N-Acetylcysteine attenuates lipopolysaccharide-induced apoptotic liver damage in D-galactosamine-sensitized mice

Melatonin-selenium nanoparticles protects liver against immunological injury induced by bacillus Calmette-Guérin and lipopolysaccharide

Aim: Melatonin-selenium nanoparticle (MT-Se), a novel complex, was synthesized by preparing selenium nanoparticles in a melatonin medium. The present investigation was designed to determine the protective effects of MT-Se against immunological liver injury in mice induced by bacillus Calmette-Guerin (BCG)/ lipopolysaccharide (LPS). Methods: The model of immunological liver injury in mice was prepared. The levels of alanine aminotransferase, aspartate aminotransferase, nitric oxide (NO) in serum, malondialdehyde content, superoxide dismutase (SOD), and glutathione peroxidase (GSH-px) activities in a liver homo-genate were assayed by spectrophotometry. The content of tumor necrosis fac-tor (TNF) and interleukin-1 (IL-1) were determined by ELISA. The splenocyte proliferation was assayed by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetra-zolium bromide (MTT) dye reduction. Meanwhile, a hepatic pathological examination was observed. Results: In the BCG/LPS-induced hepatic injury model, MT-Se administered at doses of 5,10, or 20 mg/kg to the BCG/LPS-treated mice for 10 d significantly reduced the increase in serum aminotransferase, reduced the severe extent of hepatic cell damage and the immigration of inflammatory cells. It also attenuated the increase in the content of thiobarbituric acid-reactive substances and enhanced the decrease in activities of SOD and GSH-px. In contrast, the treatment with MT-Se suppressed the increase in NO level in both the serum and liver tissue. Furthermore, MT-Se significantly lowered an increase in TNF and IL-1 levels in the liver and inhibited the production of TNF and IL-1 by peritoneal macrophages. A downregulation effect of MT-Se on splenocyte proliferation was also observed. Conclusion: MT-Se showed a hepatic protective action on immunological liver injury in mice.

＜正＞ Total glucosides of paeony（TGP）is an active compound extracted from roots of paeonia lactiflora Pall. Our previous study showed that TGP has anti - hepatic injury and anti - hepatofibrotic effects. In the present study, the effects of TGP on the changes of the expressions of G - protein and G - protein - AC - cAMP signal transduction pathways stimulated by platelet - derived growth factor BB （PDGF- BB） in hepatic stellate cells （HSC） were investigated. The changes of the expression of G - protein on HSC - T6 cell membrane induced by PDGFBB （10μg·L-1）were detected by Western- blot analysis. The results showed that PDGF - BB remarkably increased the expression of Gαi2,but had no effect on expression of Gαi1、Gαi3 and Gαs. PDGF - BB decreased the level of cAMP with concentration - dependently. Furthermore, the tendency of cAMP was closely related with the proliferation of HSC - T6. The expression of Gαi2 was remarkably inhibited by TGP,which also increased the level of cAMP,and inhibited the proliferation of HSC - T6. The results indicate that TGP may inhibit the proliferation of HSC - T6 induced by PDGF - BB via regulating G - protein - AC - cAMP pathway.

目的建立人血浆中莫西沙星药物浓度的液质联用(HPLC-MS/MS)检测方法,分别研究空腹和餐后条件下莫西沙星在人体的药代动力学,并评价其生物等效性。方法空腹和餐后各24例健康受试者,用单剂量、随机、开放、双交叉试验设计,受试者单次口服盐酸莫西沙星片受试药物0.4 g和参比药物0.4g,清洗期为1周,用HPLC-MS/MS法测定健康受试者血浆中莫西沙星浓度,用WinNonlin 6.1计算莫西沙星药代动力学参数,评价受试药物和参比药物的生物等效性。结果空腹单次给药后莫西沙星主要药代动力学参数如下:受试药物、参比药物的C_(max)分别为(3.72±0.83),(3.51±0.69)μg·mL^(-1);t_(max)分别为(1.63±1.52),(1.81±1.18)h;t_(1/2)分别为(11.81±1.66),(11.54±1.26)h;AUC_(0-48h)分别为(52.49±10.87),(51.93±10.31)μg·h^(-1)·mL^(-1);AUC0-∞分别为(56.05±11.57),(55.17±11.04)μg·h^(-1)·mL^(-1);餐后单次给药后莫西沙星主要药代动力学参数如下:受试药物、参比药物的C_(max)分别为(3.05±0.46),(3.03±0.61)μg·mL^(-1);t_(max)分别为(2.52±1.40),(2.62±1.23)h;t_(1/2)分别为(12.21±1.87),(12.13±2.04)h;AUC_(0-48 h)分别为(44.52±7.13),(45.93±9.09)μg·h^(-1)·mL^(-1);AUC_(0-∞)分别为(47.73±8.09),(49.31±10.07)μg·h^(-1)·mL^(-1)。结论空腹和餐后给药条件下,国产盐酸莫西沙星片受试药物和进口盐酸莫西沙星片参比药物主要药代动力学参数指标相近,具有生物等效性。

Antiproliferation and apoptosis induction of paeonol in HepG_2 cells

AIM： To investigate the antiproliferative effect of paeonol （Pae） used alone or in combination with chemotherapeutic agents [cisplatin （CDDP）, doxorubicin （DOX） and 5-fluorouracil （5-FU）] on human hepatoma cell line HepG2 and the possible mechanisms. METHODS： The cytotoxic effect of drugs on HepG2 cells was measured by 3-（4, 5-dimethylthiazol-2- yl）-2, 5-diphenyltetra-zolium bromide （MTT） assay. Morphologic changes were observed by acridine orange （AO） fuorescence staining. Cell cycle and apoptosis rate were detected by flow cytometry （FCM）. Drug-drug interactions were analyzed by the coefficient of drug RESULTS： Pae （7.81-250 mg/L） had an inhibitory effect on the proliferation of HepG2 cells in a dose-dependent manner, with the IC50 value of （104.77±7.28） mg/L. AO fluorescence staining and FCM assays showed that Pae induced apoptosis and arrested cell cycle at S phase in HepG2 cells. Further, different extent synergisms were observed when Pae （15.63, 31.25, 62.5 rag/L） was combined with CDDP （0.31-2.5 mg/L）, DOX （0.16-1.25 mg/L）, or 5-FU （12.5-100 mg/L） at appropriate concentrations. The IC50 value of the three drugs decreased dramatically when combined with Pae （P 〈 0.01）. Of the three different combinations, the sensitivity of cells to drugs was considerably ***： Pae had a significant growth-inhibitory effect on the human hepatoma cell line HepG2, which may be related to apoptosis induction and cell cycle arrest. It also can enhance the cytotoxicity of chemotherapeutic agents on HepG2 cells, and the S phase arrest induced by Pae may be one of the mechanisms of these interactions.

Potential therapeutic effect of epigenetic therapy on Ireatment-induced neuroendocrine prostate cancer

Although adenocarcinomas of the prostate are relatively indolent, some patients with advanced adenocarcinomas show recurrence of treatment-induced neuroendocrine prostate cancer, which is highly aggressive and lethal. detailed biological features of treatment-induced neuroendocrine prostate cancer have not been characterized owing to limited biopsies/resections and the lack of a cellular model. In this study, we used a unique cellular model （LNCaP/NE1.8） to investigate the potential role of cancer stem cells in treatment-induced neuroendocrine prostate cancer with acquired resistance to hormonal therapy and chemotherapy. We also studied the role of cancer stem cells in enhancing invasion in treatment-induced neuroendocrine prostate cancer cells that recurred after long-term androgen-ablation treatment. Using an in vitro system mimicking clinical androgen-ablation, our results showed that the neuroendocrine-like subclone NE1.8 cells were enriched with cancer stem cells. Compared to parental prostate adenocarcinoma LNCaP cells, NE1.8 cells are more resistant to androgen deprivation therapy and chemotherapeutic agents and show increased cancer cell invasiveness. Results from this study also suggest a potential epigenetic therapeutic strategy using suberoylanilide hydroxamic acid, a histone deacetylase inhibitor, as a chemotherapeutic agent for therapy-resistant treatment-induced neuroendocrine prostate cancer cells to minimize the risk of prostate cancer recurrence and metastasis.

Melatonin, a novel selective ATF-6 inhibitor, induces human hepatoma cell apoptosis through COX-2 downregulation
