1 Intr.ductionDefensins ar. small cationic antimicr.bial p.p.ides that function in the host's innate immune system. The human defensin family includes thr.e small p.p.ides fr.m the azur.p.il gr.nules of p.lymor.0}onuclear.cells named human neutr.p.il p.p.ide (HNp.-1, HNp.2, HNp.3,which consist 5%-7% of the p.otein of human neutr.p.il. HNp.4 is ap.r.ximately one hundr.d times less abundant. They demonstr.te antibacter.al, antifungal and antivir.l p.op.r.ies in vitr.. HNp. ar. imp.r.ant comp.nent of nonoxidative mechanism in macr.p.ages, and can dir.ct inactivate the envelop.d vir.ses. Because only sp.cial cells exp.ess defensins. And it is har. to extr.ct them natur.lly and the p.oduction is few. So r.sear.her.exp.ct to obtain defensins highly thr.ugh heter.genous exp.ession by gene engineer.ng technology. In or.er.to exp.ess HNp.1, we cloned the cDNA of HNp.1 fr.m human p.lymor.0}onuclear.cells in p.r.p.er.l blood, and constr.cted its eukar.otic exp.ession vector. which p.ovided a base for.the fur.her.study on its mechanism of antimicr.bial effect.
