We present a novel transient fault detection and classification approach in power transmission lines based on graph convolutional neural *** with the existing techniques,the proposed approach considers explicit spatial information in sampling sequences as prior knowledge and it has stronger feature extraction *** this basis,a framework for transient fault detection and classification is *** structure is generated to provide topology information to the *** approach takes the adjacency matrix of topology graph and the bus voltage signals during a sampling period after transient faults as inputs,and outputs the predicted classification results ***,the proposed approach is tested in various situations and its generalization ability is verified by experimental *** results show that the proposed approach can detect and classify transient faults more effectively than the existing techniques,and it is practical for online transmission line protection for its rapidness,high robustness and generalization ability.

目前,风电出力预测面临跨环境、跨传感器设备的多任务挑战,往往需要对不同的预测目标各自独立地展开针对性训练。鉴于此,首先提出了一种基于模型无关元学习策略(model-agnostic meta-learning,MAML)的短期预测方法,并基于该方法能够实现对新任务样本快速适应的能力设计了新型回归训练框架。然后结合卷积神经网络–长短期记忆网络、有注意力机制的Seq2Seq、有自注意力机制的Transformer、Synthesizer等时序深度网络模型,将该框架应用于风力发电预测场景。实验结果表明相比常规的预训练–微调的深度网络训练方法,所提出的方法在GEFCom2012数据集上对各算例实现了均方根误差和均方误差等指标的提高,同时各模型在短时风电出力为案例的预测任务上的泛化性能获得了一定提升。该训练框架可便捷地将主流深度学习模型和数据集转换为适应MAML策略的匹配模式。

The inhibitory receptor signal regulatory protein-α(Sirpα)is a myeloid-specific immune checkpoint that engages the“don’t eat me”signal CD47,which is expressed on tumor and normal tissue ***,the profile and regulatory mechanism of Sirpαexpression in tumor-associated macrophages(TAMs)are still not ***,we found that the expression of Sirpαin TAMs increased dynamically with colorectal cancer(CRC)***,CRC cell-derived lactate induced the nuclear translocation of the transcription factor Ap-2αfrom the cytoplasm in ***-2αfunctioned as a transcription factor for Elk-1 by binding to the conserved element GCCTGC located at−1396/−1391 in the mouse Elk-1 ***,the Elk-1 protein bound to two conserved sites,CTTCCTACA(located at−229/−221)and CTTCCTCTC(located at−190/−182),in the mouse Sirpαpromoter and promoted Sirpαexpression in ***,the macrophage-specific knockout of Ap-2αnotably promoted the phagocytic activity of TAMs and suppressed CRC progression,whereas these effects were prevented by the transgenic macrophage-specific expression of Elk-1,which regulated TAM phagocytosis and CRC development in a Sirpα-dependent ***,we showed that Elk-1 expression was positively correlated with Sirpαexpression in TAMs and was associated with poor survival in CRC *** together,our findings revealed a novel mechanism through which CRC evades innate immune surveillance and provided potential targets for macrophage-based immunotherapy for CRC patients.

目的探讨以丙酮酸脱氢酶激酶-1(pyruvate dehydrogenase kinase isozyme 1,PDK-1)为靶标的短发夹RNA(shRNA)与化疗药5-氟尿嘧啶(5-FU)联合应用对人结肠癌LS174T细胞的促凋亡作用。方法针对PDK-1序列设计3段干扰序列及1个阴性对照,分别构建pGenesil.1-PDK-1重组质粒表达载体及pGenesil.1-NC阴性对照载体,利用脂质体分别将质粒转染入LS174T,并用G418对细胞进行药物筛选,并通过Real-time PCR和Western blot检测有干扰序列的有效性。MTT实验检测5-FU对干扰组和对照组细胞生长、增殖的影响,流式细胞仪检测细胞凋亡率。结果 Real-time PCR和Western blot证实3个干扰组均抑制PDK-1表达,其中最大敲减效率超过70%。MTT实验结果显示,经5-FU同样处理48 h后,PDK-1干扰的LS174T细胞生长抑制率较对照组显著增高(P<0.05);流式细胞仪结果显示,干扰组细胞凋亡率较对照组增高(P<0.05)。结论 PDK-1 shRNA能特异性下调LS174T细胞中PDK-1的表达,PDK-1干扰联合5-FU可促进结肠癌细胞化疗诱导凋亡。

Intestinal cancers are developed from intestinal epithelial stem cells(ISCs)in intestinal crypts through a multi-step process involved in genetic mutations of oncogenes and tumor suppressor *** play a key role in maintaining the homeostasis of gut *** 2009,Sato et al established a three-dimensional culture system,which mimicked the niche microenvironment by employing the niche factors,and successfully grew crypt ISCs into organoids or Mini-guts in *** then,the intestinal organoid technology has been used to delineate cellular signaling in ISC ***,the cultured organoids consist of heterogeneous cell populations,and it was technically challenging to introduce genomic changes into three-dimensional ***,there was a technical necessity to develop a twodimensional ISC culture system for effective genomic *** this study,we established a conditionally immortalized mouse intestinal crypt(ciMIC)cell line by using a piggyBac transposon-based SV40 T antigen expression *** showed that the ciMICs maintained long-term proliferative activity under two-dimensional niche factor-containing culture condition,retained the biological characteristics of intestinal epithelial stem cells,and could form intestinal organoids in three-dimensional *** in vivo cell implantation tests indicated that the ciMICs were non-tumorigenic,the ciMICs overexpressing oncogenic b-catenin and/or KRAS exhibited high proliferative activity and developed intestinal adenoma-like pathological features in ***,these findings strongly suggested that the engineered ciMICs should be used as a valuable tool cell line to dissect the genetic and/or epigenetic underpinnings of intestinal tumorigenesis.

目的检测丙酮酸脱氢酶激酶-I(pyruvate dehydrogenase kinase-I,PDK1)在人结肠癌组织和细胞中的表达。探讨抑制PDK1对结肠癌细胞活性的影响。方法应用RT-PCR、荧光定量PCR和免疫荧光法、Western blot检测结肠癌组织、癌旁组织以及4种人结肠癌细胞PDK1基因的表达情况。在PDK1抑制剂(dichloroacetate,DCA)的诱导下,通过MTT法观察结肠癌细胞活性。结果PDK1基因在结肠癌组织中表达较癌旁组织中高;在4种人结肠癌细胞中明显表达。抑制PDK1能抑制结肠癌细胞增殖活性,抑制效应随药物浓度而递增,其中90 mmol/L DCA的活性细胞率分别为(23.90±2.79)%、(5.90±1.31)%、(32.82±4.50)%和(29.72±3.03)%。结论PDK1基因在结肠癌组织和细胞中高表达,在癌旁组织中低表达,抑制PDK1明显抑制结肠癌细胞增殖活性,PDK1可望成为结肠癌有效治疗靶点。

Pancreatic cancer is one of the leading causes of cancer-related mortality in both developed and developing *** incidence of pancreatic cancer in China accounts for about a quater of the global incidence,and the epidemiological characteristics and therapeutic strategies differ due to social,economic,cultural,environmental,and public health ***-domestic guidelines do not reflect the clinicopathologic characteristics and treat-ment patterns of Chinese ***,in 2018,the Chinese Society of Clinical Oncology(CSCO)organized a panel of senior experts from all sub-specialties within the field of pancreatic oncology to compile the Chinese guidelines for the diagnosis and treatment of pancreatic *** guidelines were made based on both the Western and Eastern clinical evidence and updated every one or two *** experts made consensus judg-ments and classified evidence-based recommendations into various grades according to the regional differences,the accessibility of diagnostic and treatment resources,and health economic indexes in *** we present the latest version of the guidelines,which covers the diagnosis,treatment,and follow-up of pancreatic *** guidelines might standardize the diagnosis and treatment of pancreatic cancer in China and will encourage oncologists to design and conduct more clinical trials about pancreatic cancer.

教育资源可达性的精准评估是衡量教育资源配置合理性的基础和前提,也是推进公共教育均等化的重要途径。针对前人在教育资源可达性研究中对设施综合服务能力研究不足和交通模式单一等问题,论文以东莞为例,结合多源数据、综合服务能力和多出行模式改进两步移动搜索法,测度东莞市小学空间可达性,并评价其公平性。研究发现:①东莞市小学可达性呈现西高东低的空间特征,高可达性的格网主要在中心城区4个街道及虎门镇、厚街镇等;低可达性的格网主要在水乡地区、大岭山和银瓶山等地形复杂地区。②从公平性视角看,东莞市域和91.18%的镇街小学空间分配处于绝对公平水平,茶山镇、东坑镇公平性最高,麻涌镇、松山湖高新区、虎门镇公平性最低。③学龄人口规模、人均GDP和教育经费支出是东莞市小学空间可达性的主要影响因素。研究可为实现东莞市教育资源的空间优化配置和构建更公平的教育发展新格局提供决策参考。
