data,the proposed method can learn the inner relationship between field geometry and *** addition,the predicted results by the regressive model are continuous values of FBs rather than the discrete ones of the binary *** picking results of synthetic data shows that the proposed method has low dependence on label data,and can obtain reliable and similar predicted results using two types of label data with large *** picking results of9380 shots for 3D seismic data generated by vibroseis indicate that the proposed method can still accurately predict FBs in low SNR *** subsequent stacked profiles further illustrate the reliability and effectiveness of the proposed *** results of model data and field seismic data demonstrate that the proposed regression method is a robust first-break picker with high potential for field application.
