Background:During Enterovirus type 71(EV71)infection,the structural viral protein 1(VP1)activates endoplasmic reticulum(ER)stress associated with peripheral myelin protein 22(PMP22)accumulation and induces ***,the speciﬁc mechanism behind this process remains ***:In this research,we used the VP1-overexpressing mouse Schwann cells(SCs)models co-transfected with a PMP22 silencing or Autocrine motility factor receptor(AMFR/gp78)overexpressing vector to explore the regulation of gp78 on PMP22 and its relationship with autophagy and ***:The activity of gp78 could be inﬂuenced by EV71-VP1,leading to a decrease in the ubiquitination and degradation of PMP22,resulting in PMP22 accumulation in *** VP1-overexpressing mouse SCs,all three ER stress sensors,including pancreatic endoplasmic reticulum kinase(PERK),activating transcription factor 6(ATF6)and inositol-requiring enzyme 1(IRE1)and the related downstream signals(C/EBP-homologous protein(CHOP)and Caspase 12)were activated,as well as the ER-resident chaperone Glucose-regulated protein 78(GRP78).In addition,VP1 upregulated the autophagy marker Microtubule-associated protein 1 light chain 3 beta(LC3B),while PMP22 silencing or gp78 overexpression reversed the ***,PMP22 silencing or gp78 overexpression increased proliferation of EV71-VP1-transfected mouse ***:Gp78 could regulate PMP22 accumulation through ubiquitination degradation and cause ER stress and autophagy in EV71-VP1-overexpressing mouse ***,the gp78/PMP22/ER stress axis might emerge as a promising therapeutic target for myelin and neuronal damage induced by EV71 infection.

As the most abundant living entities in the environment, viruses have been well recognized as crucial members in sustaining biogeochemical cycling. However, the significance of viruses in soil ecosystem multifunctionality remains under-explored. In this study, we used metagenomics and meta-viromics analysis to investigate the role of soil viruses in soil ecosystem functions under heavy, light, and no organochlorine pesticides(OCPs) contamination. In the three types of soil samples collected, lightcontaminated soils supported the highest level of multifunctionality, followed by heavy-contaminated soils and clean ***, our results revealed a positive correlation between bacterial community evenness and multifunctionality index(p <0.05). Dominant bacterial species with biodegradation and stress resistance advantages exhibited higher abundance in OCPaffected soils, potentially playing a core functional supporting role. Furthermore, our results indicated that the species richness and diversity of bacteriophages were positively correlated with multifunctionality(p < 0.05) in OCP-affected soils. Bacteriophages in OCP-affected soils regulate host metabolism and enhance soil ecosystem multifunctionality by infecting functional bacterial hosts and encoding AMGs related to soil element cycling. Our findings emphasize the potential effect of phages on ecosystem multifunctionality in contaminated soil, suggesting that phages may serve as contributors to soil ecology beyond bacteria and other microorganisms. Therefore, in polluted or constrained soils, further research could potentially translate phage communities and related ecological processes into artificial methods for application in soil pollution remediation or ecological restoration.

Here, we report an observation that illustrate the potential of polyelectrolyte microgels in salt-free solutions to display a high ionic conductivity. Laser light scattering and ionic conductivity tests on very dilute aqueous dispersions of the microgels indicate that both small size and swollen state of gel particles play vital roles, which should favor the counterions to freely penetrate and leave gel particles, and thus can contribute to the ion-conducting property. Upon discovering this on microgels that are composed of imidazolium-based poly(ionic liquid), we also illustrate the generality of the finding to single lithium-ion polyelectrolyte microgels that are of more technically relevant features for applications, for instance, as injectable liquid “microgel-in-solution” electrolytes of high conductivity(ca. 8.2 × 10^(-2)S/m at 25.0 ℃ for1.0 × 10^(-2)g/m L of microgels in a LiNO_(3)-free 1:1 v/v mixture of 1,2-dioxolane and dimethoxymethane) and high lithium-ion transference number(0.87) for use in the rechargeable lithium-sulfur battery.

Metal–organic gel(MOG)derived composites are promising multi-functional materials due to their alterable composition,identifiable chemical homogeneity,tunable shape,and porous ***,stable metal–organic hydrogels are prepared by regulating the complexation effect,solution polarity and curing ***,collagen peptide is used to facilitate the fabrication of a porous aerogel with excellent physical properties as well as the homogeneous dispersion of magnetic particles during ***,two kinds of heterometallic magnetic coupling systems are obtained through the application of Kirkendall ***/nitrogen-doped carbon(NC)aerogel demonstrates an ultra-strong microwave absorption of−85 dB at an ultra-low loading of 5%.After reducing the time taken by atom shifting,a FeCo/Fe3O4/NC aerogel containing virus-shaped particles is obtained,which achieves an ultra-broad absorption of 7.44 GHz at an ultra-thin thickness of 1.59 mm due to the coupling effect offered by dual-soft-magnetic ***,both aerogels show excellent thermal insulation property,and their outstanding radar stealth performances in j-20 aircraft are confirmed by computer simulation *** formation mechanism of MOG is also discussed along with the thermal insulation and electromagnetic wave absorption mechanism of the aerogels,which will enable the development and application of novel and lightweight stealth coatings.

Lactic acid(LA)plays a major role in the occurrence,development,and spread of *** by its high accumulation in tumor site,a novel lactate oxidase(LOD)conjugated two-dimensional Pd@Ir nanoplatform(Pd@Ir-LOD,PIL)was fabricated to combine cascade reaction with photothermal for tumor *** detail,the overexpressed LA in tumor microenvironment(TME)was a key factor to activate the PIL-based cascade reaction:(1)Plenty of H_(2)O_(2)could be generated from LA by the catalysis of LOD with O_(2);(2)potent·OH was produced from H_(2)O_(2)due to the peroxidase(POD)-like activity of PIL;(3)meantime,PIL’s catalase(CAT)-like activity could decompose part H_(2)O_(2)into O_(2)to achieve the purpose of LA cyclic ***,the reduced glutathione(GSH)scavenging capability of PIL might protect the produced reactive oxygen species(ROS)from being cleared to further improve the cascade therapeutic *** importantly,PIL had excellent photothermal conversion efficiency(37.35%)and manifested a surprising temperature rising effect in *** together,the decreasing LA concentration,accumulation of high-toxic ROS,the depletion of GSH together with the higher intra-tumoral temperature potently enhanced in vivo antitumor ***,a promising therapeutic tactic based on PIL integrating endogenous LA consumption,chemodynamic therapy(CDT),and photothermal therapy(PTT)has been put forward.

Cell Mechanics Regulates the Dynamic Anisotropic Remodeling of Fibril Matrix at Large Scale

Living tissues often have anisotropic and heterogeneous organizations, in which developmental processes are coordinated by cells and extracellular matrix modeling. Cells have the capability of modeling matrix in long distance;however, the biophysical mechanism is largely unknown. We investigated the dynamic remodeling of collagen I (COL) fibril matrix by cell contraction with designed patterns of cell clusters. By considering cell dynamic contractions, our molecular dynamics simulations predicted the anisotropic patterns of the observed COL bundling in experiments with various geometrical patterns without spatial limitation. The pattern of COL bundling was closely related to the dynamic remodeling of fibril under cell active contraction. We showed that cell cytoskeletal integrity (actin filaments and microtubules), actomyosin contractions, and endoplasmic reticulum calcium channels acting as force generations and transductions were essential for fiber bundling inductions, and membrane mechanosensory components integrin and Piezo played critical roles as well. This study revealed the underlying mechanisms of the cell mechanics-induced matrix remodeling in large scales and the associated cellular mechanism and should provide important guidelines for tissue engineering in potential biomedical applications.

Hexagonal MnMX-based(M=Co or Ni,X=Si or Ge)alloys exhibit giant reversible barocaloric ***,giant volume expansion would result in the as-cast MnMX ingots fragmenting into powders,and inevitably bring the deterioration of mechanical properties and *** fragmentation can bring degradation of structural transformation entropy change during cyclic application and removal of *** this paper,giant reversible barocaloric effects with high thermal cycle stability can be achieved in the epoxy bonded(MnCoGe)0.96(CuCoSn)0.04 *** reversible isothermal entropy change of 43.0 j·kg^(−1)·K^(−1) and adiabatic temperature change from barocaloric effects(ΔT_(BCE))of 15.6 K can be obtained within a wide temperature span of 30 K at 360 MPa,which is mainly attributed to the integration of the change in the transition temperature driven by pressure of−101 K·GPa^(−1) and suitable thermal hysteresis of 11.1 ***,the variation of reversibleΔ_(TBCE) against the applied hydrostatic pressure reaches up to 43 K·GPa^(−1),which is at the highest level among the other reported giant barocaloric *** importantly,after 60 thermal cycles,the composite does not break and the calorimetric curves coincide well,demonstrating good thermal cycle stability.
