High Expression of ERCC1 Is a Poor Prognostic Factor in Chinese Patients with Non-small Cell lung Cancer Receiving Cisplatin-based Therapy

Objective： to determine the role of excision repair cross-complementing group 1 gene （ERCC1） and ribonucleotide reductase subunit M1 （RRM1） expression in predicting response and survival in Chinese patients with advanced stage non-small cell lung cancer （NSCLC） treated with platinum-based chemotherapy. Methods： Formalin-fixed, paraffin-embedded biopsy tissues were retrospectively obtained from 160 advanced NSCLC patients. mRNA expression levels of ERCC1 and RRM1 were determined by real-time PCR. Results： Associations between mRNA expression level of ERCC1 and RRM1 and clinic-pathologic parameters were analyzed. One handred and forty two （88.75%） specimens were successfully amplified. mRNA expression level of ERCC1 and RRM1 was negatively associated with tumor response. ERCC1 expression levels ranged from 0.01 to 78.79 [median 0.63, mean 4.25 and standard deviation （SD） 9.23] and RRM1 from 0.00 to 30.91 （median 0.63, mean 0.87 and SD 3.36）. By adopting cut-off values according to median expression levels, we found that MST in patients with low ERCC1 mRNA levels was significantly longer in overall population than in patients with higher levels （18.63 versus 13.69 months, log-rank 5.73, P=0.017）, but we did not found the survival benefit of the patients with low expression level of RRM1. （19.07 versus 14.88, log-rank 1.66, P=0.197）. Further, in the multivariable Cox regression model analysis we found that low level of ERCC1 expression, the presence of weight loss and target therapy, were significant prognostic factors for survival. Conclusion： ERCC1 expression is associated with patients’ survival in Chinese advanced NSCLC patients treated with platinum-based regimen and may serve as a biomarker in predicting tumor response and clinical outcome in the patient population.

FADD is an important proapoptotic adaptor in death receptor-induced apoptosis. Recently, FADD has been found to participate in a variety of non-apoptotic processes, such as development, cell cycle progression and survival. Its non-apoptotic activities were regulated by the phosphorylated status of the serine residue located at the C-terminal region, a domain distinct from the proapoptotic function related DED and DD domains. However, due to the difficulties in expression and crystallization of natural FADD, by far the molecuiar structures of all FADD variants did not contain the C-terminal region. To elucidate the structure-function relationship of C-terminal region, we need to obtain a FADD variant that containing C-terminal region. In this study, mouse FADD （80-205） containing DD domain and C-terminal region, designated as C-FADD, was expressed in E. coli with His-tag at the N-terminus and purified by Ni^2＋ affinity chromatography. The purified protein existed as a homogenous monomer in glutaraldehyde cross-linking analysis and exhibited a typical a-helix spectrum in CD （circular dichroism） assay. In vitro His-tag pull-down assay demonstrated that the purified C-FADD possessed the CK Ia-binding activity which was important for its non-apoptotic function.

Thickening progression mechanism of silica fume-oil well cement composite system at high temperatures

This work studied the thickening progression mechanism of the silica fume-oil well cement composite system at high temperatures(110-180.C)in order to provide a theoretical guidance for the rational application of silica fume in the cementing *** showed that silica fume seldom affected the thickening progression of oil well cement slurry at 110-120.C,but when temperature reached above130.C,it would aggravate the bulging degree of thickening curves and significantly extend the thickening time,meanwhile causing the abnormal“temperature-based thickening time reversal”and“dosage-based thickening time reversal”phenomena in the range of 130-160.C and 170-180.C *** 130-160.C,the thickening time of oil well cement slurry was mainly associated with the generation rate of calcium hydroxide(CH)*** introduced silica fume would be attracted to the cement minerals'surface that were hydrating to produce CH and agglomerate together to form an“adsorptive barrier”to hinder further hydration of the inner cement ***“adsorptive barrier”effect strengthened with the rising temperature which extended the thickening time and caused the occurrence of the“temperature-based thickening time reversal”*** 170-180.C,the pozzolanic activity of silica fume significantly enhanced and considerable amount of C-S-H was generated,thus the“temperature-based thickening time reversal”vanished and the“dosage-based thickening time reversal”was presented.

COVID-19 is a contagious infection that has severe effects on the global economy and our daily *** diagnosis of COVID-19 is of importance for consultants,patients,and *** this study,we use the deep learning network AlexNet as the backbone,and enhance it with the following two aspects:1)adding batch normalization to help accelerate the training,reducing the internal covariance shift;2)replacing the fully connected layer in AlexNet with three classifiers:SNN,ELM,and ***,we have three novel models from the deep COVID network(DC-Net)framework,which are named DC-Net-S,DC-Net-E,and DC-Net-R,*** comparison,we find the proposed DC-Net-R achieves an average accuracy of 90.91%on a private dataset(available upon email request)comprising of 296 images while the specificity reaches 96.13%,and has the best performance among all three proposed *** addition,we show that our DC-Net-R also performs much better than other existing algorithms in the literature.

Objective This study aimed to evaluate the effects of in-utero exposure to HIV and ART on pregnancy outcome and early growth of *** This cohort study enrolled 802 HIV-infected pregnant women between October 2009 and May 2018 in Guangzhou, China. The women were assigned to receive combination ART(c ART) or mono/dual ART or no treatment. The primary outcomes were the combined endpoints of any adverse pregnancy outcome [including ectopic pregnancy, spontaneous abortion, stillbirth, preterm birth, small for gestational age(SGA)] and adverse early growth outcome(including infant death, HIV infection of mother-to-child transmission, and underweight, wasting and stunting of infants at 4 weeks of age).Results Adverse pregnancy outcomes occurred in 202(35.1%) of all enrolled HIV-infected women, and121(31.3%) of all infants exhibited adverse effects on early growth at 4 weeks of age. The rates of adverse pregnancy outcomes, spontaneous abortion, ectopic pregnancy, stillbirth, infant death and perinatal HIV infection were higher among women not receiving ART, compared to those treated with c ART or mono/dual ART(P < 0.05). However, women treated with c ART had a higher rate of SGA,compared to untreated women(P < 0.05). No differences in early infant growth were observed among the different treatment *** Our findings underscore the essentiality of prioritizing HIV-positive pregnant women for ART, as even mono/dual ART available in resource-limited countries could improve pregnancy outcomes and infant survival.

Night Sleep Duration and Risk of Cognitive Impairment in a Chinese Population： A Cross-sectional Study

Objective Although sleep is one of the most important health-related behavioral factors, the association between night sleep duration and cognitive impairment has not been fully understood. A cross-sectional study was conducted with a random sample of 2,514 participants （≥ 40 years of age; 46.6% women） in China to examine the association between night sleep duration and cognitive impairment. Methods Night sleep duration was categorized as ≤ 5, 6, 7, 8, or ≥ 9 h per night. Cognitive function was measured using the Mini-Mental State Examination. A multivariate regression analysis was used to analyze the association of night sleep duration with cognitive impairment. A total of 122 participants were diagnosed with cognitive impairment. Results A U-shaped association between night sleep duration and cognitive impairment was found. The odds ratios （95% confidence intervals） of cognitive impairment （with 7 h of daily sleep being considered as the reference） for individuals reporting ≤ 5, 6, 8, and ≥ 9 h were 2.14 （1.20-3.83）, 1.13 （0.67-1.89）, 1.51 （0.82-2.79）, and 5.37 （1.62-17.80）, respectively （P ≤ 0.01）. Conclusion Short or long night sleep duration was an important sleep-related factor independently associated with cognitive impairment and may be a useful marker for increased risk of cognitive impairment.

Background:Shenzao dripping pill(SZDP)is empirically prescribed for treating cardiac ***,there is a lack of comprehensive knowledge regarding the underlying mechanisms contributing to its therapeutic *** objective of this study is to investigate the underlying mechanism of SZDP against chronic myocardial ischemia(CMI)in a rat ***:In this study,we utilized electrocardiographic and echocardiographic detection along with pathological tissue analysis to evaluate the efficacy of *** integration of network pharmacology and metabolomics was conducted to investigate the *** docking and molecular dynamics simulations were used to validate the binding energy between the compounds of SZDP and the associated ***:The results showed that SZDP was able to improve T wave voltage,reverse CMI abnormalities in ejection fraction and fractional shortening,and restore histopathological heart *** results indicated that disturbances of metabolic profile in CMI rats were partly corrected after SZDP administration,mainly affecting purine metabolism.13-Docosenamide may be the potential metabolic biomarker of the therapeutic application of SZDP for *** network pharmacology and metabolomics,thiopurine S-methyltransferase(TPMT),xanthine dehydrogenase/oxidase(XDH),bifunctional purine biosynthesis protein ATIC(ATIC),and cytochrome p4501A1(CYP1A1)were identified as possible targets of SZDP to exert therapeutic effects by enhancing the metabolic levels of L-Tryptophan,Deoxyribose 1-phosphate and Phosphoribosyl ***:SZDP has a therapeutic effect on CMI by regulating metabolite levels,acting on the targets of TMPT,XDH,ATIC,and CYP1A1,and reducing cardiomyocyte injury and myocardial fibrosis.

Dear Editor,The Omicron variant of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has rapidly replaced the Delta variant as a dominating SARS-CoV-2 variant because of natural selection,which favors the variant with higher infectivity and stronger vaccine breakthrough ability.1 Among three lineages BA.1(B.1.1.529.1),BA.2(B.1.1.529.2),and BA.3(B.1.1.529.3)of Omicron,BA.2 is rising rapidly worldwide since January 2022 and become a new dominating“variant of concern”in China.2 With the current surge in Omicron in China,both the absolute number of children hospitalized with COVID-19 and the percentage of total COVID-19 hospitalizations have been boosting *** the clinical characteristics of SARS-CoV-2 Omicron subvariant BA.2 in children and the protective effect of vaccines is of great help for epidemic prevention ***,so far no sufficient experimental data have been reported about infectivity,disease severity,vaccine breakthrough capability,and antibody resistance from pediatric BA.2.
