Ancient mitochondrial genome depicts sheep maternal dispersal and migration in Eastern Asia

Sheep have been one of the most important groups of animals since ancient times.However,the knowledge of their migration routes and genetic relationships is still poorly understood.To investigate sheep maternal migration histories alongside Eurasian communications routes,in this study,we obtain mitochondrial genomes(mitogenomes)from 17 sheep remains in 6 Chinese sites and 1 Uzbekistan site dated 4429–3100 years before present(BP).By obtaining the mitogenomes from the sheep(4429–3556 BP)found in the Tongtian Cave site in Xinjiang,Altai region of northwest China,our results support the emergence of haplogroup C sheep in Xinjiang as early as 4429–3556 BP.The combined phylogenetic analyses with extant ancient and modern sheep mitogenomes suggest that the Uzbekistan-Altai region may have been a migration hub for early sheep in eastern Asia.At least two migration events have taken place for sheep crossing Eurasia to China,one passing by Uzbekistan and Northwest China to the middle and lower reaches of the Yellow River at approximately 4000 BP and another following the Altai region to middle Inner Mongolia from 4429 BP to 2500 BP.Overall,this study provides further evidence for early sheep utilization and migration patterns in Eastern Asia.

Early fire detection technology based on improved transformers in aircraft cargo compartments

The implementation of early and accurate detection of aircraft cargo compartment fire is of great significance to ensure flight safety.The current airborne fire detection technology mostly relies on single-parameter smoke detection using infrared light.This often results in a high false alarm rate in complex air transportation envi-ronments.The traditional deep learning model struggles to effectively address the issue of long-term dependency in multivariate fire information.This paper proposes a multi-technology collaborative fire detection method based on an improved transformers model.Dual-wavelength optical sensors,flue gas analyzers,and other equipment are used to carry out multi-technology collaborative detection methods and characterize various feature dimensions of fire to improve detection accuracy.The improved Transformer model which integrates the self-attention mechanism and position encoding mechanism is applied to the problem of long-time series modeling of fire information from a global perspective,which effectively solves the problem of gradient disappearance and gradient explosion in traditional RNN(recurrent neural network)and CNN(convolutional neural network).Two different multi-head self-attention mechanisms are used to classify and model multivariate fire information,respectively,which solves the problem of confusing time series modeling and classification modeling in dealing with multivariate classification tasks by a single attention mechanism.Finally,the output results of the two models are fused through the gate mechanism.The research results show that,compared with the traditional single-feature detection technology,the multi-technology collaborative fire detection method can better capture fire information.Compared with the traditional deep learning model,the multivariate fire pre-diction model constructed by the improved Transformer can better detect fires,and the accuracy rate is 0.995.

Hepatitis B virus(HBV)reactivation is a clinically significant challenge in disease management.This review explores the immunological mechanisms underlying HBV reactivation,emphasizing disease progression and management.It delves into host immune responses and reactivation’s delicate balance,spanning innate and adaptive immunity.Viral factors’disruption of this balance,as are interac-tions between viral antigens,immune cells,cytokine networks,and immune checkpoint pathways,are examined.Notably,the roles of T cells,natural killer cells,and antigen-presenting cells are discussed,highlighting their influence on disease progression.HBV reactivation’s impact on disease severity,hepatic flares,liver fibrosis progression,and hepatocellular carcinoma is detailed.Management strategies,including anti-viral and immunomodulatory approaches,are critically analyzed.The role of prophylactic anti-viral therapy during immunosuppressive treatments is explored alongside novel immunotherapeutic interventions to restore immune control and prevent reactivation.In conclusion,this compre-hensive review furnishes a holistic view of the immunological mechanisms that propel HBV reactivation.With a dedicated focus on understanding its implic-ations for disease progression and the prospects of efficient management stra-tegies,this article contributes significantly to the knowledge base.The more profound insights into the intricate interactions between viral elements and the immune system will inform evidence-based approaches,ultimately enhancing disease management and elevating patient outcomes.The dynamic landscape of management strategies is critically scrutinized,spanning anti-viral and immunomodulatory approaches.The role of prophylactic anti-viral therapy in preventing reactivation during immunosuppressive treatments and the potential of innovative immunotherapeutic interventions to restore immune control and proactively deter reactivation.

AIM:To explore the effect of epidermal growth factor receptor(EGFR)inhibition by erlotinib and EGFR siRNA on epidermal growth factor(EGF)-induced activation of retinal pigment epithelium(RPE)cells.METHODS:Human RPE cell line(ARPE-19 cells)was activated by 100 ng/mL EGF.Erlotinib and EGFR siRNA were used to intervene EGF treatment.Cellular viability,proliferation,and migration were detected by methyl thiazolyl tetrazolium(MTT)assay,bromodeoxyuridine(BrdU)staining assay and wound healing assay,respectively.EGFR/protein kinase B(AKT)pathway proteins and N-cadherin,α-smooth muscle actin(α-SMA),and vimentin were tested by Western blot assay.EGFR was also determined by immunofluorescence staining.RESULTS:EGF treatment for 24h induced a significant increase of ARPE-19 cells’viability,proliferation and migration,phosphorylation of EGFR/AKT proteins,and decreased total EGFR expression.Erlotinib suppressed ARPE-19 cells’viability,proliferation and migration through down regulating total EGFR and AKT protein expressions.Erlotinib also inhibited EGF-induced an increase of proliferative and migrative ability in ARPE-19 cells and clearly suppressed EGF-induced EGFR/AKT proteins phosphorylation and decreased expression of N-cadherin,α-SMA,and vimentin proteins.Similarly,EGFR inhibition by EGFR siRNA significantly affected EGF-induced an increase of cell proliferation,viability,and migration,phosphorylation of EGFR/AKT proteins,and up-regulation of N-cadherin,α-SMA,and vimentin proteins.CONCLUSION:Erlotinib and EGFR-knockdown suppress EGF-induced cell viability,proliferation,and migration via EGFR/AKT pathway in RPE cells.EGFR inhibition may be a possible therapeutic approach for proliferative vitreoretinopathy(PVR).

Evolution of interfacial heat transfer,contact behavior and microstructure during sub-rapid solidification of molten steel with different hydrogen contents

Typical Q235 low-carbon steel samples with different hydrogen contents(0.0004,0.0008,and 0.0013 wt.%)were prepared by adjusting the environment humidity and moisture.The effects of hydrogen on interfacial heat transfer,contact behavior,and microstructure evolution were investigated using a novel droplet solidification technique.The results revealed that when the hydrogen content increases from 0.0004 to 0.0013 wt.%,the maximum heat flux between the molten steel and cooling substrate decreases from 8.01 to 6.19 MW/m^(2),and the total heat removed in the initial 2 s reduces from 10.30 to 8.27 MJ/m^(2).Moreover,the final contact angle between the molten steel and substrate increases from 103.741°to 113.697°,and the number of pores on the droplet bottom surface increases significantly from 21 to 210 with the increase in hydrogen.The surface roughness of the droplet bottom surface increases from 20.902 to 49.181 pm.In addition,the average grain size of the droplet increases from 14.778 to 33.548 pm with the increase in the hydrogen content.The interfacial contact condition becomes worse due to the escape of hydrogen from the steel matrix during the cooling process,which leads to the reduction in the interfacial heat transfer and the increase in the grain size.

Important challenges must be addressed to make wind turbines sustainable renewable energy sources.A typical problem concerns the design of the foundation.If the pile diameter is larger than that of the jacket platform,traditional mechanical models cannot be used.In this study,relying on the seabed soil data of an offshore wind farm,the m-method and the equivalent embedded method are used to address the single-pile wind turbine foundation problem for different pile diameters.An approach to determine the equivalent pile length is also proposed accordingly.The results provide evidence for the effectiveness and reliability of the model based on the equivalent embedded method.

An experimental and theoretical study on photoluminescence of Ce^(3+)-doped ZnS quantum dots and their application in Aspergillus oryzae

ZnS and ZnS:3.125%Ce^(3+)(ZSC)quantum dots(QDs)were synthesized by a low-temperature solid-phase method.The crystal structure,photoluminescence(PL)properties of ZSC QDs,and their effects on the growth and kojic acid production of Aspergillus oryzae(A.oryzae)were investigated.It is found that both QDs have a cubic blend structure with the average particle size ranging of 2.75-6.14 nm.Ce^(3+)doping not only increases the band gap of ZnS QDs from 3.43 to 3.48 eV,but also increases the PL intensity by about 2.23 times,which is due to the fact that the electron carrier concentration is improved,and then the rate of radiative recombination is increased.The integral emission intensity,biomass,extracellular proteins yield,and kojic acid production of A.oryzae cultured with ZSC(ZnS)QDs are about 4.31(3.23),1.61(1.20),1.12(1.09),and 2.99(1.68)times higher than that of medium without QDs,respectively.

Analysis of factors impacting postoperative pain and quality of life in patients with mixed hemorrhoids:A retrospective study

BACKGROUND Hemorrhoids are among the most common and frequently encountered chronic anorectal diseases in anorectal surgery.They are venous clusters formed by con-gestion,expansion,and flexion of the venous plexus in the lower part of the rec-tum.Mixed hemorrhoids bleed easily and recurrently,and this can result in severe anemia.Hence,they may have a negative effect on the health of the patient and surgical treatment is required.Milligan-Morgan hemorrhoidectomy has been widely used since 1937 for the treatment of grade III and IV hemorrhoids.How-ever,most patients experience different degrees of postoperative pain that may cause anxiety.with mixed hemorrhoids post-surgery.METHODS The clinical data of patients with mixed hemorrhoids who underwent Milligan-Morgan hemorrhoidectomy were collected retrospectively.The basic character-istics of the enrolled patients with mixed hemorrhoids were recorded,and based on the Goligher clinical grading system,the hemorrhoids were classified as grades III or IV.The endpoint of this study was the disappearance of pain in all patients.Quantitative data were presented as mean±SD,such as age,pain score,and QoL score.Student’s t-test was used to compare the groups.RESULTS A total of 164 patients were enrolled.The distribution of the visual analog scale pain scores of all patients at 3,7,14 and 28 d after surgery showed that post-surgery pain was significantly reduced with the passage of time.Fourteen days after the operation,the pain had completely disappeared in some patients.Twenty-eight days after the surgery,none of the patients experienced any pain.Comparing the World Health Or-ganization Quality of Life-BREF self-reporting questionnaire scores of patients between 14 and 28 d after surgery,we observed that the quality-of-life scores of the patients post-surgery had significantly improved.There were six items that were compared at 14-and 28-d post-surgery.The mean QoL score 28 d after surgery(4.79±0.46)was higher than that at 14 d post-surgery(3.79±0.57).The mean health condition

Irisin-loaded electrospun core-shell nanofibers as calvarial periosteum accelerate vascularized bone regeneration by activating the mitochondrial SIRT3 pathway

The scarcity of native periosteum poses a significant clinical barrier in the repair of critical-sized bone defects.The challenge of enhancing regenerative potential in bone healing is further compounded by oxidative stress at the fracture site.However,the introduction of artificial periosteum has demonstrated its ability to promote bone regeneration through the provision of appropriate mechanical support and controlled release of proosteogenic factors.In this study,a poly(L-lactic acid)(PLLA)/hyaluronic acid(HA)-based nanofibrous membrane was fabricated using the coaxial electrospinning technique.The incorporation of irisin into the core-shell structure of PLLA/HA nanofibers(PLLA/HA@Irisin)achieved its sustained release.In vitro experiments demonstrated that the PLLA/HA@Irisin membranes exhibited favorable biocompatibility.The osteogenic differentiation of bone marrow mesenchymal stem cells(BMMSCs)was improved by PLLA/HA@Irisin,as evidenced by a significant increase in alkaline phosphatase activity and matrix mineralization.Mechanistically,PLLA/HA@Irisin significantly enhanced the mitochondrial function of BMMSCs via the activation of the sirtuin 3 antioxidant pathway.To assess the therapeutic effectiveness,PLLA/HA@Irisin membranes were implanted in situ into critical-sized calvarial defects in rats.The results at 4 and 8 weeks post-surgery indicated that the implantation of PLLA/HA@Irisin exhibited superior efficacy in promoting vascularized bone formation,as demonstrated by the enhancement of bone matrix synthesis and the development of new blood vessels.The results of our study indicate that the electrospun PLLA/HA@Irisin nanofibers possess characteristics of a biomimetic periosteum,showing potential for effectively treating critical-sized bone defects by improving the mitochondrial function and maintaining redox homeostasis of BMMSCs.

Comprehensive Summary,Valbenazine(Ingrezza),a potent and highly selective inhibitor of vesicular monoamine transporter type 2(VMAT2)through the active metabolite hydrotetrabenazine(HTBZ),has been approved for the treatment of tardive dyskinesia and,very recently,for chorea,which is associated with Huntington's disease.Despite numerous synthetic efforts dedicated to the synthesis of HTBZ,the industrial preparation of valbenazine uses dihydroisoquinoline as a starting material and the chiral resolution of racemic HTBZ derived from ketone reduction.Herein,we present a practical synthesis of HTBZ and valbenazine featuring a highly stereoselective 1,3-dipolar cycloaddition and enzymatic kinetic resolution.The cascade process includes cycloaddition,N—O bond cleavage,and lactamization,which proved to be operationally simple.The allure of the enzymatic resolution developed in this work offers a rapid access toward affording tetrahydroisoquinoline(THIQ)-fused piperidine in the production of medically significant compounds,such as yohimbine and reserpine.
