Insufficient MIRO1 contributes to declined oocyte quality during reproductive aging

Mitochondrial Rho-GTPase 1 (MIRO1) is an outer mitochondrial membrane protein which regulates mitochondrial transport and mitophagy in mitosis. In present study, we reported the crucial roles of MIRO1 in mammalian oocyte meiosis and its potential relationship with aging. We found that MIRO1 expressed in mouse and porcine oocytes, and its expression decreased in aged mice. MIRO1 deficiency caused the failure of meiotic resumption and polar body extrusion in both mouse and porcine oocytes, which could be rescued by exogenous MIRO1supplementation. Mass spectrometry data indicated that MIRO1 associated with several cytoskeleton and cell cycle-related proteins, and MIRO1 regulated motor protein Dynein for microtubule-organizing centers (MTOCs) dynamics at germinal vesicle (GV) stage, which determined meiotic resumption. Furthermore, we found that MIRO1 regulated Aurora A and kinesin family member 11 (KIF11) for meiotic spindle assembly in oocytes. Besides, MIRO1 associated with several mitochondria-related proteins dynamic-related protein 1 (DRP1),Parkin and lysosomal-associated membrane protein 2 (LAMP2) for mitochondrial dynamics and mitophagy during oocyte meiosis. Taken together, our results suggested that MIRO1 played pivotal roles in meiotic resumption, spindle assembly and mitochondrial function in mouse and porcine oocytes, and its insufficiency might contribute to the oocyte maturation defects during aging.

All-safe liquid-state lithium-ion batteries(ASLS-LIBs) is of great interest as they can potentially combine the safety of all-solid-state batteries with the high performance and low manufacturing cost of traditional liquid-state LIBs. However, the practical success of ASLS-LIBs is bottlenecked by the lack of advanced separator technology that can simultaneously realize high performances in puncturing-tolerability,fire-resistance, and importantly, wetting-capability with non-flammable liquid-electrolytes. Here, we propose a concept of inorganic in-situ separator(IISS) by hybrid-sol physical crosslinking directly onto the electrode surface to address the above challenges. Particularly, the hybrid-sol is designed with silica nanoparticles as the building block and poly(vinylidene difluoride) nanoparticles as the crosslinking agent. The critical factors for controlling the IISS microstructures and properties have been systematically investigated. The advantages of the IISS have been confirmed by its fast wetting with various fireresistant liquid-electrolytes, customizable thickness and porous structures, robust interface with planar or three-dimensional(3D)-structured electrodes, and importantly, unexpected self-adaptability against puncturing. Enabled by the above merits, a fire-resistant ASLS-LIB is successfully assembled and demonstrated with stable electrochemical performance. This sol-crosslinked IISS may open an avenue for the studies on the next-generation separator technology, cell assembling, solid electrolyte processing as well as non-flammable secondary batteries.

Understanding the mechanical properties of the lithologies is crucial to accurately determine the horizontal stress *** investigate the correlation between the rock mass properties and maximum horizontal stress,the three-dimensional(3D)stress tensors at 89 measuring points determined using an improved overcoring technique in nine mines in China were adopted,a newly defined characteristic parameter C_(ERP)was proposed as an indicator for evaluating the structural properties of rock masses,and a fuzzy relation matrix was established using the information distribution *** results indicate that both the vertical stress and horizontal stress exhibit a good linear growth relationship with *** is no remarkable correlation between the elastic modulus,Poisson's ratio and depth,and the distribution of data points is scattered and ***,there is no obvious relationship between the rock quality designation(RQD)and *** maximum horizontal stress σ_(H) is a function of rock properties,showing a certain linear relationship with the C_(ERP)at the same *** addition,the overall change trend of σ_(H) determined by the established fuzzy identification method is to increase with the increase of C_(ERP).The fuzzy identification method also demonstrates a relatively detailed local relationship betweenσ_H and C_(ERP),and the predicted curve rises in a fluctuating way,which is in accord well with the measured stress data.

The seemingly useless reeds are prepared as thermal insulation materials,which not only meet the requirements of environmental sustainability but also enhance the added value of reeds,creating new economic *** hydrophobicity of raw biomass surfaces leads to problems such as weak bonding strength and non-dense structure in the formed materials,as well as issues related to the residual insect infestations on the *** this study,reed straw was used as the raw material,and foamed geopolymer was used as the binder to prepare building insulation materials based *** improve the interfacial adhesion performance between reed straw and foamed geopolymer,a thermochemical modification method-thermal carbonization,was *** this study,the mechanical properties and hydraulic properties of the studied materials with different degrees of surface thermal modification were tested,especially the fire resistance performance,and weathering resistance performance rarely found in published *** the surface thermal modification condition of reed straw was 250℃（30 min）,the comprehensive performance of reed-based building insulation materials was the best,when the studied material density was 321.3 kg/m3;the compressive strength was 0.59 MPa;the thermal conductivity was 0.101 W/（m·K）;the pH was 11.27;the moisture absorption rate was 25.1%,and the compressive strength loss rate in wet-dry cycles was 18.5%.In addition,it had excellent fire resistance performance and weathering resistance *** new material can be widely used to improve the thermal insulation of traditional buildings and as sandwich filler in prefabricated buildings,such as preparing insulating walls.

A bearing fault diagnosis method based on the Markov transitionﬁeld(MTF)and SEnet(SE)-IShufﬂenetV2 model is proposed in this paper due to the problems of complex working conditions,low fault diagnosis accuracy,and poor generalization of rolling ***,MTF is used to encode one-dimensional time series vibration sig-nals and convert them into time-dependent and unique two-dimensional feature ***,the generated two-dimensional dataset is fed into the SE-IShufﬂenetV2 model for training to achieve fault feature extraction and classiﬁ*** paper selects the bearing fault datasets from Case Western Reserve University and Paderborn University to experimentally verify the effectiveness and superiority of the proposed *** generalization performance of the proposed method is tested under the variable load condition and different signal-to-noise ratios(SNRs).The experimental results show that the average accuracy of the proposed method under different working conditions is 99.2%without adding *** accuracy under different working conditions from 0 to 1 HP is 100%.When the SNR is 0 dB,the average accuracy of the proposed method can still reach 98.7%under varying working ***,the bearing fault diagnosis method proposed in this paper is characterized by high accuracy,strong anti-noise ability,and ***,the proposed method can also overcome the inﬂuence of variable working conditions on diagnosis accuracy,providing method support for the accurate diagnosis of bearing faults under strong noise and variable working conditions.

原油中金属元素钒和镍(V和Ni)的含量和比值可为深入了解原油的沉积环境特征和有机质类型发挥重要作用。在原油炼制过程中,V和Ni作为原油催化裂化过程中的有害金属元素,会降低催化剂的活性,严重时会导致催化剂中毒,会严重腐蚀设备,在精炼之前必须对原油中的V和Ni进行脱除处理。V和Ni具有致突变性和致癌性,识别或监测原油中V和Ni进入环境的潜在危险变得越来越重要,对原油中V和Ni的含量进行高灵敏精准测定具有重要意义。采用电感耦合等离子体串联质谱(ICP-MS/MS)测定原油中V和Ni的新策略。原油样品经航空煤油简单稀释后直接采用ICP-MS/MS进行分析,针对V和Ni受到的质谱干扰,在MS/MS模式下,采用由NH_(3)/He(He为缓冲气)和H_(2)组成的反应气混合物(NH_(3)/He/H_(2))作为反应气,基于V^(+)与NH_(3)/He/H_(2)不发生反应,而V^(+)的干扰离子与NH_(3)/He/H_(2)发生反应,采用原位质量法消除干扰,选择V^(+)进行测定;基于Ni^(+)与NH_(3)/He/H_(2)发生反应,采用质量转移法消除干扰,选择无干扰团簇离子Ni(NH_(3))_(3)^(+)进行测定。通过与NH_(3)/He反应模式进行对比研究发现,对于V的测定,加入H_(2)为反应气能快速去除氩基离子的干扰,并能协同NH_(3)消除氧化物离子的干扰;对于Ni的测定,加入H_(2)为反应气能促进全氢加合物(-NH_(3))的形成,从而提高生成Ni(NH_(3))_(3)^(+)的产率。在NH_(3)/He/H_(2)反应模式下,V和Ni的背景等效浓度(BEC)更低,Ni的灵敏度更高,V和Ni的检出限(LOD)分别低至0.83和3.76 ng·kg^(-1),定量限(LOQ)分别为2.77和12.5 ng·kg^(-1)。通过分析标准参考物质NIST SRM 1634c并进行加标回收实验评价方法的准确度和精密度,结果显示,方法的测定结果与标准参考物质的认证值基本一致,V和Ni的加标回收率分别为97.7%和103%,相对标准偏差(RSD)分别为1.8%和1.9%。所建立的分析方法避免了繁琐的样品预处理过程,具有操作简单快速、灵敏度高、准确度和精密度好的特点,适用于原油中V和Ni的检测,能为原油深加工中V和Ni的脱除提供理论依据。

The rapid development of short video platforms poses new challenges for traditional recommendation *** systems typically depend on two types of user behavior feedback to construct user interest profiles:explicit feedback(interactive behavior),which significantly influences users’short-term interests,and implicit feedback(viewing time),which substantially affects their long-term ***,the previous model fails to distinguish between these two feedback methods,leading it to predict only the overall preferences of users based on extensive historical behavior ***,it cannot differentiate between users’long-term and shortterm interests,resulting in low accuracy in describing users’interest states and predicting the evolution of their *** paper introduces a video recommendationmodel calledCAT-MFRec(CrossAttention Transformer-Mixed Feedback Recommendation)designed to differentiate between explicit and implicit user feedback within the DIEN(Deep Interest Evolution Network)*** study emphasizes the separate learning of the two types of behavioral feedback,effectively integrating them through the cross-attention ***,it leverages the long sequence dependence capabilities of Transformer technology to accurately construct user interest profiles and predict the evolution of user *** results indicate that CAT-MF Rec significantly outperforms existing recommendation methods across various performance *** advancement offers new theoretical and practical insights for the development of video recommendations,particularly in addressing complex and dynamic user behavior patterns.

Several promising plasma biomarker proteins,such as amyloid-β(Aβ),tau,neurofilament light chain,and glial fibrillary acidic protein,are widely used for the diagnosis of neurodegenerative ***,little is known about the long-term stability of these biomarker proteins in plasma samples stored at-80°*** aimed to explore how storage time would affect the diagnostic accuracy of these biomarkers using a large *** samples from 229 cognitively unimpaired individuals,encompassing healthy controls and those experiencing subjective cognitive decline,as well as 99 patients with cognitive impairment,comprising those with mild cognitive impairment and dementia,were acquired from the Sino Longitudinal Study on Cognitive Decline *** samples were stored at-80°C for up to 6 years before being used in this *** results showed that plasma levels of Aβ42,Aβ40,neurofilament light chain,and glial fibrillary acidic protein were not significantly correlated with sample storage ***,the level of total tau showed a negative correlation with sample storage ***,in individuals without cognitive impairment,plasma levels of total protein and tau phosphorylated protein threonine 181(p-tau181)also showed a negative correlation with sample storage *** was not observed in individuals with cognitive ***,we speculate that the diagnostic accuracy of plasma p-tau181 and the p-tau181 to total tau ratio may be influenced by sample storage ***,caution is advised when using these plasma biomarkers for the identification of neurodegenerative diseases,such as Alzheimer's ***,in cohort studies,it is important to consider the impact of storage time on the overall results.
