Background:Tourette syndrome(TS)is a neuropsychiatric disorder with onset in childhood that warrants effective therapies.Gut microbiota can affect central physiology and function via the microbiota-gut-brain axis.Therefore,the gut microbiota plays an important role in some mental illnesses.A small clinical trial showed that fecal microbiota transplantation(FMT)may alleviate TS symptoms in children.Herein,FMT effects and mechanisms were explored in a TS mouse model.Methods:TS mice model(TSMO)(n=80)were established with 3,3'-iminodipropionitrile,and 80 mice were used as controls.Mice were grouped into eight groups and were subjected to FMT with feces from children or mice with or without TS,or were given probiotics.Fecal specimens were collected 3 weeks after FMT.16S rRNA sequencing,behavioral observation,and serum serotonin(5-HT)assay were performed.Differences between groups were analyzed using Mann-Whitney U test and Kolmogorov-Smirnov(KS)tests.Results:A total of 18 discriminative microbial signatures(linear discriminant analysis score>3)that varied significantly between TS and healthy mice(CONH)were identified.A significant increase in Turicibacteraceae and Ruminococcaceae in TSMO after FMT was observed(Pliorate TS by promoting 5-HT secretion,and it provides new insights into the underlying mechanisms of FMT as a treatment for TS.

The T cell receptor(TCR)is one of the most complicated receptors in mammalian cells,and its triggering mechanism remains mysterious.As an octamer complex,TCR comprises an antigen-binding subunit(TCRαβ)and three CD3 signaling subunits(CD3ζζ,CD3δε,and CD3γε).Engagement of TCRαβwith an antigen peptide presented on the MHC leads to tyrosine phosphorylation of the immunoreceptor tyrosine-based activation motif(ITAM)in CD3 cytoplasmic domains(CDs),thus translating extracellular binding kinetics to intracellular signaling events.Whether conformational change plays an important role in the transmembrane signal transduction of TCR is under debate.Attracted by the complexity and functional importance of TCR,many groups have been studying TCR structure and triggering for decades using diverse biochemical and biophysical tools.Here,we synthesize these structural studies and discuss the relevance of the conformational change model in TCR triggering.

Cyanobacterial blooms have become a serious global environmental issue due to their potential risk for releasing detrimental secondary metabolites into aquatic ecosystems,posing a great threat to water quality management for public health authorities.Aphanizomenon,a common filamentous cyanobacterial genus belonging to Nostocales,is under particular concern because its several members are able to form harmful blooms.Furthermore,succession of bloom between A.flos-aquae and Microcystis occurs in many natural lakes.To evaluate the competitiveness of A.flos-aquae vs.M.aeruginosa,two sets of experiments at different ratios of biomass at 15℃and 25℃were conducted.Results show that at 15℃,the two species were able to coexist,and A.flos-aquae showed a specific higher growth rate,and its growth was promoted by the presence of M.aeruginosa.At 25℃,the growth of A.flos-aquae was inhibited by the biomass of M.aeruginosa,and M.aeruginosa suppressed A.flos-aquae in competition.Additionally,the vegetative cell size of A.flos-aquae was significantly influenced by the co-culture with M.aeruginosa,whereas the filament length of A.flos-aquae was not significantly affected.This study confirms that temperature is the dominating factor on the succession of A.flos-aquae and M.aeruginosa of a different biomass.

Chimeric antigen receptor （CAR） T cell therapy is a promising cancer treatment that has recently been undergoing rapid development. However, there are still some major challenges, including precise tumor targeting to avoid off-target or ＂on-target/off-tumor＂ toxicity, adequate T cell infiltration and migration to solid tumors and T cell proliferation and persistence across the physical and biochemical barriers of solid tumors. In this review, we focus on the primary challenges and strafegies to design safe and effective CAR T cells, including using novel cutting-edge technologies for CAR and vecfor designs to increase both the safety and efficacy, further T cell modification to overcome the tumorassociated immune suppression, and using gene editing technologies to generate universal CAR T cells. All these efforts promote the development and evolution of CAR T cell therapy and move toward our ultimate goal--curing cancer with high safety, high efficacy, and low cost.

A highly dispersed Pt/copper modified-MnO_(2)catalyst for the complete oxidation of volatile organic compounds:The effect of oxygen species on the catalytic mechanism

Manganese oxide(MnO_(2))exhibits excellent activity for volatile organic compound oxidation.However,it is currently unknown whether lattice oxygen or adsorbed oxygen is more conducive to the progress of the catalytic reaction.In this study,novel hollow highly dispersed Pt/Copper modified-MnO_(2)catalysts were fabricated.Cu^(2+)was stabilized into theδ-MnO_(2)cladding substituting original K+,which produced lattice defects and enhance the content of adsorbed oxygen.The 2.03 wt%Pt Cu_(0.050)-MnO_(2)catalyst exhibited the highest catalytic activity and excellent stability for toluene and benzene oxidation,with T_(100)=160℃under high space velocity(36,000 mL g^(-1)h^(-1)).The excellent performance of catalytic oxidation of VOCs is attributed to the abundant adsorbed oxygen content,excellent low-temperature reducibility and the synergistic catalytic effect between the Pt nanoparticles and Cu_(0.050)-MnO_(2).This study provides a comprehensive understanding of the Langmuir-Hinshelwood(L-H)mechanism occurring on the catalysts.

Severe acute respiratory syndrome coronavirus 2(SARSCoV-2)infection has been extensively shown to cause many neurological sequelae,and cognitive deficits(known as“brain fog”)may particularly and widely occur even in individuals with mild symptoms[1].Peripheral hyperinflammation as well as central nervous system(CNS)local immune responses may synergistically contribute to brain autoimmune injury.In addition to the direct neuroinvasion of SARS-CoV-2 and nonimmune effects such as severe systemic hypoxemia and vascular thrombosis,the central mechanism by which SARSCoV-2 accelerates cognitive-related symptoms may be related to immune effects[2].However,the precise neuroinflammatory mechanisms of SARS-CoV-2 infection have not been fully established.Fernández-Casta-da et al.[3]provided direct evidence and unique insights into the potential mechanism of cognitive impairment in mild respiratory coronavirus disease 2019(COVID-19)cases.
