Deep learning identification of novel autophagic protein-protein interactions and experimental validation of Beclin 2-Ubiquilin 1 axis in triple-negative breast cancer

A Mn-porphyrinic metal–organic framework immobilizing glucose oxidase for combined photodynamic/chemodynamic/starvation therapy

The combination of photodynamic therapy（PDT） and chemodynamic therapy（CDT） in tumor treatment has attracted considerable ***,tumor hypoxia and glutathione（GSH） overproduction in the tumor tissue restricted the progress of their ***,a Mn-porphyrinic metal-organic framework（Mn-TCPP） was constructed by the one-pot method and further used for immobilizing glucose oxidase（GOx） to obtain GOx@*** would rapidly exhaust endogenous glucose into hydrogen peroxide（H2O2） and gluconic acid,thus shutting off the energy supply of tumor cells for starvation ***-TCPP catalyzed H2O2to produce oxygen,regulating the hypoxic tumor microenvironment and in turn improving1O2generation under laser ***,Mn-TCPP can reduce reactive oxygen species consumption owing to the redox reaction between Mn3+and GSH,thus greatly enhancing ***,benefiting from GOx-mediated starvation therapy,Mn2+produced by Mn3+reduction can react with sufficient intracellular H2O2to generate ·OH with high cytotoxicity through a Fenton-like *** treatment by GOx@Mn-TCPP under laser irradiation in vitro,the cell viability of 4T1 and A549 tumor cells reached to 20%,reflecting GOx@Mn-TCPP could give full play to the advantages of PDT/CDT/starvation *** results in vivo demonstrated that GOx@Mn-TCPP mediated synergistic cascade therapy could significantly inhibit tumor growth and improve the therapeutic effect.

The application of industrial solid wastes as environmentally functional materials for air pollutants control has gained much attention in recent years due to its potential to reduce air pollution in a cost-effective *** this review,we investigate the development of industrialwaste-based functional materials for various gas pollutant removal and consider the relevant reaction mechanism according to different types of industrial solid *** see a recent effort towards achieving high-performance environmental functional materials via chemical or physical modification,in which the active components,pore size,and phase structure can be *** review will discuss the potential of using industrial solid wastes,these modified materials,or synthesized materials from raw waste precursors for the removal of air pollutants,including SO_(2),NO_(x),Hg^(0),H_(2)S,VOCs,and CO_(2).The challenges still need to be addressed to realize this potential and the prospects for future research *** suggestions for future directions include determining the optimal composition of these materials,calculating the real reaction rate and turnover frequency,developing effective treatment methods,and establishing chemical component databases of raw industrial solid waste for catalysts/adsorbent preparation.

Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. Since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome,proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial sc RNA-seq and CRISPR screening. Single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

Bai Le Mian capsule（BLMC） which is composed of fifteen kinds of traditional Chinese Medicine,Lilium Brownii,Acanthopanax Senticosus,Polygonum Multiflorum,Albizia Julibrissin,etc,is a popular traditional Chinese patent medicine for the treatment of insomnia with potential effect on *** the present study,The effects of acute and chronic effects of BLMC（81,162,325 mg/kg,ig） on antianxiety was tested in the elevated plus maze,open field,and light/dark box,in normal mice and an anxious model using Meta-chlorophenyl-piperazine（mCPP）,a compound with known anxiogenic *** 1 mg/kg was used as reference *** administered BLMC *** 325 mg/kg prolonged the time spent in central area 1.1-fold,and two-week medication,the time spent in central area,open arms area and light box were increased by 2.8-2.0-and 1.7-fold,respectively,compared with vehicle control in normal ***,in anxious model,Acute treatment of BLMC（325 mg/kg） mice spent notably more time in the open arms area 2.1-fold,andconsecutive administration for two weeks,BLMC（325 mg/kg） increased the time spent in central area,open area and light box by 1.6-,2.3-,2.1-fold,respectively,compared with vehicle ***,BLMC（162 mg/kg） also increased the time of light box 2.1-fold in mCPP anxious mice *** study showed that mCPP increased c-Fos expression in the neurons of central nucleus of the amygdala and bed nucleus of stria terminalis by 57% and 60%,respectively,which could be reversed by *** together,these results indicate that BLMC is an effective treatment for anxiety in mice and may in patients.

We spend nearly one-third of our lives asleep and some rodents even spend half or more experience in this ***,the neuronal mechanism of sleep is still one of the great unsolved scientific *** hypothesis is that sleep results from the inhibition of wake-promoting *** rostromedial tegmental nucleus（RMTg）,a newly identified structure in the brainstem,is rich in μ-opioid receptors and primarily comprises GABAergic *** this study,we found that activation of rat RMTg neurons by pharmacogenetics promoted non-rapid eye movement（NREM）sleep with increased slow-wave activity（SWA）,which were mimicked through specific inhibition of ventral tegmental area（VTA）/substantia nigra compacta（SNc）dopaminergic *** contrast,lesion or inactivation of rat RMTg neurons led to decreased NREM sleep with reduced SWA,which were also mimicked by specific activation of the midbrain dopaminergic neuronal ***,electrophysiological experiments combined with optogenetics illustrated a direct inhibitory connection between the terminals of RMTg neurons and midbrain dopaminergic *** study thus provides an insight into the essential sleep-promoting role of the RMTg in both sleep amount and sleep quality.

Insufficient MIRO1 contributes to declined oocyte quality during reproductive aging

Mitochondrial Rho-GTPase 1 (MIRO1) is an outer mitochondrial membrane protein which regulates mitochondrial transport and mitophagy in mitosis. In present study, we reported the crucial roles of MIRO1 in mammalian oocyte meiosis and its potential relationship with aging. We found that MIRO1 expressed in mouse and porcine oocytes, and its expression decreased in aged mice. MIRO1 deficiency caused the failure of meiotic resumption and polar body extrusion in both mouse and porcine oocytes, which could be rescued by exogenous MIRO1supplementation. Mass spectrometry data indicated that MIRO1 associated with several cytoskeleton and cell cycle-related proteins, and MIRO1 regulated motor protein Dynein for microtubule-organizing centers (MTOCs) dynamics at germinal vesicle (GV) stage, which determined meiotic resumption. Furthermore, we found that MIRO1 regulated Aurora A and kinesin family member 11 (KIF11) for meiotic spindle assembly in oocytes. Besides, MIRO1 associated with several mitochondria-related proteins dynamic-related protein 1 (DRP1),Parkin and lysosomal-associated membrane protein 2 (LAMP2) for mitochondrial dynamics and mitophagy during oocyte meiosis. Taken together, our results suggested that MIRO1 played pivotal roles in meiotic resumption, spindle assembly and mitochondrial function in mouse and porcine oocytes, and its insufficiency might contribute to the oocyte maturation defects during aging.

Conductive polymer foam(CPF)with excellent compressibility and variable resistance has promising applications in electromagnetic interference(EMI)shielding and other integrated functions for wearable ***,its insufficient change amplitude of resistance with compressive strain generally leads to a degradation of shielding performance during ***,an innovative loading strategy of conductive materials on polymer foam is proposed to significantly increase the contact probability and contact area of conductive components under *** inter-skeleton conductive films are constructed by loading alginate-decorated magnetic liquid metal on the polymethacrylate films hanged between the foam skeleton(denoted as AMLM-PM foam).Traditional point contact between conductive skeletons under compression is upgraded to planar contact between conductive ***,the resistance change of AMLM-PM reaches four orders of magnitude under ***,the inter-skeleton conductive films can improve the mechanical strength of foam,prevent the leakage of liquid metal and increase the scattering area of EM ***-PM foam has strain-adaptive EMI shielding performance and shows compression-enhanced shielding effectiveness,solving the problem of traditional CPFs upon *** upgrade of resistance response also enables foam to achieve sensitive pressure sensing over a wide pressure range and compression-regulated Joule heating function.
