Helicobacter pylori infection in relation to E-cadherin gene promoter polymorphism and hypermethylation in sporadic gastric carcinomas

AIM: To study Helicobacter pylori (H pylori) infection in relation to E-cadherin (E-cad) promoter polymorphism and hypermethylation in ***: Specimens were taken from representative cancerous lesions and adjacent non-cancerous epithelia of 67 resected GCs. Hpyloriwas detected by real-time PCR of the cagA gene from non-neoplastic epithelium.E-cad promoter polymorphism and hypermethylation were determined by restriction fragment length polymorphism analysis and methylation-specific PCR, respectively. Expression of E-cad protein was determined by ***: Hpyloriwas found in 57% of patients with GC.H pylori infection was more frequently found in tumors with the -160C/C genotype than those with the -160C/A and -160A/A genotypes (74% vs47%, P = 0.02). Hpylori infection was associated with E-cad methylation in nonneoplastic epithelium; however, no significant difference in H pylori was observed between methylated and unmethylated cancerous ***: Patients with the -160C/C genotype might require Hpyloriinfection to promote the inactivation of CDH1, suggesting that Hpylori infection might affect GC in an initial stage because polymorphism is germ *** of hypermethylation of CDH1 promoter in GC is complex, and Hpyloriinfection might affect it in an initial stage.

AIM： To study the role of CDH1/E-cadherin （E-cad） gene alteration profiles including mutation, loss of heterozygosity （LOH）, promoter polymorphism and hypermethylation in mechanisms of CDH1 inactivation in gastric carcinoma （GC）. METHODS： Specimens were collected surgically from 70 patients with GC. Allelotyping PCR and detection of LOH, denaturing high pressure liquid chromatography and DNA sequencing, restriction fragment length polymorphism analysis, methylation specific PCR, and immunohistochemical staining were used. RESULTS： Promoter polymorphism was not a major mechanism of E-cad inactivation. Only one truncating mutation was found in a diffuse type tumor （3%）. Both LOH and promoter hypermethylation were major mechanisms of E-cad inactivation, but interestingly, there was a negative association between the fraction of allelic loss （LOH） in tumors and hypermethylation of CDH1. Therefore LOH and hypermethylation were two different tumorigenic pathways involved in GC. CONCLUSION： Given the findings that somatic mutation was extremely low and the relationship between LOH and hypermethylation was inverse, any two combinations of these three factors cannot fulfill the classical two-hit hypothesis of CDH1 inactivation. Thus, other mechanisms operating at the transcriptional level or at the post-translational level might be required to induce E-cadherin inactivation.

Developing New Small Molecular Drugs for Prostate Cancer Therapy

Most of the early prostate cancer has no obvious symptoms, but its malignancy metastasis will cause largely deaths. The treatment options for patients with prostate cancer include traditional surgery, external beam therapy, hormone therapy, small molecular drug and cryosurgery. It was considered non-traditional treatments also can be used in alternative medicines for prostate cancer therapy. There are well-known molecular mechanisms and their pathogenesis, which provide potential targets for drug screening on the prostate cancer. Currently, natural plant extracts or human tissues active ingredients are widely used for the treatment of cancer. Then isolated effective substances in the extract, and further prepared large amounts of small molecule drugs by chemical synthesis. In this review, we summarized four small molecular drugs, abiraterone, docetaxel, isochaihulactone and butylidenephthalide, and their detailed anti-tumor mechanisms. These indicate that small molecular drug is a very efficient way and can be used for prostate cancer treatment.

目的基于皮肤类器官模型探索生物力学-化学信号促进皮肤及毛囊再生的机制,为解决皮肤创伤和脱发问题提供新的思路。方法本研究通过微流控类器官培养模型、细胞延时摄影、单细胞多组学技术、力学环境重编程、分子功能干扰等技术探究了生物力学-化学信号如何参与皮肤类器官的自组装过程,通过裸鼠移植实验评价了类器官诱导毛发再生的效果。结果通过延时摄影对类器官自组装过程中的细胞行为进行追踪,发现真皮细胞和表皮细胞经历了“聚集-极化-联合-平面化”4个阶段,形成功能性皮肤结构。真皮细胞的收缩力激活了表皮聚集体的Piezo1信号,进而形成极化的细胞形态发生单位,此结构通过局部微环境力学硬度增加激活了Yap1及下游的Wnt/Mmp信号,诱导相邻的CMU结构联合。在缺氧诱导因子Hif1a的驱动下真皮细胞和表皮细胞通过代谢适应完成相位分离,形成功能性平面化皮肤。将完成自组装的类器官移植裸鼠背部后,成功诱导移植部位的毛发再生。结论本研究深入分析了皮肤类器官的自组装过程,揭示了力学-化学信号引起的组织形态相变影响毛囊再生的机制。这一自组装途径为利用解离的皮肤前体细胞提供了一种重塑形态的有效途径,也为未来修复或改造器官提供了可能性。

Trends and driving forces of low-carbon energy technology innovation in China’s industrial sectors from 1998 to 2017: from a regional perspective

Low-carbon energy technology(LC)innovation contributes to both environmental protection and economic *** the panel data of 30 provinces/autonomous regions/municipalities in China from 1998 to 2017,this paper constructs a two-layer logarithmic mean Divisia index(LMDI)model to uncover the factors influencing the variation of the innovation of LC in China’s industrial sectors,including the alternative energy production technology(AEPT)and the energy conversation technology(ECT).The results show that China’s industrial LC patent applications rapidly increased after 2005 and AEPT patent applications outweighed ECT patent applications all the time with a gradually narrowing ***-carbon degree played the dominant role in promoting the increase in China’s industrial LC patent applications,followed by the economic scale,R&D(research and development)efficiency,and R&D *** structure contributed to the increases in LC patent applications in the central and the western regions,while led to the decreases in the eastern region,the north-eastern region,and Chinese ***-carbon degree and economic scale were two main contributors to the growths of both industrial AEPT patent applications and ECT patent applications in Chinese mainland and the four *** policy recommendations are made to further promote industrial innovation in China.

PUF-Based Key Distribution in Wireless Sensor Networks

Physical Unclonable Functions(PUFs)can be seen as kind of hardware one-way functions,who are easily fabricated but difficult to clone,duplicate or ***,PUFs with unclonable and unpredictable properties are welcome to be applied in designing lightweight cryptography *** this paper,a Basic Key Distribution Scheme(Basic-KDS)based on PUFs is firstly ***,by employing different deployment modes,a Random Deployment Key Distribution Scheme(RD-KDS)and a Grouping Deployment Key Distribution Scheme(GD-KDS)are further proposed based on the Basic-KDS for large scale wireless sensor *** our proposals,a sensor is not pre-distributed with any keys but will generate one by the embedded PUF when receiving a challenge from the gateway,which provides perfect resilience against sensor capture ***,the unclonable and unpredictable properties of PUF guarantee the key uniqueness and two-way *** and experiment results show that our proposals have better performances in improving the resilience,secure-connectivity,and efficiency as compared to other schemes.

Chemometrics of differentially expressed proteins from colorectal cancer patients

AIM:To evaluate the usefulness of differentially expressed proteins from colorectal cancer (CRC) tissues for differentiating cancer and normal ***:A Proteomic approach was used to identify the differentially expressed proteins between CRC and normal *** proteins were extracted using Tris buffer and thiourea lysis buffer (TLB) for extraction of aqueous soluble and membrane-associated proteins,***,namely principal component analysis (PCA) and linear discriminant analysis (LDA),were used to assess the usefulness of these proteins for identifying the cancerous state of ***:Differentially expressed proteins identified were 37 aqueous soluble proteins in Tris extracts and 24 membrane-associated proteins in TLB *** on the protein spots intensity on 2D-gel images,PCA by applying an eigenvalue > 1 was successfully used to reduce the number of principal components (PCs) into 12 and seven PCs for Tris and TLB extracts,respectively,and subsequently six PCs,respectively from both the extracts were used for *** LDA classification for Tris extract showed 82.7% of original samples were correctly classified,whereas 82.7% were correctly classified for the cross-validated *** LDA for TLB extract showed that 78.8% of original samples and 71.2% of the cross-validated samples were correctly ***:The classification of CRC tissues by PCA and LDA provided a promising distinction between normal and cancer *** methods can possibly be used for identification of potential biomarkers among the differentially expressed proteins identified.

Analysis of differentially expressed proteins in cancerous and normal colonic tissues

AIM： To identify and analyze the differentially expressed proteins in normal and cancerous tissues of four patients suffering from colon cancer. METHODS： Colon tissues （normal and cancerous） were homogenized and the proteins were extracted using three protein extraction buffers. The extraction buffers were used in an orderly sequence of increasing extraction strength for proteins with hydrophobic properties. The protein extracts were separated using the SDS-PAGE method and the images were captured and analyzed using Quantity One software. The target protein bands were subjected to in-gel digestion with trypsin and finally analyzed using an ESI-ion trap mass spectrometer. RESULTS： A total of 50 differentially expressed proteins in colonic cancerous and normal tissues were identified. CONCLUSION： Many of the identified proteins have been reported to be involved in the progression of similar or other types of cancers. However, some of the identified proteins have not been reported before. In addition, a number of hypothetical proteins were also identified.
